  • Paracetamol analogues conjugated by FAAH induce TRPV1-mediated antinociception without causing acute liver toxicity

    • Paracetamol, one of the most widely used pain-relieving drugs, is deacetylated to 4-aminophenol (4-AP) that undergoes fatty acid amide hydrolase (FAAH)-dependent biotransformation into N-arachidonoylphenolamine (AM404), which mediates TRPV1-dependent antinociception in the brain of rodents. However, paracetamol is also converted to the liver-toxic metabolite N-acetyl-p-benzoquinone imine already at therapeutic doses, urging for safer paracetamol analogues. Primary amine analogues with chemical structures similar to paracetamol were evaluated for their propensity to undergo FAAH-dependent N-arachidonoyl conjugation into TRPV1 activators both in vitro and in vivo in rodents. The antinociceptive and antipyretic activity of paracetamol and primary amine analogues was examined with regard to FAAH and TRPV1 as well as if these analogues produced acute liver toxicity. 5-Amino-2-methoxyphenol (2) and 5-aminoindazole (3) displayed efficient target protein interactions with a dose-dependent antinociceptive effect in the mice formalin test, which in the second phase was dependent on FAAH and TRPV1. No hepatotoxicity of the FAAH substrates transformed into TRPV1 activators was observed. While paracetamol attenuates pyrexia via inhibition of brain cyclooxygenase, its antinociceptive FAAH substrate 4-AP was not antipyretic, suggesting separate mechanisms for the antipyretic and antinociceptive effect of paracetamol. Furthermore, compound 3 reduced fever without a brain cyclooxygenase inhibitory action. The data support our view that analgesics and antipyretics without liver toxicity can be derived from paracetamol. Thus, research into the molecular actions of paracetamol could pave the way for the discovery of analgesics and antipyretics with a better benefit-to-risk ratio.

  • A convex optimization approach to complexity constrained analytic interpolation with applications to ARMA estimation and robust control

    • Analytical interpolation theory has several applications in systems and control. In particular, solutions of low degree, or more generally of low complexity, are of special interest since they allow for synthesis of simpler systems. The study of degree constrained analytic interpolation was initialized in the early 80's and during the past decade it has had significant progress. This thesis contributes in three different aspects to complexity constrained analytic interpolation: theory, numerical algorithms, and design paradigms. The contributions are closely related; shortcomings of previous design paradigms motivate development of the theory, which in turn calls for new robust and efficient numerical algorithms. Mainly two theoretical developments are studied in the thesis. Firstly, the spectral Kullback-Leibler approximation formulation is merged with simultaneous cepstral and covariance interpolation. For this formulation, both uniqueness of the solution, as well as smoothness with respect to data, is proven. Secondly, the theory is generalized to matrix-valued interpolation, but then only allowing for covariance-type interpolation conditions. Again, uniqueness and smoothness with respect to data is proven. Three algorithms are presented. Firstly, a refinement of a previous algorithm allowing for multiple as well as matrix-valued interpolation in an optimization framework is presented. Secondly, an algorithm capable of solving the boundary case, that is, with spectral zeros on the unit circle, is given. This also yields an inherent numerical robustness. Thirdly, a new algorithm treating the problem with both cepstral and covariance conditions is presented. Two design paradigms have sprung out of the complexity constrained analytical interpolation theory. Firstly, in robust control it enables low degree Hinf controller design. This is illustrated by a low degree controller design for a benchmark problem in MIMO sensitivity shaping. Also, a user support for the tuning of controllers within the design paradigm for the SISO case is presented. Secondly, in ARMA estimation it provides unique model estimates, which depend smoothly on the data as well as enables frequency weighting. For AR estimation, a covariance extension approach to frequency weighting is discussed, and an example is given as an illustration. For ARMA estimation, simultaneous cepstral and covariance matching is generalized to include prefiltering. An example indicates that this might yield asymptotically efficient estimates.

  • To boldly go: individual differences in boldness influence migratory tendency.

    • Ecology Letters (2011) ABSTRACT: Partial migration, whereby only a fraction of the population migrates, is thought to be the most common type of migration in the animal kingdom, and can have important ecological and evolutionary consequences. Despite this, the factors that influence which individuals migrate and which remain resident are poorly understood. Recent work has shown that consistent individual differences in personality traits in animals can be ecologically important, but field studies integrating personality traits with migratory behaviour are extremely rare. In this study, we investigate the influence of individual boldness, an important personality trait, upon the migratory propensity of roach, a freshwater fish, over two consecutive migration seasons. We assay and individually tag 460 roach and show that boldness influences migratory propensity, with bold individuals being more likely to migrate than shy fish. Our data suggest that an extremely widespread personality trait in animals can have significant ecological consequences via influencing individual-level migratory behaviour.

  • Traffic-generated emissions of ultrafine particles from pavement-tire interface

