  • The association of coffee intake with liver cancer risk is mediated by biomarkers of inflammation and hepatocellular injury : data from the European Prospective Investigation into Cancer and Nutrition

    • Background: Higher coffee intake has been purportedly related to a lower risk of liver cancer. However, it remains unclear whether this association may be accounted for by specific biological mechanisms. Objective: We aimed to evaluate the potential mediating roles of inflammatory, metabolic, liver injury, and iron metabolism biomarkers on the association between coffee intake and the primary form of liver cancer-hepatocellular carcinoma (HCC). Design: We conducted a prospective nested case-control study within the European Prospective Investigation into Cancer and Nutrition among 125 incident HCC cases matched to 250 controls using an incidence-density sampling procedure. The association of coffee intake with HCC risk was evaluated by using multivariable-adjusted conditional logistic regression that accounted for smoking, alcohol consumption, hepatitis infection, and other established liver cancer risk factors. The mediating effects of 21 biomarkers were evaluated on the basis of percentage changes and associated 95% CIs in the estimated regression coefficients of models with and without adjustment for biomarkers individually and in combination. Results: The multivariable-adjusted RR of having >= 4 cups (600mL) coffee/d compared with <2 cups (300 mL)/d was 0.25 (95% CI: 0.11, 0.62; P-trend = 0.006). A statistically significant attenuation of the association between coffee intake and HCC risk and thereby suspected mediation was confirmed for the inflammatory biomarker IL-6 and for the biomarkers of hepatocellular injury glutamate dehydrogenase, alanine aminotransferase, aspartate aminotransferase (AST), gamma-glutamyltransferase (GGT), and total bilirubin, which-in combination-attenuated the regression coefficients by 72% (95% CI: 7%, 239%). Of the investigated biomarkers, IL-6, AST, and GGT produced the highest change in the regression coefficients: 40%, 56%, and 60%, respectively. Conclusion: These data suggest that the inverse association of coffee intake with HCC risk was partly accounted for by biomarkers of inflammation and hepatocellular injury.

    • The linear thermal expansion of compacted Ar-doped fullerite C60(ArxC60) is investigated at 2–12 K using a dilatometric method. The thermal expansion of ArxC60 is also studied after partial desaturation of argon from fullerite. It is revealed that argon doping resulted in a considerable change of the temperature dependence of the thermal expansion of fullerite. An explanation of the observed effects is proposed.

    • Inequalities of Mandelstam-Tamm (MT) and Margolus-Levitin (ML) type provide lower bounds on the time that it takes for a quantum system to evolve from one state into another. Knowledge of such bounds, called quantum speed limits, is of utmost importance in virtually all areas of physics, where determination of the minimum time required for a quantum process is of interest. Most MT and ML inequalities found in the literature have been derived from growth estimates for the Bures length, which is a statistical distance measure. In this paper we derive such inequalities by differential geometric methods, and we compare the quantum speed limits obtained with those involving the Bures length. We also characterize the Hamiltonians which optimize the evolution time for generic finite-level quantum systems.

  • Terahertz absorption of illuminated photosynthetic reaction center solution: a signature of photoactivation?

    • Photosynthetic reaction centers develop a stable charge separated state upon illumination. To investigate the molecular vibrations associated with the illuminated state of a reaction center we recorded terahertz absorption spectra of the photosynthetic reaction center from Rhodobacter sphaeroides in the dark and upon illumination and observed a small, but significant THz absorption increase in the 20 to 130 cm(-1) spectral region. Reaction centers show very similar terahertz absorption increase when solubilized in detergents and in a lipidic sponge phase indicating that the nature of the bulk solvent has limited influence on the vibrational spectrum. The absorption change of the isolated LM subunit is very similar to that of the intact reaction center. Through temperature control experiments we show that 89% of the absorption change is likely attributed to the non-thermal activation of the protein molecules. These results indicate that picosecond molecular vibrations change primarily in the cofactors and/or in the evolutionary conserved core of the reaction center upon illumination, whereas the nuclear motions of the H-subunit and the bulk solvent have limited impact on the terahertz spectral changes.

