    • Intestinal mucus barriers normally prevent microbial infections but are sensitive to diet-dependent changes in the luminal environment. Here we demonstrate that mice fed a Western-style diet (WSD) suffer regiospe-cific failure of the mucus barrier in the small intestinal jejunum caused by diet-induced mucus aggregation. Mucus barrier disruption due to either WSD exposure or chromosomal Muc2 deletion results in collapse of the commensal jejunal microbiota, which in turn sensitizes mice to atypical jejunal colonization by the enteric pathogen Citrobacter rodentium. We illustrate the jejunal mucus layer as a microbial habitat, and link the re-giospecific mucus dependency of the microbiota to distinctive properties of the jejunal niche. Together, our data demonstrate a symbiotic mucus-microbiota relationship that normally prevents jejunal pathogen colo-nization, but is highly sensitive to disruption by exposure to a WSD.

    • Denna rapport är ett resultat av en förstudie kring en logistik- och transportlösning (som kan realiseras i form av en gemensam hubb) för aktörer som är placerade i evenemangsstråket i Göteborg. Rapporten svarar på frågor kring nulägesanalys av aktörernas logistiklösningar, leveranstrafik inom evenemangsstråket, samt hur leveranstrafik kan se ut under 2030+ om inga förändringar i leverans/logistikupplägget görs. En viktig slutsats som framgår är att det krävs åtgärder som minskar leveranstrafiken, eftersom elektrifiering av fordon inte kan eliminera trängsel och buller som står för den största delen av de samhällsekonomiska kostnaderna. Utan förändringar i logistikupplägget och fordonsflottan förväntas de samhällsekonomiska kostnaderna öka med cirka 30%. Förändringar i logistikupplägget kan nås genom ett optimerad trafikflöde, vilket i sin tur kan realiseras genom en gemensam samlastningshubb för små och fragmenterade flöden som kan minska antalet anlöp och fordonstrafiken i området. Baserat på intervjuer med aktörerna, studiebesök, studerande av underlag, workshops och analys av logistikbehoven, kan det sammanfattas att verksamheterna, utmaningar och logistikbehoven i stort sett är ganska lika. En gemensam extern hubb kan frigöra yta hos verksamheterna som kan användas för att skapa nya affärsmöjligheter. En möjlighet för att upphandla en tredjeparts logistik- och transportlösning är att bygga ett konsortium där alla aktörer delar på kostnaden för terminalhantering samt samlastning/samkörning. Det finns stora möjligheter att hitta lösningar för logistikhubb som uppfyller kraven på geografisk närhet och säkra leveranser i trafiksystemet – och det är klokt att anpassa leveranstider för att undvika trängsel. Det finns en vilja hos aktörerna att minska transporterna, anpassa sin verksamhet för effektivisering vid mottagning och förvaring, ställa om mot fossilfrihet och på det sättet bidra till transporteffektivitet som krävs för att nå mål inom klimatneutralitet 2030. Det finns stora behov för förändring i trafikflödet inom GGCZ och det finns en stor potential att optimera det genom att konsolidera och styra leveranser.

  • A Comparison of Rule-based Analysis with Regression Methods in Understanding the Risk Factors for Study Withdrawal in a Pediatric Study

    • Regression models are extensively used in many epidemiological studies to understand the linkage between specific outcomes of interest and their risk factors. However, regression models in general examine the average effects of the risk factors and ignore subgroups with different risk profiles. As a result, interventions are often geared towards the average member of the population, without consideration of the special health needs of different subgroups within the population. This paper demonstrates the value of using rule-based analysis methods that can identify subgroups with heterogeneous risk profiles in a population without imposing assumptions on the subgroups or method. The rules define the risk pattern of subsets of individuals by not only considering the interactions between the risk factors but also their ranges. We compared the rule-based analysis results with the results from a logistic regression model in The Environmental Determinants of Diabetes in the Young (TEDDY) study. Both methods detected a similar suite of risk factors, but the rule-based analysis was superior at detecting multiple interactions between the risk factors that characterize the subgroups. A further investigation of the particular characteristics of each subgroup may detect the special health needs of the subgroup and lead to tailored interventions.

  • Experienced consequences of being diagnosed with ADHD as an adult - a qualitative study

    • Background: Despite increasing knowledge of attention deficit hyperactivity disorder (ADHD) across the life span, there is still little research on adults' own experiences of being diagnosed with ADHD. The aim of the present study was to explore and describe patients' experiences and perceptions of being diagnosed with ADHD in adulthood. The study can be seen as an attempt to validate the diagnosis from a patient perspective. Methods: Twenty-one adults diagnosed with ADHD were individually interviewed. The interviews were open-ended and exploratory, analysed with a qualitative phenomenographical approach, and the results were described in categories. Results: Positive experiences were dominant, but there was a complex intra-and inter-individual variation of experiences. Descriptions focused on the diagnosis, on identity, and on life. The diagnosis was described as explaining a previously inexplicable life history, but was also questioned, both as a phenomenon and in relation to the individual (the diagnosis in focus). It was experienced as providing self-knowledge and increased value, but could also cause devaluation and concern about identity (identity in focus). It meant help to achieve a better life, but was also perceived to restrict possibilities and cause disappointment over lack of professional help. It could lead to a wish for an earlier diagnosis that could have spared suffering, as well as to a changed view of the participants' relatives (life in focus). All but one of the interviewees expressed important positive consequences of being diagnosed with ADHD. About half of them acknowledged negative aspects of being diagnosed, but none regretted going through the neuropsychiatric evaluation. Conclusions: From a patient perspective, there are major positive consequences of being diagnosed with ADHD, compared to the undiagnosed situation. Knowledge of the individual's combination of experiences is important for professionals, as these experiences can affect well-being and interfere with treatment. Negative experiences in particular might need to be addressed in the treatment work.