    • In a road simulator study, a significant source of sub-micrometer fine particles produced by the road-tire interface was observed. Since the particle size distribution and source strength is dependent on the type of tire used, it is likely that these particles largely originate from the tires, and not the road pavement. The particles consisted most likely of mineral oils from the softening filler and fragments of the carbon-reinforcing filler material (soot agglomerates). This identification was based on transmission electron microscopy studies of collected ultrafine wear particles and on-line thermal treatment using a thermodesorber. The mean particle number diameters were between 15-50 nm, similar to those found in light duty vehicle (LDV) tail-pipe exhaust. A simple box model approach was used to estimate emission factors in the size interval 15-700 nm. The emission factors increased with increasing vehicle speed, and varied between 3.7 x 10(11) and 3.2 x 10(12) particles vehicle(-1) km(-1) at speeds of 50 and 70 km h(-1). This corresponds to between 0.1-1% of tail-pipe emissions in real-world emission studies at similar speeds from a fleet of LDV with 95% gasoline and 5% diesel-fueled cars. The emission factors for particles originating from the road-tire interface were, however, similar in magnitude to particle number emission factors from liquefied petroleum gas-powered vehicles derived in test bench studies in Australia 2005.     • Systemic inflammation activates central autonomic circuits, such as neurons in the pontine parabrachial nucleus. This activation may be the result of afferent signaling through the vagus nerve, but it may also depend on central prostaglandin-mediated mechanisms. Recently, we have shown that neurons in the parts of the parabrachial nucleus that are activated by immune challenge express prostaglandin receptors of the EP3 and EP4 subtypes, but it remains to be determined if the prostaglandin receptor-expressing neurons are identical to those that respond to immune stimuli. In the present study, bacterial wall lipopolysaccharide was injected intravenously in adult male rats and the expression of c-fos mRNA and of EP3 and EP4 receptor mRNA was examined with complementary RNA probes labeled with digoxigenin and radioisotopes, respectively. Large numbers of neurons in the external lateral parabrachial subnucleus, a major target of vagal-solitary tract efferents, expressed c-fos mRNA. Quantitative analysis showed that about 60% (range 40%–79%) of these neurons also expressed EP3 receptor mRNA. Conversely, slightly more than 50% (range 48%–63%) of the EP3 receptor-expressing neurons in the same subnucleus coexpressed c-fos mRNA. In contrast, few EP4 receptor-expressing neurons were c-fos positive, with the exception of a small population located in the superior lateral and dorsal lateral subnuclei. These findings show that immune challenge activates central autonomic neurons that could be the target of centrally produced prostaglandin E2, suggesting that synaptic signaling and paracrine mechanisms may interact on these neurons.     • By using in situ hybridization, the distribution of mRNA for the PGE2 receptors EP3 and EP4 was examined in the rat parabrachial nucleus (PB), a major brain stem relay for autonomic and nociceptive processing. EP3 receptor mRNA was present in most subnuclei, with the densest labeling in the external lateral, dorsal lateral, superior lateral, central lateral and Kölliker–Fuse nuclei. EP4 receptor mRNA expressing cells had a more restricted distribution, largely being confined to the superior lateral and adjacent parts of the dorsal and central lateral nuclei in a pattern complementary to that for EP3 receptor mRNA. These findings suggest that EP3 and EP4 receptors in PB have distinct functional roles that include nociceptive processing, blood pressure regulation and feeding behavior.

    • This study examines the distribution of prostaglandin E2 receptors of subtype EP3 and EP4 among brain stem parabrachial neurons that were characterized with respect to their neuropeptide expression. By using a dual-labeling in situ hybridization method, we show that preprodynorphin mRNA expressing neurons in the dorsal and central lateral subnuclei express EP3 receptor mRNA. Such receptors are also expressed in preproenkephalin, calcitonin gene related peptide and preprotachykinin mRNA positive neurons in the external lateral subnucleus, whereas preprodynorphin mRNA expressing neurons in this subnucleus are EP receptor negative. In addition, EP3 receptor expression is seen among some enkephalinergic neurons in the Kölliker-Fuse nucleus. Neurons in the central part of the cholecystokininergic population in the regions of the superior lateral subnucleus express EP4 receptor mRNA, whereas those located more peripherally express EP3 receptors. Taken together with previous findings showing that discrete peptidergic cell groups mediate nociceptive and/or visceral afferent information to distinct brain stem and forebrain regions, the present results suggest that the processing of this information in the parabrachial nucleus is influenced by prostaglandin E2. Recent work has shown that prostaglandin E2 is released into the brain following peripheral immune challenge; hence, the parabrachial nucleus may be a region where humoral signaling of peripheral inflammatory events may interact with neuronal signaling elicited by the same peripheral processes.

    • Although central nervous symptoms such as hyperalgesia, fatigue, malaise, and anorexia constitute major problems in the treatment of patients suffering from chronic inflammatory disease, little has been known about the signaling mechanisms by which the brain is activated during such conditions. Here, in an animal model of rheumatoid arthritis, we show that microsomal prostaglandin E-synthase, the inducible terminal isomerase in the prostaglandin E2-synthesizing pathway, is expressed in endothelial cells along the blood-brain barrier and in the parenchyma of the paraventricular hypothalamic nucleus. The endothelial cells but not the paraventricular hypothalamic cells displayed a concomitant induction of cyclooxygenase-2 and expressed interleukin-1 type 1 receptors, which indicates that the induction is due to peripherally released cytokines. In contrast to cyclooxygenase-2, microsomal prostaglandin E synthase had very sparse constitutive expression, suggesting that it could be a target for developing drugs that will carry fewer side effects than the presently available cyclooxygenase inhibitors. These findings, thus, suggest that immune-to-brain communication during chronic inflammatory conditions involves prostaglandin E2-synthesis both along the blood-brain barrier and in the parenchyma of the hypothalamic paraventricular nucleus and point to novel avenues for the treatment of the brain-elicited disease symptoms during these conditions.

    • Upon immune challenge the brain launches a wide range of responses, such as fever, anorexia, and hyperalgesia that serve to maintain homeostasis. While these responses are adaptive during acute infections, they may be destructive during chronic inflammatory conditions. Research performed during the last decade has given us insight into how the brain monitors the presence of a peripheral inflammation and the mechanisms underlying the brain-mediated acute-phase reactions. Here we give a brief review on this subject, with focus on the role of prostaglandin E2 produced in cells associated with the blood-brain barrier in immune-to-brain signaling. The recent advances in this field have not only elucidated the mechanisms behind the anti-pyretic and anti-hyperalgesic effects of cyclooxygenase inhibitors, but have also identified novel and more-selective potential drug targets.

