  • Atrophy of the posterior subiculum is associated with memory impairment, Tau- and Aβ pathology in non-demented individuals

    • Alzheimer's disease (AD) is associated with atrophy of the cornu ammonis (CA) 1 and the subiculum subfield of the hippocampus (HC), and with deficits in episodic memory and spatial orientation. These deficits are mainly associated with the functionality of the posterior HC. We therefore hypothesized that key AD pathologies, i.e., β-amyloid and tau pathology would be particularly associated with the volume of the posterior subiculum in non-demented individuals. In our study we included 302 cognitively normal elderly participants (CN), 183 patients with subjective cognitive decline (SCD) and 171 patients with amnestic mild cognitive impairment (MCI), all of whom underwent 3T magnetic resonance images (MRI). The subicular subfield was segmented using Freesurfer 5.3 and divided into 10 volumetric segments moving from the most posterior (segment 1) to the most anterior part along the axis of the hippocampal head and body (segment 10). Cerebrospinal fluid (CSF) Aβ42 and phosphorylated tau (P-tau) were quantified using ELISA and were used as biomarkers for β-amyloid and tau pathology, respectively. In the total sample, tau-pathology and Aβ-pathology and (measured by elevated P-tau and low Aβ42 levels in CSF) and mild memory dysfunction were mostly associated with the volume changes of the posterior subiculum. Both SCD and MCI patients with elevated P-tau or low Aβ42 levels displayed predominantly posterior subicular atrophy in comparisons to control subjects with normal CSF biomarker levels. Finally, there was no main effect of Aβ42 or P-tau when comparing SCD with abnormal P-tau or Aβ42 with SCD with normal levels of these CSF-biomarkers. However, in the left subiculum there was a significant interaction revealing atrophy in the left posterior but not the anterior subiculum in participants with low Aβ42 levels. The same pattern was observed on the contralateral side in participants with elevated P-tau levels. In conclusion, AD pathologies and mild memory dysfunction are mainly associated with atrophy of the posterior parts of the subicular subfields of the HC in non-demented individuals. In light of these findings we suggest that segmentation of the HC subfields may benefit from considering the volume of the different anterior-posterior subsections of each subfield.

    • In vitro studies suggest that the G protein-coupled receptor (GPR) 30 is a functional estrogen receptor. However, the physiological role of GPR30 in vivo is unknown, and it remains to be determined whether GPR30 is an estrogen receptor also in vivo. To this end, we studied the effects of disrupting the GPR30 gene in female and male mice. Female GPR30((-/-)) mice had hyperglycemia and impaired glucose tolerance, reduced body growth, increased blood pressure, and reduced serum IGF-I levels. The reduced growth correlated with a proportional decrease in skeletal development. The elevated blood pressure was associated with an increased vascular resistance manifested as an increased media to lumen ratio of the resistance arteries. The hyperglycemia and impaired glucose tolerance in vivo were associated with decreased insulin expression and release in vivo and in vitro in isolated pancreatic islets. GPR30 is expressed in islets, and GPR30 deletion abolished estradiol-stimulated insulin release both in vivo in ovariectomized adult mice and in vitro in isolated islets. Our findings show that GPR30 is important for several metabolic functions in female mice, including estradiol-stimulated insulin release.

  • Diffusion Tensor Tractography versus Volumetric Imaging in the Diagnosis of Behavioral Variant Frontotemporal Dementia