  • Evidence for production of single top quarks and first direct measurement of vertical bar V-tb vertical bar

    • The D0 Collaboration presents first evidence for the production of single top quarks at the Fermilab Tevatron < p(p)over bar > collider. Using a 0.9 fb(-1) dataset, we apply a multivariate analysis to separate signal from background and measure sigma(< p(p)over bar >-> tb+X,tqb+X)=4.9 +/- 1.4 pb. The probability to measure a cross section at this value or higher in the absence of a signal is 0.035%, corresponding to a 3.4 standard deviation significance. We use the cross section measurement to directly determine the Cabibbo-Kobayashi-Maskawa matrix element that describes the Wtb coupling and find 0.68 <= 1 at 95% C.L. within the standard model.

    • We present the first experimental discrimination between the 2e/3 and 4e/3 top quark electric charge scenarios, using top quark pairs (t (t) over bar) produced in p (p) over bar collisions at root s =1.96 TeV by the Fermilab Tevatron Collider. We use 370 pb(-1) of data collected by the D0 experiment and select events with at least one high transverse momentum electron or muon, high transverse energy imbalance, and four or more jets. We discriminate between b- and (b) over bar -quark jets by using the charge and momenta of tracks within the jet cones. The data are consistent with the expected electric charge, |q|=2e/3. We exclude, at the 92% C.L., that the sample is solely due to the production of exotic quark pairs Q (Q) over bar with |q|=4e/3. We place an upper limit on the fraction of Q (Q) over bar pairs rho < 0.80 at the 90% C.L.

  • Lifetime difference and CP-violating phase in the B-s(0) system

    • From an analysis of the decay B-s(0)-> J/psi phi, we obtain the width difference between the light and heavy mass eigenstates Delta Gamma equivalent to(Gamma(L)-Gamma(H))=0.17 +/- 0.09(stat)+/- 0.02(syst) ps(-1) and the CP-violating phase phi(s)=-0.79 +/- 0.56(stat)(-0.01)(+0.14)(syst). Under the hypothesis of no CP violation (phi(s)equivalent to 0), we obtain 1/Gamma=tau/(B-s(0))=1.52 +/- 0.08(stat)(-0.03)(+0.01)(syst) ps and Delta Gamma=0.12(-0.10)(+0.08)(stat)+/- 0.02(syst) ps(-1). The data sample corresponds to an integrated luminosity of about 1.1 fb(-1) accumulated with the D0 detector at the Fermilab Tevatron collider. This is the first direct measurement of the CP-violating mixing phase in the B-s(0) system.

  • Measurement of the charge asymmetry in semileptonic B-s(0) decays

    • We have performed the first direct measurement of the time-integrated flavor untagged charge asymmetry in semileptonic B-s(0) decays A(SL)(s,unt) by comparing the decay rate of B-s(0) -> mu(+) D-s(-) nu X, where D-s(-) -> phi pi(-) and phi -> K+K-, with the charge-conjugate (B) over bar (0)(s) decay rate. This sample was selected from 1: 3 fb(-1) of data collected by the D0 experiment in run II of the Fermilab Tevatron collider. We obtain A(SL)(s,unt) = [1.23 +/- 0.97(stat) +/- 0.17(syst)] x 10(-2). Assuming that Delta m(s)/(Gamma) over bar (s) >> 1, this result can be translated into a measurement of the CP-violating phase in B-s(0) mixing: Delta Gamma(s)/Delta m(s) tan phi(s) = [2.45 +/- 1.93(stat) +/- 0.35(syst)] x 10(-2).

  • Measurement of the top quark mass in the lepton plus jets channel using the ideogram method

    • A measurement of the top quark mass using events with one charged lepton, missing transverse energy, and jets in the final state, collected by the D0 detector from p (p) over bar collisions at root s=1.96 TeV at the Fermilab Tevatron collider, is presented. A constrained fit is used to fully reconstruct the kinematics of the events. For every event a top quark mass likelihood is calculated taking into account all possible jet assignments and the probability that an event is signal or background. Lifetime-based identification of b jets is employed to enhance the separation between t (t) over bar signal and background from other physics processes and to improve the assignment of the observed jets to the quarks in the t (1) over bar hypothesis. We extract a multiplicative jet energy scale (JES) factor in situ, greatly reducing the systematic effect related to the jet energy measurement. In a data sample with an integrated luminosity of 425 pb(-1), we observe 230 candidate events, with an estimated background of 123 events, and measure m(t)=173.7 +/- 4.4(stat+JES)(-2.0)(+2.1)(syst) GeV. This result represents the first application of the ideogram technique to the measurement of the top quark mass in lepton+jets events.

  • Measurement of the W boson helicity in top quark decays at D0

    • We present a measurement of the fraction f(+) of right-handed W bosons produced in top quark decays, based on a candidate sample of tt events in the l + jets and dilepton decay channels corresponding to an integrated luminosity of 370 pb(-1) collected by the D0 detector at the Fermilab Tevatron pp Collider at root s = 1.96 TeV. We reconstruct the decay angle theta* for each lepton. By comparing the cos theta* distribution from the data with that for the expected background and signal for various values of f(+) (where we assume that the fraction of longitudinally-polarized W bosons has the standard model value of 0.70), we find f(+) = 0.056 +/- 0.080 (stat) +/- 0.057 (syst) (f(+) < 0.23 at 95% C. L.), consistent with the standard model prediction of f(+) = 3.6 X 10(-4).