  • Lipopolysaccharide-Induced Fever Depends on Prostaglandin E2 Production Specifically in Brain Endothelial Cells

    • Immune-induced prostaglandin E2 (PGE2) synthesis is critical for fever and other centrally elicited disease symptoms. The production of PGE2 depends on cyclooxygenase-2 and microsomal prostaglandin E synthase-1 (mPGES-1), but the identity of the cells involved has been a matter of controversy. We generated mice expressing mPGES-1 either in cells of hematopoietic or nonhematopoietic origin. Mice lacking mPGES-1 in hematopoietic cells displayed an intact febrile response to lipopolysaccharide, associated with elevated levels of PGE2 in the cerebrospinal fluid. In contrast, mice that expressed mPGES-1 only in hematopoietic cells, although displaying elevated PGE2 levels in plasma but not in the cerebrospinal fluid, showed no febrile response to lipopolysaccharide, thus pointing to the critical role of brain-derived PGE2 for fever. Immunohistochemical stainings showed that induced cyclooxygenase-2 expression in the brain exclusively occurred in endothelial cells, and quantitative PCR analysis on brain cells isolated by flow cytometry demonstrated that mPGES-1 is induced in endothelial cells and not in vascular wall macrophages. Similar analysis on liver cells showed induced expression in macrophages and not in endothelial cells, pointing at the distinct role for brain endothelial cells in PGE2 synthesis. These results identify the brain endothelial cells as the PGE2-producing cells critical for immune-induced fever.

    • The aims of this project have been to describe PM10 from studded tyre wear and to study their inflammatory effects in human airway cells. The VTI circular road simulator has been used to generate "clean" wear particles from two different pavements; asphalt concrete (ABT) and stone mastic asphalt (ABS), with granite respectively quartzite as the main stone materials. The advantage of using the road simulator is that the contribution from other sources can be minimised. During the project time, the project was expanded also to study particle generation by non-studded winter tyres (friction tyres) and two kinds of winter sanding agents, namely washed crushed stone and unwashed natural sand in combination with both studded and friction tyres. Most of these parts of the project are presented in VTI report 520. The results show that pavement wear by studded tyres generates about 40-50 times as much PM10 as that by friction tyres, but also that the ABT pavement generates several times more PM10 than the ABS pavement. The size distribution within PM10 has a maximum around 3-4 ?m and more than 95 % of the mass is larger than 1 ?m. PM10 is totally dominated by fresh stone material. A fraction of very small particles, with a number concentration peak at around 30-40 nm, was also discovered. Their origin is unknown, but since the number distribution shifted depending on which tyre type was used, a possible origin is the tyres. In the cell studies the inflammation potential of the wear particles was compared with PM10 from Hornsgatan in Stockholm and PM10 from a Stockholm subway station. Later on in the project, diesel particles were made available for comparison. The cell study results show that PM10 from the ABT pavement is at least as inflammatory as diesel particles and more inflammatory than PM10 from the subway. The PM10 from Hornsgatan generally had the highest potential, but PM10 from the ABT pavement was often in parity. This despite the fact that the Hornsgatan PM10 were the only particle sample where endotoxin could be detected. Endotoxin causes an additive effect in studies of inflammation potential. PM10 from the ABS pavement generally had a lower potential than PM10 from the ABT pavement.

Friction tyres on natural sand caused higher concentrations than studded tyres on washed crushed stone.     • In regions where studded tyres and traction material are used during winter, e.g. the Nordic countries, northern part of USA, Canada, and Japan, mechanically generated particles from traffic is the main reason for high particle concentrations in busy street- and road environments. In many Nordic municipalities the European environmental quality standard for inhalable particles (PM10) is exceeded due to these particles. In this study, particles from the wear of studded and studless friction tyres on two pavements and traction sanding were generated using a road simulator. The particles were characterized using particle sizers, PIXE and electron microscopy. Cell studies were conducted on particles sampled from the tests with studded tyres and compared with street environment, diesel exhaust and subway PM10, respectively. The results show that in the road simulator, where resuspension is minimised, studded tyres produce tens of times more particles than friction tyres. Chemical analysis of the sampled particles shows that the generated wear particles consists almost entirely of minerals from the pavement stone material, but also that S is enriched for the sub-micron particles and that Zn is enriched for friction tyres for all particles sizes. The chemical data can be used for source identification and apportionment in urban aerosol studies. A mode of ultra-fine particles was also present and is hypothesised to originate in the tyres. Further, traction material properties affect PM10 emission. The inflammatory potential of the particles from wear of pavements seems to depend on type of pavement and can be at least as potent as diesel exhaust particles. The results implies that there is a need and a good potential to reduce particle emission from pavement wear and winter time road and street operation by adjusting both studded tyre use as well as pavement and traction material properties.

    • Under slutet på 90-talet genomfördes mätningar av luften partikelhalter i sex svenska städer där syftet var att studera vilka halter som förekommer i gatu-, respektive taknivå och studera partiklarnas ursprung (Areskoug m. fl. 2001). Något förvånande visade det sig att den tydligt dominerande källan till partiklar i gatunivå under vinterhalvåret då halterna är som högst, inte är avgasrelaterade partiklar utan härrör från slitage av vägbana, däck och bromsar. Slitage av vägbanan orsakad av dubbdäcksanvändning bidrar mycket starkt till partikelhalterna, men även vintersandningen är en bidragande orsak. Den nyvunna kunskapen om slitagepartiklarnas betydelse för partikelhalterna kräver ökad kunskap om vilka faktorer som styr bildningen, om partiklarnas egenskaper, om deras eventuella betydelse för hälsoeffekter och om vilka åtgärder som kan bidra till att sänka bidraget.Projektet WearTox är ett av de första som Vägverket initierade för att fylla dessa kunskapsluckor. Projektet har utförts av VTI i samarbete med Lunds universitet och Linköpings universitet.Syftet med projektet har varit att studera den toxiska potentialen hos slitagepartiklar från beläggning och däck hos exponerade mänskliga luftvägsceller. slitagepartiklarnas fysikaliska, kemiska och morfologiska egenskaper. Under arbetets gång tillkom önskemål om karaktärisering av ytterligare kombinationer av däck vägbana och även från försök med sandad vägbana. Dessa tilläggsbeställningar inkluderade dock inte toxikologiska studier utan begränsade sig till karaktäriseringen av partiklarna.