    • MRI diffusion tensor imaging (DTI) studies of white matter integrity in behavioral variant frontotemporal dementia have consistently shown involvement of frontal and temporal white matter, corresponding to regional loss of cortical volume. Volumetric imaging has a suboptimal sensitivity as a diagnostic tool and thus we wanted to explore if DTI is a better method to discriminate patients and controls than volumetric imaging. We examined the anterior cingulum bundle in 14 patients with behavioral variant frontotemporal dementia and 22 healthy controls using deterministic manual diffusion tensor tractography, and compared DTI parameters with two measures of cortical atrophy, VBM and cortical thickness, of the anterior cingulate cortex (ACC). Statistically significant changes between patients and controls were detected in all DTI parameters, with large effect sizes. ROC-AUC was for the best DTI parameters: 0.92 (fractional anisotropy) to 0.97 (radial diffusivity), 0.82 for the best cortical parameter, VBM of the ACC. Results from the AUC were confirmed with binary logistic regression analysis including demographic variables, but only for fractional anisotropy and mean diffusivity. Ability to classify patient/nonpatient status was significantly better for mean diffusivity vs. VBM (p = 0.031), and borderline significant for fractional anisotropy vs. VBM (p = 0.062). The results indicate that DTI could offer advantages in comparison with the assessment of cortical volume in differentiating patients with behavioral variant frontotemporal dementia and controls.

    • Many extensions of the Standard Model predict the production of dark matter particles at the LHC. Sufficiently light dark matter particles may be produced in decays of the Higgs boson that would appear invisible to the detector. This Letter presents a statistical combination of searches for H→invisible decays where multiple production modes of the Standard Model Higgs boson are considered. These searches are performed with the ATLAS detector using 139 fb−1 of proton–proton collisions at a centre–of–mass energy of   at the LHC. In combination with the results at √s=7TeV and 8TeV, an upper limit on the H→invisible branching ratio of 0.107 (0.077) at the 95% confidence level is observed (expected). These results are also interpreted in the context of models where the 125 GeV Higgs boson acts as a portal to dark matter, and limits are set on the scattering cross-section of weakly interacting massive particles and nucleons.

    • Measurements of transverse energy-energy correlations and their associated azimuthal asymmetries in multijet events are presented. The analysis is performed using a data sample corresponding to 139 fb-1 of proton-proton collisions at a centre-of-mass energy of root s = 13TeV, collected with the ATLAS detector at the Large Hadron Collider. The measurements are presented in bins of the scalar sum of the transverse momenta of the two leading jets and unfolded to particle level. They are then compared to next-to-next-to-leading-order perturbative QCD calculations for the first time, which feature a significant reduction in the theoretical uncertainties estimated using variations of the renormalisation and factorisation scales. The agreement between data and theory is good, thus providing a precision test of QCD at large momentum transfers Q. The strong coupling constant alpha(s) is extracted as a function of Q, showing a good agreement with the renormalisation group equation and with previous analyses. A simultaneous fit to all transverse energy-energy correlation distributions across different kinematic regions yields a value of alpha(s)( mZ) = 0.1175 +/- 0.0006 (exp.)(+0.0034) (-0.0017) (theo.), while the global fit to the asymmetry distributions yields alpha(s)(m(Z)) = 0.1185 +/- 0.0009 (exp.)(+0.0025)(-0.0012)(theo.).

    • Measurements of differential cross sections are presented for inclusive isolated photon production in pp collisions at a centre-of-mass energy of 13TeV provided by the LHC and using 139 fb-1 of data recorded by the ATLAS experiment. The cross sections are measured as functions of the photon transverse energy in different regions of photon pseudorapidity. The photons are required to be isolated by means of a fixed-cone method with two different cone radii. The dependence of the inclusive-photon production on the photon isolation is investigated by measuring the fiducial cross sections as functions of the isolation-cone radius and the ratios of the differential cross sections with different radii in different regions of photon pseudorapidity. The results presented in this paper constitute an improvement with respect to those published by ATLAS earlier: the measurements are provided for different isolation radii and with a more granular segmentation in photon pseudorapidity that can be exploited in improving the determination of the proton parton distribution functions. These improvements provide a more in-depth test of the theoretical predictions. Next-to-leading-order QCD predictions from JETPHOX and SHERPA and next-to-next-to-leading-order QCD predictions from NNLOJET are compared to the measurements, using several parameterisations of the proton parton distribution functions. The measured cross sections are well described by the fixed-order QCD predictions within the experimental and theoretical uncertainties in most of the investigated phase-space region.