  • Model-independent measurement of the W-boson helicity in top-quark decays at D0

    • We present the first model-independent measurement of the helicity of W bosons produced in top quark decays, based on a 1 fb(-1) sample of candidate t (t) over bar events in the dilepton and lepton plus jets channels collected by the D0 detector at the Fermilab Tevatron p (p) over bar Collider. We reconstruct the angle theta(*) between the momenta of the down-type fermion and the top quark in the W boson rest frame for each top quark decay. A fit of the resulting cos theta(*) distribution finds that the fraction of longitudinal W bosons f(0)=0.425 +/- 0.166(stat)+/- 0.102(syst) and the fraction of right-handed W bosons f(+)=0.119 +/- 0.090(stat)+/- 0.053(syst), which is consistent at the 30% C.L. with the standard model.

    • We report results from a search for the decay B-s(0)->mu(+)mu(-) using 1.3 fb(-1) of p (p) over bar collisions at root s = 1.96 TeV collected by the D0 experiment at the Fermilab Tevatron Collider. We find two candidate events, consistent with the expected background of 1.24 +/- 0.99, and set an upper limit on the branching fraction of B(B-s(0)->mu(+)mu(-))< 1.2x10(-7) at the 95% C.L.

    • lWe report on a search for second generation leptoquarks (LQ(2)) which decay into a muon plus quark in (p) over barp collisions at a center-of-mass energy of root s = 1.96 TeV in the DO detector using an integrated luminosity of about 300 pb(-1). No evidence for a leptoquark signal is observed and an upper bound on the product of the cross section for single leptoquark production times branching fraction into a quark and a muon was determined for second generation scalar leptoquaiks as a function of the leptoquark mass. This result has been combined with a previously published DO search for leptoquark pair production to obtain leptoquark mass limits as a function of the leptoquark-muon-quark coupling, lambda. Assuming lambda = 1, lower limits on the mass of a second generation scalar leptoquark coupling to a u quark and a muon are m(LQ2) > 274 GeV and m(LQ2) > 226 GeV for beta = 1 and beta = 1/2, respectively.

  • Search for techniparticles in e plus jets events at D0

    • We search for the technicolor process p(p) over bar ->rho(T)/omega(T)-> W pi(T) in events containing one electron and two jets, in data corresponding to an integrated luminosity of 390 pb(-1), recorded by the D0 experiment at the Fermilab Tevatron. Technicolor predicts that technipions pi(T) decay dominantly into b(b) over bar, b(c) over bar, or (b) over barc, depending on their charge. In these events b and c quarks are identified by their secondary decay vertices within jets. Two analysis methods based on topological variables are presented. Since no excess above the standard model prediction was found, the result is presented as an exclusion in the pi(T) vs rho(T) mass plane for a given set of model parameters.

    • We report a study of the decay B-s(0)->(DsDs(*))-D-(*) using a data sample corresponding to 1.3 fb(-1) of integrated luminosity collected by the D0 experiment in 2002-2006 during run II of the Fermilab Tevatron collider. One D-s((*)) meson was partially reconstructed in the decay D-s ->phi mu nu, and the other D-s((*)) meson was identified using the decay D-s ->phi pi where no attempt was made to distinguish D-s and D-s(*) states. For the branching fraction Br(B-s(0)->(DsDs(*))-D-(*)) we obtain a 90% C.L. range [0.002,0.080] and central value 0.039(-0.017)(+0.019)(stat)(-0.015)(+0.016)(syst). This was subsequently used to make the most precise estimate of the width difference Delta Gamma(CP)(s) in the B-s(0)-(B)over bar(s)(0) system: Delta Gamma(CP)(s)/Gamma(s)=0.079(-0.035)(+0.038)(stat)(-0.030)(+0.031)(syst).

  • 10 Years of research on debugging concurrent and multicore software : a systematic mapping study

    • Debugging – the process of identifying, localizing and fixing bugs – is a key activity in software development. Due to issues such as non-determinism and difficulties of reproducing failures, debugging concurrent software is significantly more challenging than debugging sequential software. A number of methods, models and tools for debugging concurrent and multicore software have been proposed, but the body of work partially lacks a common terminology and a more recent view of the problems to solve. This suggests the need for a classification, and an up-to-date comprehensive overview of the area. This paper presents the results of a systematic mapping study in the field of debugging of concurrent and multicore software in the last decade (2005– 2014). The study is guided by two objectives: (1) to summarize the recent publication trends and (2) to clarify current research gaps in the field.Through a multi-stage selection process, we identified 145 relevant papers. Based on these, we summarize the publication trend in the field by showing distribution of publications with respect to year , publication venues , representation of academia and industry , and active research institutes . We also identify research gaps in the field based on attributes such as types of concurrency bugs, types of debugging processes , types of research  and research contributions.The main observations from the study are that during the years 2005–2014: (1) there is no focal conference or venue to publish papers in this area, hence a large variety of conferences and journal venues (90) are used to publish relevant papers in this area; (2) in terms of publication contribution, academia was more active in this area than industry; (3) most publications in the field address the data race bug; (4) bug identification is the most common stage of debugging addressed by articles in the period; (5) there are six types of research approaches found, with solution proposals being the most common one; and (6) the published papers essentially focus on four different types of contributions, with ”methods” being the type most common one.We can further conclude that there is still quite a number of aspects that are not sufficiently covered in the field, most notably including (1) exploring correction  and fixing bugs  in terms of debugging process; (2) order violation, suspension  and starvation  in terms of concurrency bugs; (3) validation and evaluation research  in the matter of research type; (4) metric  in terms of research contribution. It is clear that the concurrent, parallel and multicore software community needs broader studies in debugging.This systematic mapping study can help direct such efforts.