    • Svenska Historikermötet i Umeå 14–16 juni 2023Titel: Högskolan Dalarna och folkskoleseminariets historia – Perspektiv på lärarutbildningen i Falun, ca 1875–1970Den som har inflytande över utformandet av en lärarutbildning och blivande lärares kunskapssyn, kan också komma att påverka framtida generationers sätt att se på samhället och världen (Hartman 2012, Bejerot & Hasselbladh 2020). Vid folkskoleseminariet i Falun bedrevs lärarutbildning från etablerandet 1875 fram till det att verksamheten succesivt övergick i lärarhögskolans regi 1968–70.  I en mening sammanfaller dessa nära på hundra år av lärarutbildning därmed också med framväxten av det moderna Sverige.I antologiprojektet ’Högskolan Dalarna och folkskoleseminariets historia’ riktas fokus mot en rad tidigare outforskade aspekter av hur seminarietraditionen, i ett specifikt lokalsamhälle, etableras och förändras under olika tidsperioder. Enligt tidigare forskning hade den 1842 inrättade folkskolan uppgiften att legitimera ett traditionellt, religiöst och auktoritärt samhällssystem (Petterson 1993, Lundahl 2005). Längre fram i tiden, inte minst från sekelskiftet 1900 och framåt, kom folkskolan dock att alltmer betraktas som ett centralt redskap i det moderna samhällets utvecklande av medborgares agens, kvinnlig frigörelse och samhällsutveckling (Linne 1996). Under femton år med start 1864 etablerades i Sverige fem folkskoleseminarier för kvinnor. Först ut var Stockholm och Skara som 1864 ombildades från att utbilda män till att utbilda kvinnor. Därefter etablerades folkskollärarinneseminarier i både Falun och Kalmar år 1875 och fyra år senare följde ett femte i Umeå (Sörensen 1942). Seminarieetableringarna i Falun och övriga orter innebar en ökning i andelen kvinnliga lärare under slutet av 1800-talet och i förlängningen kom den patriarkala skolordningen att utmanas (Florin 1987).Beslutet den 30 april 1875 att anlägga ett folkskoleseminarium i Falun, säger något om behovet av lärare på den svenska landsbygden, men också något om Faluns historiska bakgrund som lärdomsstad (Claesson 2015, Liljas 2019:2021, Ödman 1995). Möjligen finns också lokala och aktörsburna förklaringar till den utpräglat progressiva profil som med tiden kom att växa fram vid folkskoleseminariet i Falun (Linné 1996:2006). I projektets delstudier görs exempel på pregnanta avsteg från folkskolans normer av framträdande lärarutbildare som Valborg Olander och Anna Sörensen. Med utgångspunkt i ett mycket rikt arkivmaterial blir det också möjligt att följa hur de och andra lärarutbildare i valet av uppgifter och utformningen av textnära kommentarer söker påverka de blivande lärarinnorna i deras sätt att se på kunskap, undervisning och lärarskap. Utifrån än mer kritiskt orienterade perspektiv på verksamheten granskas också urvalsprinciperna för rekryteringen av nya lärarinnor och interkulturella aspekter av den faktiska verksamheten.Genom dessa nedslag berörs därmed också hur professionaliseringen av läraryrket tar form i relation till ett bildningsanspråk - vilket ofta tycks gravitera mot medborgerliga och könsutjämnande principer. Under den föreslagna sessionen till historikermötet i Umeå presenteras fem av dessa bidrag:Juvas Marianne Liljas presentation fokuserar på Valborg Olanders (1861-1943) reformerande insatser vid folkskoleseminariet i Falun och har titeln ’’’Det varma hjärtat”: Valborg Olanders pedagogiska arv i Faluseminariets historia’.I presentationen ’Folkskolelärarinneseminarier i Sverige: en jämförelse mellan Falun och Umeå, 1875–1915’ intar Emil Marklund en komparativ ansats. Folkskollärarinneseminariet i Umeå grundades 1879, blott fyra år efter seminariet i Falun. Utifrån rubriken ’Kunskaps- och undervisningsideal i förändring: Pedagogiska examensuppsatser vid Falu folkskoleseminarium 1915–1937’ analyserar Anders Persson de skriftliga kommentarer som dåtidens lärarutbildare lämnar på de blivande lärarinnornas texter.I ´En lärare blir till. Om ideal och normer för blivande lärare på folkskollärarseminariet i Falun år 1876–1909´presenterar Ulrika Norburg exempel på hur rekryteringsprocessen av blivande lärarinnor vid seminariet gick till.Anders Blomqvist presentation ’Perspektiv på seminariet i krigs- och brytningstider 1940–1960:  genus, klass och etnicitet’ fokuserar på erfarenheter från Faluseminariet under tiden för Andra världskriget och efterföljande år. Ordförande: Simon Lundberg, Umeå universitet.Discussant: Anna Götlind, Stockholms universitet.

Lately, wear particles generated from traffic have been recognized to be a major contributing source to the overall particle load, especially in the Nordic countries were studded tires are used. In this work, we investigated the inflammatory effect of PM10 generated from the wear of studded tires on two different types of pavement. As comparison, we also investigated PM10 from a traffic-intensive street, a subway station, and diesel exhaust particles (DEP). Human monocyte-derived macrophages, nasal epithelial cells (RPMI 2650), and bronchial epithelial cells (BEAS-2B) were exposed to the different types of particles, and the secretion of IL-6, IL-8, IL-10, and TNF-α into the culture medium was measured. The results show a significant release of cytokines from macrophages after exposure for all types of particles. When particles generated from asphalt/granite pavement were compared to asphalt/quartzite pavement, the granite pavement had a significantly higher capacity to induce the release of cytokines. The granite pavement particles induced cytokine release at the same magnitude as the street particles did, which was higher than what particles from both a subway station and DEP did. Exposure of epithelial cells to PM 10 resulted in a significant increase of TNF-α secreted from BEAS-2B cells for all types of particles used (DEP was not tested), and the highest levels were induced by subway particles. None of the particle types were able to evoke detectable cytokine release from RPMI 2650 cells. The results indicate that PM10 generated by the wear of studded tires on the street surface is a large contributor to the cytokine-releasing ability of particles in traffic-intensive areas and that the type of pavement used is important for the level of this contribution. Furthermore, the airway inflammatory potential of wear particles from tires and pavement might be of a greater magnitude than that of DEP.