  • Considerations for three-dimensional image reconstruction from experimental data in coherent diffractive imaging

    • Diffraction before destruction using X-ray free-electron lasers (XFELs) has the potential to determine radiation-damage-free structures without the need for crystallization. This article presents the three-dimensional reconstruction of the Melbournevirus from single-particle X-ray diffraction patterns collected at the LINAC Coherent Light Source (LCLS) as well as reconstructions from simulated data exploring the consequences of different kinds of experimental sources of noise. The reconstruction from experimental data suffers from a strong artifact in the center of the particle. This could be reproduced with simulated data by adding experimental background to the diffraction patterns. In those simulations, the relative density of the artifact increases linearly with background strength. This suggests that the artifact originates from the Fourier transform of the relatively flat background, concentrating all power in a central feature of limited extent. We support these findings by significantly reducing the artifact through background removal before the phase-retrieval step. Large amounts of blurring in the diffraction patterns were also found to introduce diffuse artifacts, which could easily be mistaken as biologically relevant features. Other sources of noise such as sample heterogeneity and variation of pulse energy did not significantly degrade the quality of the reconstructions. Larger data volumes, made possible by the recent inauguration of high repetition-rate XFELs, allow for increased signal-to-background ratio and provide a way to minimize these artifacts. The anticipated development of three-dimensional Fourier-volume-assembly algorithms which are background aware is an alternative and complementary solution, which maximizes the use of data.

    • In this paper, we propose a method for adaptive selection of the kernel size used in estimation of the wall shear stress (WSS) based on flow velocity profiles obtained by the blood flow imaging method based on enhancement of RF echo signals from blood cells using a singular value decomposition based clutter filter. Two simulation phantom experiments with the Field II program were performed to validate the proposed method. In the simulation experiments, the average bias error (BE) between the WSSs estimated by the conventional method and theoretical one was 16.5%. The proposed method could reduce the average BE to 12.3%. Also, the proposed method was applied to RF echo signals from a common carotid artery (CCA) of a healthy male subject. The WSSs in the CCA were estimated during one cardiac cycle and in a range similar to that in previous reports.

  • Age-related alteration of functional connectivity in the default mode network and insular cortex detected by quantitative data-driven analysis of resting-state fMRI

    • Background: Age-related changes in the brain-parenchyma may alter the brains functional connectivity. Studies have consistently reported significant age-effects in the default mode network (DMN). Less is known about sex differences and the interaction effect with older age. Both independent component analysis and seed-based analysis have been widely used in the literature.Method: We investigated 29 young (age=25.0±3.4) and 31 older (n=31, age=68.4±2.7) healthy adults (♀50%) with a resting-state fMRI protocol at 3T. We evaluated voxel-wise differences in brain functional connectivity in relation to age and sex of the participants by taking advantage of a recently developed quantitative data-driven analysis (QDA) method, where the connection counters (CCI) and strength (CSI) indies for each voxel in the brain are measured.Results: Compared with the CSI result, more extensive brain regions showed significantly reduction in CCI in older age. Besides the brain regions involved in the DMN, we found that older age induced also functional connectivity decline in thalamus and insula cortex. On average, females had lower CCI and CSI in the right middle frontal gyrus. The anterior cingulate, right middle frontal gyrus, and left lentiform nucleus showed age and sex interaction effect. The finding of reduced functional connectivity in older adults was also confirmed by the result from linear regression analysis.Conclusion: Age-related effect as detected by the QDA method is largely consistently with previously reported findings. Furthermore, our study added interesting new findings in age-related differences. This indicates that QDA provides a sensitive model-free approach for voxel-wise analysis of functional connectivity.

    • Here, we introduce a novel scFv antibody, G2-D11, specific for two adjacent Tn-antigens (GalNAc- Ser/Thr) binding equally to three dimeric forms of the epitope, Ser-Thr, Thr-Thr and Thr-Ser. Compared to other anti-Tn reagents, the binding of G2-D11 is minimally influenced by the peptide structure, which indicates a high degree of carbohydrate epitope dominance and a low influence from the protein backbone. With a high affinity (KDapp = 1.3 × 10-8 M) and no cross-reactivity to either sialyl-Tn epitope or blood group A antigens, scFv G2-D11 is an excellent candidate for a well-defined anti-Tn-antigen reagent. Detailed immunohistochemical evaluation of tissue sections from a cohort of 80 patients with gastric carcinoma showed in all cases positive tumor cells. The observed staining was localized to the cytoplasm and in some cases to the membrane, whereas the surrounding tissue was completely negative demonstrating the usefulness of the novel Tn-antigen binding antibody.

    • This work presents a novel modeling methodology which allows one to predict slag infiltration in a 3D slab model by coupling the slag behavior with the metal flow, heat transfer, oscillation and solidification within the mold. The model is able to describe the dynamic behavior of liquid/solid slag in the shell mold gap and the bed, as well as its effects on heat extraction and shell formation. This paper also illustrates both the application of the model to industrial practice and the challenges faced during its implementation. Furthermore, the model allows one to predict the conditions leading to irregular shell growth, which is likely to generate defects such as cracking and bulging. It also provides, at the same time, a more fundamental understanding of the mechanisms responsible for defect formation.