    • With the development of multicore hardware, concurrent, parallel and multicore software are becoming increasingly popular. Software companies are spending a huge amount of time and resources to nd and debug the bugs. Among all types of software bugs, concurrency bugs are also important and troublesome. This type of bugs is increasingly becoming an issue particularly due to the growing prevalence of multicore hardware. In this position paper, we propose a model for monitoring and debugging Starvation bugs as a type of concurrency bugs in multicore software. The model is composed into three phases: monitoring, detecting and debugging. The monitoring phase can support detecting phase by storing collected data from the system execution. The detecting phase can support debugging phase by comparing the stored data with starvation bug's properties, and the debugging phase can help in reproducing and removing the Starvation bug from multicore software. Our intention is that our model is the basis for developing tool(s) to enable solving Starvation bugs in software for multicore platforms.

    • This article presents a runtime verification tool for embedded software executing under the open source real-time operating system FreeRTOS. The tool detects and diagnoses concurrency bugs such as deadlock, starvation, and suspension based-locking. The tool finds concurrency bugs at runtime without debugging and tracing the source code. The tool uses the Tracealyzer tool for logging relevant events. Analysing the logs, our tool can detect the concurrency bugs by applying algorithms for diagnosing each concurrency bug type individually. In this paper, we present the implementation of the tool, as well as its functional architecture, together with illustration of its use. The tool can be used during program testing to gain interesting information about embedded software executions. We present initial results of running the tool on some classical bug examples running on an AVR 32-bit board SAM4S.

Similarly, to design effective tools and techniques for testing and debugging concurrent software understanding the differences between non-concurrency and concurrency bugs in real-word software would be useful.

    • Cyber Physical Systems (CPS) bridge the cyber-world of computing and communications with the physical world and require development of secure and reliable software. It asserts a big challenge not only on testing and verifying the correctness of all physical and cyber components of such big systems, but also on integration of these components. This paper develops a categorization of multiple levels of testing required to test CPS and makes a comparison of these levels with the levels of software testing based on the V-model. It presents a detailed state-of-the-art survey on the testing approaches performed on the CPS. Further, it provides challenges in CPS testing.

  • Concurrency Bugs : Characterization, Debugging and Runtime Verification

    • Concurrent software has been increasingly adopted in recent years, mainly due to the introduction of multicore platforms. However, concurrency bugs are still difficult to test and debug due to their complex interactions involving multiple threads (or tasks). Typically, real world concurrent software has huge state spaces. Thus, testing techniques and handling of concurrency bugs need to focus on exposing the bugs in this large space. However, existing solutions typically do not provide debugging information to developers (and testers) for understanding the bugs.Our work focuses on improving concurrent software reliability via three contributions: 1) An investigation of concurrent software challenges with the aim to help developers (and testers) to better understand concurrency bugs. We propose a classification of concurrency bugs and discuss observable properties of each type of bug. In addition, we identify a number of gaps in the body of knowledge on concurrent software bugs and their debugging. 2) Exploring concurrency related bugs in real-world software with respect to the reproducibility of bugs, severity of their consequence and effort required to fix them. Our findings here is that concurrency bugs are different from other bugs in terms of their fixing time and severity, while they are similar in terms of reproducibility. 3) A model for monitoring concurrency bugs and the implementation and evaluation of a related runtime verification tool to detect the bugs. In general, runtime verification techniques are used to (a) dynamically verify that the observed behaviour matches specified properties and (b) explicitly recognize understandable behaviors in the considered software. Our implemented tool is used to detect concurrency bugs in embedded software and is in its current form tailored for the FreeRTOS operating system. It helps developers and testers to automatically identify concurrency bugs and subsequently helps to reduce their finding and fixing time.

  • Concurrency bugs in open source software : a case study

    • Concurrent programming puts demands on software debugging and testing, as concurrent software may exhibit problems not present in sequential software, e.g., deadlocks and race conditions. In aiming to increase efficiency and effectiveness of debugging and bug-fixing for concurrent software, a deep understanding of concurrency bugs, their frequency and fixing-times would be helpful. Similarly, to design effective tools and techniques for testing and debugging concurrent software, understanding the differences between non-concurrency and concurrency bugs in real-word software would be useful. This paper presents an empirical study focusing on understanding the differences and similarities between concurrency bugs and other bugs, as well as the differences among various concurrency bug types in terms of their severity and their fixing time, and reproducibility. Our basis is a comprehensive analysis of bug reports covering several generations of five open source software projects. The analysis involves a total of 11860 bug reports from the last decade, including 351 reports related to concurrency bugs. We found that concurrency bugs are different from other bugs in terms of their fixing time and severity while they are similar in terms of reproducibility. Our findings shed light on concurrency bugs and could thereby influence future design and development of concurrent software, their debugging and testing, as well as related tools.