Lately, wear particles generated from traffic have been recognized as a major contributing source to the overall particle load, especially in the Nordic countries where studded tires are used. The aim of this study was to further investigate and compare the ability to induce inflammatory mediators of different traffic-related wear particles collected from an urban street, a subway station, and studded tire-pavement wear. Inflammatory effects were measured as induction of nitric oxide (NO), IL-6, TNF-α, arachidonic acid (AA), and lipid peroxidation after exposure of the murine macrophage like cell line RAW 264.7. In addition, the redox potential of the particles was measured in a cell-free system. The results show that all particles tested induce IL-6, TNF-α, and NO, and those from the urban street were the most potent ones. In contrast, particles collected from a subway station were most potent to induce lipid peroxidation, A A release, and formation of ROS. Particles from studded tire-pavement wear, generated using a road simulator, were able to induce inflammatory cytokines, NO, lipid peroxidation, and ROS formation. Interestingly, particles generated from pavement containing granite as the main stone material were more potent than those generated from pavement containing quartzite as the main stone material.

    • The mode of action of paracetamol (acetaminophen), which is widely used for treating pain and fever, has remained obscure, but may involve several distinct mechanisms, including cyclooxygenase inhibition and transient receptor potential ankyrin 1 (TRPA1) channel activation, the latter being recently associated with paracetamol’s propensity to elicit hypothermia at higher doses. Here, we examined whether the antipyretic effect of paracetamol was due to TRPA1 activation or cyclooxygenase inhibition. Treatment of wild-type and TRPA1 knockout mice rendered febrile by immune challenge with LPS with a dose of paracetamol that did not produce hypothermia (150 mg/kg) but is known to be analgetic, abolished fever in both genotypes. Paracetamol completely suppressed the LPS-induced elevation of prostaglandin E2 in the brain and also reduced the levels of several other prostanoids. The hypothermia induced by paracetamol was abolished in mice treated with the electrophile-scavenger N-acetyl cysteine. We conclude that paracetamol’s antipyretic effect in mice is dependent on inhibition of cyclooxygenase activity, including the formation of pyrogenic prostaglandin E2, whereas paracetamol-induced hypothermia likely is mediated by the activation of TRPA1 by electrophilic metabolites of paracetamol, similar to its analgesic effect in some experimental paradigms.

  • The grade of individual prostate cancer lesions predicted by magnetic resonance imaging and positron emission tomography

    • Background: Multiparametric magnetic resonance imaging (mpMRI) and positron emission tomography (PET) are widely used for the management of prostate cancer (PCa). However, how these modalities complement each other in PCa risk stratification is still largely unknown. We aim to provide insights into the potential of mpMRI and PET for PCa risk stratification.Methods: We analyzed data from 55 consecutive patients with elevated prostate-specific antigen and biopsy-proven PCa enrolled in a prospective study between December 2016 and December 2019. [68Ga]PSMA-11 PET (PSMA-PET), [11C]Acetate PET (Acetate-PET) and mpMRI were co-registered with whole-mount histopathology. Lower- and higher-grade lesions were defined by International Society of Urological Pathology (ISUP) grade groups (IGG). We used PET and mpMRI data to differentiate between grades in two cases: IGG 3 vs. IGG 2 (case 1) and IGG ≥ 3 vs. IGG ≤ 2 (case 2). The performance was evaluated by receiver operating characteristic (ROC) analysis.Results: We find that the maximum standardized uptake value (SUVmax) for PSMA-PET achieves the highest area under the ROC curve (AUC), with AUCs of 0.72 (case 1) and 0.79 (case 2). Combining the volume transfer constant, apparent diffusion coefficient and T2-weighted images (each normalized to non-malignant prostatic tissue) results in AUCs of 0.70 (case 1) and 0.70 (case 2). Adding PSMA-SUVmax increases the AUCs by 0.09 (p < 0.01) and 0.12 (p < 0.01), respectively.Conclusions: By co-registering whole-mount histopathology and in-vivo imaging we show that mpMRI and PET can distinguish between lower- and higher-grade prostate cancer, using partially discriminative cut-off values.

    • The dominant road traffic particle sources are wear particles from the road and tire interface, and from vehicle brake pads. The aim of this work was to investigate the effect of road and brake wear particles on pulmonary function and biomarkers in isolated perfused rat lungs. Particles were sampled from the studded tire wear of three road pavements containing different rock materials in a road simulator; and from the wear of two brake pad materials using a pin-on-disk machine. Isolated rat lungs inhaled the coarse and fine fractions of the sampled particles resulting in an estimated total particle lung dose of 50 μg. The tidal volume (TV) was measured during the particle exposure and the following 50 min. Perfusate and BALF were analyzed for the cytokines TNF, CXCL1 and CCL3. The TV of lungs exposed to rock materials was significantly reduced after 25 min of exposure compared to the controls, for quartzite already after 4 min. The particles of the heavy-duty brake pads had no effect on the TV. Brake particles resulted in a significant elevation of CXCL1 in the perfusate. Brake particles showed significant elevations of all three measured cytokines, and quartzite showed a significant elevation of TNF in BALF. The study shows that the toxic effect on lungs exposed to airborne particles can be investigated using measurements of tidal volume. Furthermore, the study shows that the choice of rock material in road pavements has the potential to affect the toxicity of road wear PM10.