    • Background: Optibirth is a funded FP7 research programme using a cluster randomised trial in Ireland, Germany and Italy, with 15 clusters of 94 women per cluster. The aim of OptiBIRTH is to evaluate the effectiveness of a complex intervention aimed at increased VBAC rates through enhanced women-centred care1 .The intervention consisted of motivationally enhanced, evidence-based, educational information for women and clinicians for use in face to face and online modes. Purpose/Objective: This paper reports the design process associated with creating three interrelated mobile apps, as part of a complex intervention, to enable women with a previous caesarean section to decide autonomously between a repeat caesarean section and a vaginal birth. The emphasis has been on using technology creatively and effectively maximizing optimality bearing in mind the needs of the new “Z” generation. Method: Focus groups were undertaken across six European countries to illicit women’s need for information about caesarean section and VBAC. Motivational theory underpinned the development of subsequent e-learning materials to enhance woman-led-decision-making. Ethical approval was obtained from Trinity College University, Dublin. Following analysis of focus group data, consultation with experts and exploration of technological solutions, three electronic applications were designed. Following three iterations, the Apps were reviewed for content, motivational design and functionality by Irish, German and Italian midwifery experts and translated into German and Italian. Key Findings: The following interrelated Apps were designed to provide women and health professionals with rapid access to women’s perceptions of their previous birth experience and their current decision-making-in-progress: “My birth story” uses a wordle to communicate past birth experience “My birth thoughts” enables women to communicate their current decision making “My Birth plan” generates a personalised birth plan Discussion: Discussion will focus on challenges facing midwives including, embedding the Apps into a motivationally-designed portal, staff support and training, timeline, cultural differences and translation issues

    • The Journal of Intelligence Studies in Business (JISIB) has performed a survey, or done a spot-check, to learn more about its users at the end of three years of publications. Users were found via the journal’s site on LinkedIn and a web-survey was sent from there as an announcement. 18 respondents answered completely. This was only 3,2% of the total member group, but we still think we can draw a number of conclusion from it, also as compared to feedback gathered during the years. Users are looking for more case study material in the articles. There is an even balance between those who think there is too much technical material and too little. The discussion about what languages to publish articles in is likely to continue. It is not given that this should be exclusively English in the future. At the same time publishing non-English articles present a number of challenges.

    • This study focused on the attainment of Theory of Mind (ToM) in children (aged 6 to 13) with complex communication needs who used augmentative and alternative communication (AAC). The AAC group (n = 14) was matched to a younger group, without disabilities, vis-à-vis nonverbal mental age. A second comparison group consisting of children with mild intellectual disabilities and matched nonverbal mental age and chronological age was also included. A test battery that included tests of cognitive and language development and tests of first- and second-order ToM understanding and understanding of more advanced ToM was used. Nonverbal intelligence proved to be a good indicator of ToM ability. Contrary to previous research, the AAC group did not differ significantly from the comparison groups on the test results. The results are discussed in terms of the necessary cognitive capacity underpinning ToM development.

  • Vasomotor symptoms in postmenopausal women : the role of acupuncture and calcitonin gene-related peptide

    • A majority of postmenopausal women experience vasomotor symptoms, which have a significant impact on the women's quality of life. Estrogens are the treatment of choice but can for different reasons not be used by all women. There is therefore a great need for viable alternative treatments. The patophysiology behind hot flushes and sweatings is so far not fully understood. Several studies have concluded that low levels of estrogens after menopause will lead to instability in the thermoregulatory center in the hypothalamus probably due to low [ß-endorphin levels. Since acupuncture is known to increase central [ß-endorphin activity, we wanted to evaluate if this treatment could ameliorate vasomotor symptoms which, to our knowledge, has not been scientifically evaluated previously. We also aimed to reveal if the vasoactive neuropeptides Calcitonin Gene-related peptide (CGRP), neuropeptide Y (NPY), neurokinin A, and substance P were involved in the mechanisms behind these symptoms.Results: Electro-acupuncture decreased flushes by 50 %, and superficial needle insertion by 30 %. The difference in reduction between the groups was not significant. Along with the decrease of flushes we found a significant reduction of the 24h urinary excretion of the neuropeptide CGRP, which is one of the most potent vasodilators known. When we later compared electro-acupuncture (EA), superficial needle insertion (SNI) and estrogen treatment, we found a significant reduction of 24h flushes by almost 60 % after EA and SNI. However, about 20% were non-responders in both groups. The responders in the EA group reduced their flushes and sweatings by 82 % and in the SNI group the reduction was 68%. We found no significant difference in effect between the acupuncture groups. Estrogen was the most effective treatment with a 91% reduction of flushes (range 58-100 %). We found a higher excretion of CGRP/24h urine in postmenopausal women with vasomotor symptoms, than in women without symptoms. Furthermore we found a 73 % elevation of CGRP in plasma, along with a 34% increase ofNPY concentration, during flushes in postmenopausal women.Conclusion: The results indicate that acupuncture is as a viable alternative or complement to traditional pharmacological treatments in postmenopausal women with vasomotor symptoms. The vasoactive neuropeptide CGRP is most likely involved in the mechanisms of vasomotor symptoms, probably as an executor or mediator of the skin vasodilatation and sweating that occur during the hot flush.