    • Multicore hardware development increases the popularity of parallel and multicore software, while testing and debugging the software become more difficult, frustrating and costly. Among all types of software bugs, concurrency bugs are both important and troublesome. This type of bugs is increasingly becoming an issue, particularly due to the growing prevalence of multicore hardware. Suspension-based-locking bug is one type of concurrency bugs. This position paper proposes a model based on runtime verification and reflection technique in the context of multicore and parallel software to monitor and detect suspension-based-locking bugs. The model is not only able to detect faults, but also diagnose and even repair them. The model is composed of four layers: Logging, Monitoring, Suspension Bug Diagnosis and Mitigation. The logging layer will observe the events and save them into a file system. The monitoring layer will detect the presents of bugs in the software. The suspension bug diagnosis will identify Suspension bugs by comparing the captured data with the suspension bug properties. Finally, the mitigation layer will reconfigure the software to mitigate the suspension bugs. A functional architecture of a runtime verification tool is also proposed in this paper. This architecture is based on the proposed model and is comprised of different modules.

    • In software engineering, classification is a way to find an organized structure of knowledge about objects. Classification serves to investigate the relationship between the items to be classified, and can be used to identify the current gaps in the field. In many cases users are able to order and relate objects by fitting them in a category. This paper presents initial work on a taxonomy for classification of errors (bugs) related to concurrent execution of application level software threads. By classifying concurrency bugs based on their corresponding observable properties, this research aims to examine and structure the state of the art in this field, as well as to provide practitioner support for testing and debugging of concurrent software. We also show how the proposed classification, and the different classes of bugs, relates to the state of the art in the field by providing a mapping of the classification to a number of recently published papers in the software engineering field.

    • Overexpression of HER2 in breast carcinomas predicts response to trastuzumab therapy. Affibody molecules based on a non-immunoglobulin scaffold have demon-strated high potential for in vivo molecular imaging of HER2-expressing tumors. Re-engineering of the molecular scaffold has led to a second generation of optimized Affibody molecules, having a surface distinctly different from the parental protein domain from staphylococcal protein A. The new tracer showed further increased melting point, stability and overall hydrophilicity compared to the parental molecule, and was shown to be more amenable for chemical peptide synthesis. The goal of this study was to assess potential effects of this extensive re-engineering on HER2 targeting, using ABY-025, a DOTA conjugated variant of the novel tracer. Methods: 111In-ABY-025 was compared with previously evaluated parent HER2-binding Affibody tracers in vitro and in vivo. The in vivo behavior was further evaluated in mice bearing SKOV-3 xenografts, in rats and in cynomolgus macaques. Results: 111In-ABY-025 bound specifically to HER2 in vitro and in vivo. Direct comparison with the previous generation of HER2-binding tracers showed that ABY-025 retained excellent targeting properties. Rapid blood clearance was shown in mice, rats and macaques. A highly specific tumor uptake of 16.7 ± 2.5 %IA/g was seen at 4 h after injection. The tumor-to-blood ratio was 6.3 at 0.5 h, 88 at 4 h, and increased up to 3 days after injection. Gamma camera imaging of tumors was already possible 0.5 h after injection. Furthermore, repeated i.v. administration of ABY-025 did not induce antibody formation in rats. Conclusions: The biodistribution of 111In-ABY-025 was in remarkably good agreement with the parent tracers, despite profound re-engineering of the non-binding surface. The molecule displayed rapid blood clearance in all species investigated and excellent targeting capacity in tumor bearing mice, leading to high tumor-to-organ-ratios and high contrast imaging shortly after injection.

    • The Autism – Tics, AD/HD, and other Comorbidities inventory (A-TAC) is a comprehensive interview for evaluating problems related to autism spectrum disorders (ASD), tic disorders, attention-deficit/hyperactivity disorder (AD/HD), and common comorbid conditions in children and adolescents. A-TAC telephone interviews were administered to parents of 2,957 children aged nine- or twelve-years, representing one in each twin pair included in the population- based Child and Adolescent Twin Study in Sweden (CATSS). A total of 16.4% were screen-positive for one or several of the targeted disorder, 1.3% for ASD and 5.6% for AD/HD. All types of problems were more common among boys, with the exception of those related to “eating habits”. They were all dimensionally/continuously distributed, highly inter-correlated, and overlapped across types. They aggregated in three ba- sic factors corresponding to externalizing/disruptiveness, socio-communicative problems, and compulsiveness. Population-based data on problems in children thus challenge current categorical diagnostic definitions, calling for dimen- sional and complementary models of problem descriptions.

    • BACKGROUND: Lung transplantation (LTx) is established as a life-saving treatment in end-stage lung disease. However, long-term survival is hampered by the development of chronic rejection, almost synonymous with bronchiolitis obliterans syndrome (BOS). The rejection is characterized by deposition of extracellular matrix in small airways. Fibroblasts/myofibroblasts are the main producers of extracellular matrix molecules such as proteoglycans. This study compared fibroblast phenotype and activity in the wound healing process at different points after LTx in patients who later did, or did not, develop BOS. METHODS: Distally derived fibroblasts from patients 6 and 12 months after LTx and from healthy controls were analyzed for production of the proteoglycans versican, perlecan, biglycan, and decorin, with and without transforming growth factor (TGF)-β(1). Fibroblast migration and proliferation were also studied. RESULTS: At 6 and 12 months after LTx, versican production was higher in fibroblasts from LTx patients (p < 0.01 p < 0.01) than from controls. Fibroblasts from patients who later developed BOS were more responsive to TGF-β(1)-induced synthesis of versican and biglycan than patients without signs of rejection (p < 0.05). Production of perlecan and decorin was negatively correlated with fibroblast proliferation in fibroblasts at 6 months after LTx. In a more detailed case study of 2 patients, one with and one without BOS, the altered proteoglycan profile was associated with impaired lung function. CONCLUSIONS: LTx changes the phenotype of fibroblasts to a non-proliferative but extracellular matrix-producing cell due to wound healing involving TGF-β(1). If not controlled, this may lead to development of BOS.