    • Using dual-labeling in situ hybridization histochemistry, the neurotransmitter expression of immune-responsive neurons in the pontine parabrachial nucleus, a major relay for interoceptive information, was investigated. Intravenous injection of bacterial wall lipopolysaccharide resulted in dense c-fos mRNA expression in the external lateral parabrachial nucleus, and a majority of the c-fos expressing cells also expressed calcitonin gene-related peptide (CGRP) mRNA. In contrast CGRP-posi- tive cells in the adjoining external medial subnucleus were c-fos negative. Taken together with previous hodological and behavioral studies, these data suggest that CGRPergic parabrachial neurons may mediate lipopolysaccharide-induced anorexia by means of their projection to central nucleus of the amygdala.

    • Objective: PET/CT and multiparametric MRI (mpMRI) are important diagnostic tools in clinically significant prostate cancer (csPC). The aim of this study was to compare csPC detection rates with [68Ga]PSMA-11-PET (PSMA)-PET, [11C] Acetate (ACE)-PET, and mpMRI with histopathology as reference, to identify the most suitable imaging modalities for subsequent hybrid imaging. An additional aim was to compare inter-reader variability to assess reproducibility.Methods: During 2016–2019, all study participants were examined with PSMA-PET/mpMRI and ACE-PET/CT prior to radical prostatectomy. PSMA-PET, ACE-PET and mpMRI were evaluated separately by two observers, and were compared with histopathology-defined csPC. Statistical analyses included two-sided McNemar test and index of specific agreement.Results: Fifty-five study participants were included, with 130 histopathological intraprostatic lesions >0.05 cc. Of these, 32% (42/130) were classified as csPC with ISUP grade ≥2 and volume >0.5 cc. PSMA-PET and mpMRI showed no difference in performance (P = 0.48), with mean csPC detection rate of 70% (29.5/42) and 74% (31/42), respectively, while with ACE-PET the mean csPC detection rate was 37% (15.5/42). Interobserver agreement was higher with PSMA-PET compared to mpMRI [79% (26/33) vs 67% (24/38)]. Including all detected lesions from each pair of observers, the detection rate increased to 90% (38/42) with mpMRI, and 79% (33/42) with PSMA-PET.Conclusion: PSMA-PET and mpMRI showed high csPC detection rates and superior performance compared to ACE-PET. The interobserver agreement indicates higher reproducibility with PSMA-PET. The combined result of all observers in both PSMA-PET and mpMRI showed the highest detection rate, suggesting an added value of a hybrid imaging approach.

    • Background and purpose: The diagnostic accuracy of new imaging techniques requires validation, preferably by histopathological verification. The aim of this study was to develop and present a registration procedure between histopathology and in-vivo magnetic resonance imaging (MRI) of the prostate, to estimate its uncertainty and to evaluate the benefit of adding a contour-correcting registration.Materials and methods: For twenty-five prostate cancer patients, planned for radical prostatectomy, a 3D-printed prostate mold based on in-vivo MRI was created and an ex-vivo MRI of the specimen, placed inside the mold, was performed. Each histopathology slice was registered to its corresponding ex-vivo MRI slice using a 2D-affine registration. The ex-vivo MRI was rigidly registered to the in-vivo MRI and the resulting transform was applied to the histopathology stack. A 2D deformable registration was used to correct for specimen distortion concerning the specimen's fit inside the mold. We estimated the spatial uncertainty by comparing positions of landmarks in the in-vivo MRI and the corresponding registered histopathology stack.Results: Eighty-four landmarks were identified, located in the urethra (62%), prostatic cysts (33%), and the ejaculatory ducts (5%). The median number of landmarks was 3 per patient. We showed a median in-plane error of 1.8 mm before and 1.7 mm after the contour-correcting deformable registration. In patients with extraprostatic margins, the median in-plane error improved from 2.1 mm to 1.8 mm after the contour-correcting deformable registration.Conclusions: Our registration procedure accurately registers histopathology to in-vivo MRI, with low uncertainty. The contour-correcting registration was beneficial in patients with extraprostatic surgical margins.

    • This report is an outcome of a major research effort within the Swedish National Road Vehicle Emission Research Programme EMFO, carried out 2005-2008 in collaboration between IVL, Lund University, the City of Stockholm and VTI. Within the project extensive data have been collected by a variety of methods for measuring, sampling and analysing the chemical composition of primarily three different fractions of particulate matter - PM10, PM2.5, PM1. Emphasis has been on the PM10 fraction, in the case of which preferably the major Swedish cities experience significant problems with complying with the legally binding air quality standards. The collected data originate from indoor measurements in controlled runs with the VTI circular road simulator as well as ambient air measurements at both street and roof level in a variety of Swedish cities. Based on elemental (metals etc.) source profiles of various sources to the different particle fractions, derived from the literature or from measurements carried out within the project, several different receptor models (e.g. COPREM, PMF) were applied to the collected data to derive the contributions from exhaust, brake wear, tyre wear, road surface wear, long-range transport etc., to the measured concentrations of PM10 and other particle fractions in urban environments. Furthermore, from the measurements, emission factors (expressed in grams per vehkm) for the various particle fractions, as well as for a large number of contained metals (about 30 metals) were derived for a major city street in Stockholm. Total emission factors as well as emission factors for each of the different source types (exhaust, brake wear, tyre wear and road surface wear) have also been derived from the measurements.

