  • Robust estimation of bacterial cell count from optical density
  • 2020
  • Ingår i: Communications Biology. - : Springer Science and Business Media LLC. - 2399-3642. ; 3:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Installation of microseismic monitoring networks in geothermal fields
  • 2023
  • Ingår i: JOURNAL OF THE FACULTY OF ENGINEERING AND ARCHITECTURE OF GAZI UNIVERSITY. - : GAZI UNIV, FAC ENGINEERING ARCHITECTURE. - 1300-1884 .- 1304-4915. ; 38:3, s. 1307-1319

    • Purpose: How the optimum microseismic monitoring network should be installed for induced seismicity studies in selected geothermal area. So, noise analysis results are given in borehole and surface locations using different type of seismometers in selected locations. Theory and Methods: In induced seismicity analysis, high frequency, low energy and small amplitude microseismic events are obtained within continuous passive seismic records. Noise analysis important step of the network installation before the data analysis and determining microseismic events. So, the background noise at the locations are compared from borehole and surface stations in same points. Results: According to compared results, the noise level in the well is very low compared to the surface and this is at suitable level to obtain microseismic events clearly. So, borehole seismic sensors can be preferred in microseismic monitoring networks in induced seismicity studies. Conclusion: Reservoir management is crucial subject for geothermal studies. So, microseismic monitoring method provides continues results about the geothermal reservoir and these results are used in the reservoir management. In fact, microseismic monitoring method is generally standard in abroad but there are no detailed studies in our country. This study is the first study to monitor microseismic activities using borehole seismometers. The study includes network installations and optimum array design also. Noise level must be low level to obtain microseismic events clearly. It can be provided using borehole seismometers in indused seismicity studies.