    • Mg-chelatase catalyzes the insertion of Mg2+ into protoporphyrin IX at the first committed step of the chlorophyll biosynthetic pathway. It consists of three subunits: I, D, and H. The I subunit belongs to the AAA protein superfamily ( ATPases associated with various cellular activities) that is known to form hexameric ring structures in an ATP-dependant fashion. Dominant mutations in the I subunit revealed that it functions in a cooperative manner. We demonstrated that the D subunit forms ATP-independent oligomeric structures and should also be classified as an AAA protein. Furthermore, we addressed the question of cooperativity of the D subunit with barley (Hordeum vulgare) mutant analyses. The recessive behavior in vivo was explained by the absence of mutant proteins in the barley cell. Analogous mutations in Rhodobacter capsulatus and the resulting D proteins were studied in vitro. Mixtures of wild-type and mutant R. capsulatus D subunits showed a lower activity compared with wild-type subunits alone. Thus, the mutant D subunits displayed dominant behavior in vitro, revealing cooperativity between the D subunits in the oligomeric state. We propose a model where the D oligomer forms a platform for the stepwise assembly of the I subunits. The cooperative behavior suggests that the D oligomer takes an active part in the conformational dynamics between the subunits of the enzyme.

    • Background: Neuroinflammation is implicated in the pathophysiology of Parkinson's disease (PD) and related conditions, yet prior clinical biomarker data report mixed findings. Objectives: The aim was to measure a panel of neuroinflammatory acute phase response (APR) proteins in the cerebrospinal fluid (CSF) of participants with PD and related disorders. Methods: Eleven APR proteins were measured in the CSF of 867 participants from the BioFINDER cohort who were healthy (612) or had a diagnosis of PD (155), multiple system atrophy (MSA) (26), progressive supranuclear palsy (PSP) (22), dementia with Lewy bodies (DLB) (23), or Parkinson’s disease with dementia (PDD) (29). Results: CSF APR proteins were mostly unchanged in PD, with only haptoglobin and α1-antitrypsin significantly elevated compared to controls. These proteins were variably increased in the other disorders. Certain protein components yielded unique signatures according to diagnosis: ferritin and transthyretin were selectively elevated in MSA and discriminated these patients from all others. Haptoglobin was selectively increased in PSP, discriminating this disease from MSA when used in combination with ferritin and transthyretin. This panel of proteins did not correlate well with severity of motor impairment in any disease category, but several (particularly ceruloplasmin and ferritin) were associated with memory performance (Mini-Mental State Examination) in patients with DLB and PDD. Conclusions: These findings provide new insights into inflammatory changes in PD and related disorders while also introducing biomarkers of potential clinical diagnostic utility.

In this paper we focus on ethical issues that arise when human beings are directly affected in the performance of research, by comparing traffic research to the ethical requirements imposed on biomedical research. After introducing the basic ethical requirements on biomedical research, each of the major requirements is discussed in relation to traffic research. We identify the main areas where biomedical research and traffic research differ, and where the ethical requirements from the former cannot easily be transferred to the latter. Finally, we argue that there is a need for systematic studies of the ethics of traffic research and point to some of the issues that need to be addressed (A).     • Essay 1: Traffic research shares a fundamental dilemma with other areas of empirical research in which humans are potentially put at risk. Research is justified because it can improve safety in the long run. Nevertheless, people can be harmed in the research situation. Hence, we need to balance short-term risks against long-term safety improvements, much as in other areas of research with human subjects. In this paper we focus on ethical issues that arise when human beings are directly affected in the performance of research by examining how the ethical requirements in biomedical research can inform traffic research. After introducing the basic ethical requirements on biomedical research, each of the major requirements is discussed in relation to traffic research. We identify the main areas where biomedical research and traffic research differ, and where the ethical requirements from the former cannot easily be transferred to the latter. We then point to some of the issues that need to be addressed for a systematic approach to the ethics of traffic research.Essay 2: The requirement of always obtaining participants’ informed consent in research with human subjects cannot always be met, for a variety of reasons. In this paper, research situations where informed consent is unobtainable are described and categorised. Some of these kinds of situations, common in biomedicine and psychology, have been previously much discussed, whereas others, more prevalent in for example infrastructure research, introduce new perspectives. The advancement of new technology may lead to an increase in research of these kinds. The paper also provides a review of methods intended to compensate for a lack of consent and their applicability and usefulness for the different categories of situations are discussed, thereby providing insights into one important aspect of relevance for the question of permitting research without informed consent: how well that which informed consent is meant to safeguard can be achieved by other means.Essay 3: This paper starts with the assumption that it is morally problematic when people in need are offered money in exchange for research participation if the amount offered is unfair. Such offers are called “coercive”, and the degree of coerciveness is said to be determined by the offer’s potential to cause exploitation and its irresistibility. Depending on what view we take on the possibility to compensate for the sacrifices made by research participants, a wish to avoid “coercive offers” leads to policy recommendations concerning payment for participation. For sacrifices considered compensable we ought to offer either no payment or payment at a level deemed fair, while for sacrifices deemed incompensable we always ought to offer no payment.Essay 4: It is commonly thought that transactions that are the result of voluntary gift-giving do not constitute exploitation. This paper argues that exploitation is indeed possible in such situations, by showing how gift-giving can fulfil the two commonly proposed criteria for exploitation, namely that in an interaction between two persons one receives disproportionally little and the other disproportionally much of the resulting benefits, and that this disproportion is caused by the latter making inappropriate use of a disadvantage of the former. A theoretical approach to what such inappropriate use would amount to in cases of gift-giving is lacking. The paper therefore aims at spelling out such an approach. The method of reflective equilibrium inspires this endeavour, which proceeds by testing intuitions about examples that embody a set of possible conditions. It is concluded that three of the conditions are necessary for exploitation of gift-giving, namely (1) the giver incurs a loss, (2) the recipient has aimed for the gift, and (3) the gift is undeserved.