    • BACKGROUND: A substantial proportion of individuals with COPD have never smoked, and it is implied to be more common than previously anticipated but poorly studied.AIM: To describe the process of recruitment of never-smokers with COPD from a population-based cohort (n = 30 154).METHODS: We recruited never-smokers with COPD, aged 50-75 years, from six University Hospitals, based on: 1) post broncho-dilator forced expiratory volume in 1 second/forced vital capacity (FEV1/FVC) < 0.70 and 2) FEV1 50-100% of predicted value and 3) being never-smokers (self-reported). In total 862 SCAPIS participants were identified, of which 652 were reachable and agreed to a first screening by telephone. Altogether 128 (20%) were excluded due to previous smoking or declined participation. We also applied a lower limit of normal (LLN) of FEV1/FVC (z-score<-1.64) according to the Global Lung Initiative to ensure a stricter definition of airflow obstruction.RESULTS: Data on respiratory symptoms, health status, and medical history were collected from 492 individuals, since 32 were excluded at a second data review (declined or previous smoking), prior to the first visit. Due to not matching the required lung function criteria at a second spirometry, an additional 334 (68%) were excluded. These exclusions were by reason of: FEV1/FVC ≥0.7 (49%), FEV1 > 100% of predicted (26%) or z-score ≥ -1,64 (24%). Finally, 154 never-smokers with COPD were included: 56 (36%) women, (mean) age 60 years, FEV1 84% of predicted, FEV1/FVC: 0.6, z-score: -2.2, Oxygen saturation: 97% and BMI: 26.8 kg/m2.CONCLUSIONS: The challenges of a recruitment process of never-smokers with COPD were shown, including the importance of correct spirometry testing and strict inclusion criteria. Our findings highlight the importance of repeated spirometry assessments for improved accuracy in diagnosing COPD.

    • BACKGROUND: The delineation of intraprostatic lesions is vital for correct delivery of focal radiotherapy boost in patients with prostate cancer (PC). Errors in the delineation could translate into reduced tumour control and potentially increase the side effects. The purpose of this study is to compare PET-based delineation methods with histopathology.MATERIALS AND METHODS: The study population consisted of 15 patients with confirmed high-risk PC intended for prostatectomy. [68Ga]-PSMA-PET/MR was performed prior to surgery. Prostate lesions identified in histopathology were transferred to the in vivo [68Ga]-PSMA-PET/MR coordinate system. Four radiation oncologists manually delineated intraprostatic lesions based on PET data. Various semi-automatic segmentation methods were employed, including absolute and relative thresholds, adaptive threshold, and multi-level Otsu threshold.RESULTS: The gross tumour volumes (GTVs) delineated by the oncologists showed a moderate level of interobserver agreement with Dice similarity coefficient (DSC) of 0.68. In comparison with histopathology, manual delineations exhibited the highest median DSC and the lowest false discovery rate (FDR) among all approaches. Among semi-automatic approaches, GTVs generated using standardized uptake value (SUV) thresholds above 4 (SUV > 4) demonstrated the highest median DSC (0.41), with 0.51 median lesion coverage ratio, FDR of 0.66 and the 95th percentile of the Hausdorff distance (HD95%) of 8.22 mm.INTERPRETATION: Manual delineations showed a moderate level of interobserver agreement. Compared to histopathology, manual delineations and SUV > 4 exhibited the highest DSC and the lowest HD95% values. The methods that resulted in a high lesion coverage were associated with a large overestimation of the size of the lesions.

    • The fourth generation of the Sloan Digital Sky Survey (SDSS-IV) has been in operation since 2014 July. This paper describes the second data release from this phase, and the 14th from SDSS overall (making this Data Release Fourteen or DR14). This release makes the data taken by SDSS-IV in its first two years of operation (2014-2016 July) public. Like all previous SDSS releases, DR14 is cumulative, including the most recent reductions and calibrations of all data taken by SDSS since the first phase began operations in 2000. New in DR14 is the first public release of data from the extended Baryon Oscillation Spectroscopic Survey; the first data from the second phase of the Apache Point Observatory (APO) Galactic Evolution Experiment (APOGEE-2), including stellar parameter estimates from an innovative data-driven machine-learning algorithm known as "The Cannon"; and almost twice as many data cubes from the Mapping Nearby Galaxies at APO (MaNGA) survey as were in the previous release (N = 2812 in total). This paper describes the location and format of the publicly available data from the SDSS-IV surveys. We provide references to the important technical papers describing how these data have been taken (both targeting and observation details) and processed for scientific use. The SDSS web site (www.sdss.org) has been updated for this release and provides links to data downloads, as well as tutorials and examples of data use. SDSS-IV is planning to continue to collect astronomical data until 2020 and will be followed by SDSS-V.

    • A double-labelling procedure combining immunohistochemical staining with in situ hybridization using a radiolabelled cRNA probe was employed to demonstrate oestrogen receptor-like immunoreactivity and preproenkephalin-A mRNA in the medullary and spinal dorsal horn of female rats. Both markers labelled large numbers of neurons in the substantia gelatinosa and its trigeminal homologue. Many of these neurons were double-labelled, displaying both oestrogen receptor-like-immunoreactivity and preproenkephalin-A mRNA; cell counts showed that 40-60% of the of the oestrogen receptor-like-immunoreactive cells in the superficial laminae also were labelled for preproenkephalin-A mRNA, and that 60-70% of the preproenkephalin-A mRNA-labelled neurons in the same laminae displayed oestrogen receptor-like immunoreactivity. Previous studies have shown that oestrogen receptors can bind to the promoter region of the preproenkephalin-A gene, and studies on the hypothalamus have demonstrated that oestrogen regulates enkephalin expression in select neuronal populations. The present results demonstrate that enkephalinergic neurons in the superficial dorsal horn contain oestrogen receptors and suggest that oestrogen may play an important role in the modulation of sensory and nociceptive processing in the lower medulla and spinal cord.