The course gives an introduction to the use of OpenSource softwarefor CFD applications. It has a strong focus on how to efficiently use the Linux operatingsystem and different softwares that are useful for CFD. A major individual project work in OpenFOAMforms a large part of the course. The project is defined according to the student’s specialinterests. The result of the project is a detailed tutorial for a specific application or library ofOpenFOAM, which is presented to, and peer-reviewed by the students. The tutorials thus forma part of the course (Docendo discimus - Latin: ’by teaching, we learn’). The tutorials are madeavailable through the proceedings, as a contribution to the OpenFOAM community, and to contributeto global learning. The tutorials are typically made available in the form of a report, aset of slides, possible source code, possible cases, and additional material, to be downloaded athttp://www.tfd.chalmers.se/~hani/kurser/OS_CFD, under the corresponding year.The main requirements of a tutorial is that it should consider the following four aspects:• How to use it.• The theory of it.• How it is implemented.• How to modify it.This should be reflected by the specified learning outcomes of each tutorial, and should be evaluatedby a set of study questions.It should be recognized that the tutorials are student project work, done as part of a course.For many of the students it is the first time they are introduced to OpenFOAM, C++ and objectoriented programming, and some of the students have limited previous experience with Linux. Anyreader should be aware that the tutorials might not be free of errors. Still, the tutorials might beuseful for someone who would like learn something similar as the students learnt while doing theproject work. The material has gone through a review process. The role of the reviewers is to gothrough the tutorial and make sure that it works, that it is possible to follow, and to some extentcorrect the writing.     • The current study investigated the associations between work-related perceived stress and surface electromyographic (sEMG) parameters (muscle activity and muscle rest) during standardized simulated computer work (typing, editing, precision, and Stroop tasks). It was part of the European case-control study, NEW (Neuromuscular assessment in the Elderly Worker). The present cross-sectional study was based on a questionnaire survey and sEMG measurements among Danish and Swedish female computer users aged 45 or older (n=49). The results show associations between work-related perceived stress and trapezius muscle activity and rest during standardized simulated computer work, and provide partial empirical support for the hypothesized pathway of stress induced muscle activity in the association between an adverse psychosocial work environment and musculoskeletal symptoms in the neck and shoulder.

    • The article is about the attitudes of corner boys and college boys’ to social mobility, showing that the 1930s was a watershed era. The first generation of Italians were often farmers and fishermen who became industrial workers in the United States. The second generation of Italians had adapted to American cultural conditions to varying degrees, with commitments in both the local community and the majority society. Corner boys planned for a future in the local community. College boys strove to pursue a career outside the social and geographical boundaries of the community. Corner boys learned the rules of society on the street. College boys were handpicked at school and in the settlement house to become middle-class Americans. Corner boys regarded college boys as hypocrites who put their short-term interests before those of the local community. The schisms between corner boys and college boys were an indication that the family values of the local Italian community were changing. But it was not until some time after the Second World War that the third and fourth generations of Italians had acquired moral values similar to those of the average American about abortion, mixed marriages, homosexuality, sexual abstinence before marriage, and divorce.

This is achievable via monitoring patient's vital signs continuously and is often manually done with paper and pen. However, because of the constraints in healthcare resources and the high hospital costs, the patient might not be hospitalized for the whole period of the treatments, which has lead to a demand for in-home or portable EWS systems. Such a personalized EWS system needs to monitor the patient at anytime and anywhere even when the patient is carrying out daily activities. In this paper, we propose a self-aware EWS system which is the reinforced version of the existing EWS systems by using the Internet of Things technologies and the self-awareness concept. Our self-aware approach provides (i) system adaptivity with respect to various situations and (ii) system personalization by paying attention to critical parameters. We evaluate the proposed EWS system using a full system demonstration.