    • Neuroinflammation is implicated in the development and maintenance of persistent pain states, but there are limited data linking cerebrospinal fluid (CSF) inflammatory mediators with neurophysiological pain processes in humans. In a prospective observational study, CSF inflammatory mediators were compared between patients with osteoarthritis (OA) who were undergoing total hip arthroplasty due to disabling pain symptoms (n = 52) and pain-free comparison controls (n = 30). In OA patients only, detailed clinical examination and quantitative sensory testing were completed. Cerebrospinal fluid samples were analyzed for 10 proinflammatory mediators using Meso Scale Discovery platform. Compared to controls, OA patients had higher CSF levels of interleukin 8 (IL-8) (P = 0.002), intercellular adhesion molecule 1 (P = 0.007), and vascular cell adhesion molecule 1 (P = 0.006). Osteoarthritis patients with central sensitization possibly indicated by arm pressure pain detection threshold <250 kPa showed significantly higher CSF levels of Fms-related tyrosine kinase 1 (Flt-1) (P = 0.044) and interferon gamma-induced protein 10 (IP-10) (P = 0.024), as compared to subjects with PPDT above that threshold. In patients reporting pain numerical rating scale score >/=3/10 during peripheral venous cannulation, Flt-1 was elevated (P = 0.025), and in patients with punctate stimulus wind-up ratio >/=2, CSF monocyte chemoattractant protein 1 was higher (P = 0.011). Multiple logistic regression models showed that increased Flt-1 was associated with central sensitization, assessed by remote-site PPDT and peripheral venous cannulation pain, and monocyte chemoattractant protein-1 with temporal summation in the area of maximum pain. Multiple proinflammatory mediators measured in CSF are associated with persistent hip OA-related pain. Pain phenotype may be influenced by specific CSF neuroinflammatory profiles.

Although pain is the most disabling symptom of osteoarthritis (OA), limited knowledge exists regarding the CNS mechanisms that amplify pain and drive sensitization processes in humans.Objectives:The main objective of this study was to evaluate associations between cerebrospinal fluid (CSF) monoamine metabolites, pain severity, and central sensitization in patients with OA undergoing total hip arthroplasty (THA).Methods:Patients with OA (n = 52) and pain-free controls (n = 30) provided CSF samples for measurement of serotonin (5-hydroxyindoleacetic acid [5-HIAA]), noradrenaline (3-methoxy-4-hydroxyphenylglycol [HMPG]), and dopamine (homovanillic acid [HVA]) monoamine metabolites. Patients with OA completed longitudinal evaluation of pain using clinical measures and quantitative sensory testing.Results:Patients with OA had higher HMPG levels when compared with controls (P = 0.036). Within patients with OA undergoing THA, higher 5-HIAA and HVA levels were consistently associated with higher preoperative pain severity. Higher concentrations of 5-HIAA and HVA were also associated with lower conditioned pain modulation levels, whereas higher HMPG levels were linked to more efficient conditioned pain modulation. Patients with higher levels of CSF HVA exhibited increased pressure pain sensitivity (arm pressure pain detection threshold < 250 kPa vs ≥ 250 kPa, P = 0.042). Higher preoperative levels of CSF 5-HIAA predicted poorer pain control 6 months postoperatively (brief pain inventory pain severity; adjusted β = 0.010, 95% CI 0.001-0.019).Conclusions:In OA patients with disabling pain, higher CSF levels of serotonin and dopamine metabolites are associated with increased pain severity and central sensitization. Increased noradrenergic activity may be associated with more efficient pain inhibitory capacity.

    • White matter hyperintensities (WMH), perivascular spaces (PVS) and lacunes are common MRI features of small vessel disease (SVD). However, no shared underlying pathological mechanism has been identified. We investigated whether SVD burden, in terms of WMH, PVS and lacune status, was related to changes in the cerebral arterial wall by applying global cerebral pulse wave velocity (gcPWV) measurements, a newly described marker of cerebral vascular stiffness. In a population-based cohort of 190 individuals, 66–85 years old, SVD features were estimated from T1-weighted and FLAIR images while gcPWV was estimated from 4D flow MRI data. Additionally, the gcPWV’s stability to variations in field-of-view was analyzed. The gcPWV was 10.82 (3.94) m/s and displayed a significant correlation to WMH and white matter PVS volume (r = 0.29, p < 0.001; r = 0.21, p = 0.004 respectively from nonparametric tests) that persisted after adjusting for age, blood pressure variables, body mass index, ApoB/A1 ratio, smoking as well as cerebral pulsatility index, a previously suggested early marker of SVD. The gcPWV displayed satisfactory stability to field-of-view variations. Our results suggest that SVD is accompanied by changes in the cerebral arterial wall that can be captured by considering the velocity of the pulse wave transmission through the cerebral arterial network.