    • Enkephalin-synthesizing neurons in the super®cial laminae of the spinal and trigeminal dorsal horn are critical components of the endogenous pain-modulatory system. We have previously demonstrated that these neurons display intracellular estrogen receptors, suggesting that estrogen can potentially influence their enkephalin expression. By using Northern blot, we now show that a bolus injection of estrogen results in a rapid increase in spinal cord enkephalin mRNA levels in ovariectomized female rats. Thus, 4 h after estrogen administration the enkephalin mRNA-expression in the lumbar spinal cord was on average 68% higher (P , 0:05) than in control animals injected with vehicle only. A small increase in the amount of enkephalin mRNA was also seen after 8 h (P , 0:05), whereas no difference between estrogen-injected and control animals was found after 24 h or at time periods shorter than 4 h. Taken together with the previous anatomical data, the present findings imply that estrogen has an acute effect on spinal opioid levels in areas involved in the transmission of nociceptive information.

    • Estrogens have a remarkably wide range of actions in the mammalian brain. They not only play a pivotal role in reproductive behavior and sexual differentiation, but also contribute to e.g. thermoregulation, feeding, memory, neuronal survival and the perception of somatosensory stimuli. A multitude of studies on both animals and human subjects has demonstrated potential effects of gonadal hormones, such as estrogens, on pain transmission. These effects most likely involve multiple neuroanatomical circuits as well as diverse neurochemical systems and therefore need to be evaluated specifically in relation to the localization and intrinsic characteristics of the neurons engaged. The overall aim of this thesis is to gain specific knowledge of the possible cellular mechanisms by which estrogens may influence the transmission of nociceptive stimuli at the level of the spinal cord. The estrogen receptors, by which estrogens regulate non-genomic as well as genomic mechanisms, are crucial to estrogen signaling in general and essential to the estrogen-induced effects in the brain. In Paper I, we use immunohistochemistry to label neurons containing estrogen receptor-! (ERα) in the medullary and spinal dorsal horn of female rats. Large numbers of ER!-expressing neurons were found in lamina I and lamina II, i.e. in the areas involved in the processing of primary afferent nociceptive information. This distribution in part overlaps that of enkephalin, a potent pain-inhibiting endogenous opioid. The effects of gonadal hormones on pain modulation may, to a great extent, be blocked by the opioid antagonist naloxone, suggesting an involvement of the endogenous opioid system in the prosecution of hormonal pain regulation. By combining immunohistochemical labeling of ERα with in situ hybridization of preproenkephalin mRNA (Paper II), we demonstrate that the majority of enkephalinergic neurons in the superficial laminae of the spinal and medullary dorsal horn express ER!. This co-localization and the fact that the preproenkephalin gene contains a sequence that binds ERs, suggest that estrogens may potentially regulate enkephalin expression in these cells. This is further supported by the findings in Paper III in which we show that a single subcutaneous injection of estradiol induces a significant increase (on average 68%) in preproenkephalin mRNA content in the spinal cord after 4 hours. The expression of the enkephalin gene in the spinal cord is thus sensitive to fluctuating estradiol levels. In Paper IV, a noxious injection of formalin is used to induce activation of a neuronal population involved in nociceptive transmission from the face. By using a dual-labeling immunohistochemistry protocol, we were able to identify ER!-expressing cells within this neuronal population suggesting that nociceptive-responsive neurons in the medullary dorsal horn express ER!. In all, our findings provide morphological as well as biochemical evidence in support for an estrogen-dependent modulation of nociceptive processing at the level of the dorsal horn.

    • Using an immunohistochemical technique, we demonstrate that large numbers of neurons in the laminar spinal trigeminal nucleus and spinal gray matter of the female rat express estrogen receptors (ER). Densely packed ER-immunoreactive neurons were present in lamina II, but labeled neurons were also present in lamina I, the neck of the dorsal horn, and in lamina X. Labeling was present throughout the length of the spinal cord, with the exception of segments caudal to S1, which were unlabeled. The distribution of ER-containing neurons to areas that are involved in processing of primary afferent nociceptive information suggests that the pain modulatory effects of estrogen may be exerted at the spinal level.

    • Estrogens exert a substantial influence on the transmission of nociceptive stimuli and the susceptibility to pain disorders as made evident by studies in both animals and human subjects. The estrogen receptor (ER) seems to be of crucial importance to the cellular mechanisms underlying such an influence. However, it has not been clarified whether nociceptive neurons activated by pain express ERs. In this study, a noxious injection of formalin was given into the lower lip of female rats, thereby activating nociceptive neurons in the trigeminal subnucleus caudalis as demonstrated by immunohistochemical labeling of Fos. Using a dual-label immunohistochemistry protocol ERalpha-containing cells were visualized in the same sections. In the superficial layers of the medullary dorsal horn, 12% of ERalpha-labeled cells, mainly located in lamina II, also expressed noxious-induced Fos. These findings show that nociceptive-responsive neurons in the medullary dorsal horn express ERalpha, thus providing a possible morphological basis for the hypothesis that estrogens directly regulate pain transmission at this level.

  • Investigation of fire emissions from Li-ion batteries

    • This report presents an investigation on gases emitted during Lithium-ion battery fires. Details of the calibration of an FTIR instrument to measure HF, POF3 and PF5 gases are provided as background to the minimum detection limits for each species. The use of FTIR in tests has been verified by repeating experiments reported in the literature. The study reports on gases emitted both after evaporation and after ignition of the electrolyte fumes. Tests were conducted where electrolyte is injected into a propane flame and the influence of the addition of water is studied. Finally three types of battery cells were burnt and emission of fluorine and/or phosphorous containing species quantified.

    • Batteries, in particular lithium-ion (Li-ion) batteries, are seen as an alternative to fossil fuels in the automotive sector. Li-ion batteries, however, have some safety issues including possible emissions of toxic fluorine-containing compounds during fire and other abuse situations. This paper demonstrates the possibilities to use the Fourier transform infrared technique to assess some of the most important compounds, including hydrogen fluoride and the far less often measured POF3 and PF5. The study is conducted in the cone calorimeter with different solvents used in Li-ion batteries. The measurements show that, in addition to hydrogen fluoride, with a known high toxicity, POF3 is emitted and can be quantified using Fourier transform infrared.