    • A linear reference is a reference guaranteed to be unaliased. Thisis a powerful property that simplifies reasoning about programs,but is also a property that is too strong for certainapplications. For example, lock-free algorithms, which implementprotocols to ensure safe concurrent access to data structures, aregenerally not typable with linear references as they involvesharing of mutable state.This paper presents a type system with a relaxed notion oflinearity that allows an unbounded number of aliases to an objectas long as at most one alias at a time owns the right to accessthe contents of the object. This ownership can be transferredbetween aliases, but can never be duplicated. The resultinglanguage is flexible enough to express several lock-freealgorithms and at the same time powerful enough to guarantee theabsence of data-races when accessing owned data. The language isformalised and proven sound, and is also available as a prototype implementation.

    • We have developed a combinatory antibody-antigen microarray for direct screening of multiple single-chain fragment variable (scFv) clones with no need for pre-purification or enrichment before screening. The straightforward workflow allows for early selection of binders to predefined peptide and glycopeptide targets. A capture antibody is contact printed on microarray slides, side by side with the antigens of interest. A large number of scFv clones, in supernatants, are printed on top of the capture antibody and the antigen in a "spot-on-spot" print. The printed scFv clones, which bind to the capture antibody, are detected using biotinylated antigen, while the binding of scFv clones to the printed antigen is detected through a mouse anti-tag antibody. Two different analyses are thus performed on the same slide, generating two kinds of information: one on the ability of an individual scFv clone to bind to the soluble form of the antigen, which may favour selection for higher affinity rather than avidity, while the other allows the identification of large numbers of clones, simultaneously, due to the binding of scFv clones to densely presented antigens, thus providing an overall increased hit rate. The functionality of the new screening approach was illustrated through the generation of antibodies against peptides from the chaperone complex Ku70/Ku80 and the GalNAcαserine/threonine epitope on the IgA1 alpha chain hinge region. In total, 659 scFv clones were screened with a hit rate of approximately 20%. This approach allowed the identification of functional antibodies in both cases, illustrating the usefulness and capacity of this combinatory microarray screening technique for efficient analysis and validation of antibodies at an early stage of antibody generation.

    • Trade shows have consistently been neglected in marketing research, especially the gathering of market information, what is studied under Market Intelligence and directly related to a company’s Knowledge and Innovation. A major reason is that the marketing discipline traditionally has had a strong focus on customers instead of competitors and influencers. In this article we show how the field of Competitive Intelligence is a useful part of the company’s Integrated Marketing Communications. Based on a qualitative method encompassing a case study and exploratory research we followed, coached and traveled with two different groups of companies to major International Trade shows. Based on the information gathered a division into three areas of Intelligence was useful. These were intelligence about products, the booths and the behavior in the booths. For each group a number of Key Intelligence Topics and a set of specific research methods were identified which can make the Market Intelligence process more efficient. We explain why Exhibits or trade shows have much in common with Event Marketing and could be considered a part of the latter.

    • BACKGROUND: Few large-scale studies have examined the association between sociodemographic factors and the probability of reaching the recommended levels of HbA1c and blood lipids in patients with type 2 diabetes. The aim was to investigate whether sociodemographic characteristics of type 2 diabetes patients affected the odds that they would reach recommended levels of blood lipids and HbA1c. METHODS: This study included 2912 men and 2136 women, aged 35-74, with diagnosed type 2 diabetes from 25 primary health care centers in Stockholm, Sweden. National population registers were linked to clinical data from electronic records and logistic regression was used to estimate odds ratios. RESULTS: Less than half of the diabetic men and women reached the recommended levels of HbA1c. Even fewer reached the recommended levels for total cholesterol and LDL cholesterol. The gender differences favored women, for HbA1c, and men, for blood lipids. Individuals with the lowest income levels were less likely to reach the recommended level of HbA1c. Country of birth showed that immigrants from Middle Eastern countries and Other countries had lower odds of reaching the recommended levels of HbA1c. CONCLUSION: This study confirmed that risk factor control among patients with type 2 diabetes treated in primary health care is inadequate and that sociodemographic factors were associated with metabolic control. Future studies could include new strategies for the control of modifiable risk factors in patients with type 2 diabetes. Copyright © 2010 John Wiley & Sons, Ltd.