    • We studied clinical and immunological outcome of Covid-19 in consecutive CLL patients from a well-defined area during month 1–13 of the pandemic. Sixty patients (median age 71 y, range 43–97) were identified. Median CIRS was eight (4–20). Patients had indolent CLL (n = 38), had completed (n = 12) or ongoing therapy (n = 10). Forty-six patients (77%) were hospitalized due to severe Covid-19 and 11 were admitted to ICU. Severe Covid-19 was equally distributed across subgroups irrespective of age, gender, BMI, CLL status except CIRS (p < 0.05). Fourteen patients (23%) died; age ≥75 y was the only significant risk factor (p < 0.05, multivariate analysis with limited power). Comparing month 1–6 vs 7–13 of the pandemic, deaths were numerically reduced from 32% to 18%, ICU admission from 37% to 15% whereas hospitalizations remained frequent (86% vs 71%). Seroconversion occurred in 33/40 patients (82%) and anti-SARS-CoV-2 antibodies were detectable at six and 12 months in 17/22 and 8/11 patients, respectively. Most (13/17) had neutralizing antibodies and 19/28 had antibodies in saliva. SARS-CoV-2-specific T-cells (ELISpot) were detected in 14/17 patients. Covid-19 continued to result in high admission even among consecutive and young early- stage CLL patients. A robust and durable B and/or T cell immunity was observed in most convalescents.

  • Going for gold : A spectroelectrochemical and catalytic study of gold materials

    • With the increase in demand for renewable energy, understanding chemical processes is essential for improving the design of catalysts in order to achieve better performance. This thesis summarises the experimental investigation of three types of catalytic gold materials: gold oxide formed from gold films, oxide-derived gold (OD-Au) produced from gold films, and gold nanoparticles supported on metal oxides. Different spectroscopic techniques were employed, such as operando sum frequency generation (SFG) and in situ and ex situ X-ray spectroscopies. These methods allowed the probing of the electronic and chemical states of gold after oxidising electrochemical treatments. The results indicate the presence of subsurface gold oxide remnants after formation of OD-Au, which may help explain its improved catalytic properties with respect to pure gold. In addition, a mathematical model to couple the early stages of gold oxide formation with the nonlinear optical response of gold during this process is presented. This model suggests that the growth proceeds from small oxide islands to 3D oxide growth, while SFG oxidation variation is due to the suppression of the free electron density by negatively-charged adsorbing oxygen atoms. Gold oxide was also studied with both in situ and operando X-ray spectroscopies, showing the importance of a continuous electrochemical treatment during measurements to avoid beam induced effects. Furthermore, gold nanoparticles supported on metal oxides (TiO2 and γ-Fe2O3) were investigated mainly with mass spectrometry. The results indicate two different reaction pathways for oxidation of CO to CO2 depending on the type of metal oxide support. These findings could be used to help design future gold-based catalysts.

    • Oxide-derived metals are produced by reducing an oxide precursor. These materials, including gold, have shown improved catalytic performance over many native metals. The origin of this improvement for gold is not yet understood. In this study, operando non-resonant sum frequency generation (SFG) and ex situ high-pressure X-ray photoelectron spectroscopy (HP-XPS) have been employed to investigate electrochemically-formed oxide-derived gold (OD-Au) from polycrystalline gold surfaces. A range of different oxidizing conditions were used to form OD-Au in acidic aqueous medium (H3PO4, pH = 1). Our electrochemical data after OD-Au is generated suggest that the surface is metallic gold, however SFG signal variations indicate the presence of subsurface gold oxide remnants between the metallic gold surface layer and bulk gold. The HP-XPS results suggest that this subsurface gold oxide could be in the form of Au2O3 or Au(OH)3. Furthermore, the SFG measurements show that with reducing electrochemical treatments the original gold metallic state can be restored, meaning the subsurface gold oxide is released. This work demonstrates that remnants of gold oxide persist beneath the topmost gold layer when the OD-Au is created, potentially facilitating the understanding of the improved catalytic properties of OD-Au.

This thesis summarizes the experimental investigation of redox reactions on polycrystalline gold surfaces by combining electrochemistry with operando surface-specific sum frequency generation (SFG) for a wider anodic potential interval than previously studied in acidic aqueous medium (H3PO4, pH 1). This configuration provides new insight about the chemical processes involved in the formation and reduction of gold oxides. More precisely, the migration of oxygen below the gold surface leading to a stratified structure has been suggested for the first time. This information is relevant to bring more clarity about redox reactions on gold surface during catalytic reactions.

    • To determine the incidence of physical inactivity and factors prior to stroke and in acute stroke that are associated with physical inactivity 1 year after stroke Design: Prospective longitudinal cohort Patients: A total of 190 consecutively included individuals with acute stroke Methods: A follow-up questionnaire, relating to physical activity level using the Saltin-Grimby Physical Activity Scale, was sent to participants in The Fall Study of Gothenburg 1 year after stroke. Predictors of physical inactivity at baseline were identified using univariable and multivariable logistic regression analyses.Physical inactivity 1 year after stroke was reported by 70 (37%) of the 190 patients who answered the questionnaire and was associated with physical inactivity before the stroke, odds ratio (OR) 4.07 (95% confidence interval (95% CI) 1.69-9.80, p=0.002); stroke severity (assessed by National Institutes of Health Stroke Scale (NIHSS), score 1-4), OR 2.65 (95% CI) 1.04-6.80, p=0.042) and fear of falling in acute stroke, OR 2.37 (95% CI 1.01-5.60, p=0.048).Almost 4 in 10 participants reported physical inactivity 1 year after stroke. Physical inactivity before the stroke, stroke severity and fear of falling in acute stroke are the 3 main factors that predict physical inactivity 1 year after stroke.