    • The production of a W boson in association with a single charm quark is studied using 140  fb−1 of √s=13  TeV proton-proton collision data collected with the ATLAS detector at the Large Hadron Collider. The charm quark is tagged by the presence of a charmed hadron reconstructed with a secondary-vertex fit. The W boson is reconstructed from the decay to either an electron or a muon and the missing transverse momentum present in the event. The charmed mesons reconstructed are D+→K−π+π+ and D*+→D0π+→(K−π+)π+ and the charge conjugate decays in the fiducial regions where pT(e,μ)>30  GeV, |η(e,μ)|<2.5, pT(D(*))>8  GeV, and |η(D(*))|<2.2. The integrated and normalized differential cross sections as a function of the pseudorapidity of the lepton from the W boson decay, and of the transverse momentum of the charmed hadron, are extracted from the data using a profile likelihood fit. The measured total fiducial cross sections are Results are compared with the predictions of next-to-leading-order quantum chromodynamics calculations performed using state-of-the-art parton distribution functions. Additionally, the ratio of charm to anticharm production cross sections is studied to probe the quark asymmetry. The ratio is found to be . The ratio and cross-section measurements are consistent with the predictions obtained with parton distribution function sets that have a symmetric sea, indicating that any asymmetry in the Bjorken-x region relevant for this measurement is small.

  • Observation of four-top-quark production in the multilepton final state with the ATLAS detector

    • This paper presents the observation of four-top-quark () production in proton-proton collisions at the LHC. The analysis is performed using an integrated luminosity of 140 fb−1 at a centre-of-mass energy of 13 TeV collected using the ATLAS detector. Events containing two leptons with the same electric charge or at least three leptons (electrons or muons) are selected. Event kinematics are used to separate signal from background through a multivariate discriminant, and dedicated control regions are used to constrain the dominant backgrounds. The observed (expected) significance of the measured signal with respect to the standard model (SM) background-only hypothesis is 6.1 (4.3) standard deviations. The production cross section is measured to be fb, consistent with the SM prediction of 12.0±2.4 fb within 1.8 standard deviations. Data are also used to set limits on the three-top-quark production cross section, being an irreducible background not measured previously, and to constrain the top-Higgs Yukawa coupling and effective field theory operator coefficients that affect production.

    • Means to measure the temporal evolution following a photo-excitation in conjugated polymers are a key for the understanding and optimization of their function in applications such as organic solar cells. In this paper we study the electronic structure dynamics by direct pump-probe measurements of the excited electrons in such materials. Specifically, we carried out a time-resolved photoelectron spectroscopy (TRPES) study of the polymer PCPDTBT by combining an extreme ultraviolet (XUV) high harmonic generation source with a time-of-flight spectrometer. After excitation to either the 1st excited state or to a higher excited state, we follow how the electronic structure develops and relaxes on the electron binding energy scale. Specifically, we follow a less than 50 fs relaxation of the higher exited state and a 10 times slower relaxation of the 1st excited state. We corroborate the results using DFT calculations. Our study demonstrates the power of TRPES for studying photo-excited electron energetics and dynamics of solar cell materials.

  • A deep learning model for brain age prediction using minimally preprocessed T1w images as input

    • Introduction: In the last few years, several models trying to calculate the biological brain age have been proposed based on structural magnetic resonance imaging scans (T1-weighted MRIs, T1w) using multivariate methods and machine learning. We developed and validated a convolutional neural network (CNN)-based biological brain age prediction model that uses one T1w MRI preprocessing step when applying the model to external datasets to simplify implementation and increase accessibility in research settings. Our model only requires rigid image registration to the MNI space, which is an advantage compared to previous methods that require more preprocessing steps, such as feature extraction. Methods: We used a multicohort dataset of cognitively healthy individuals (age range = 32.0–95.7 years) comprising 17,296 MRIs for training and evaluation. We compared our model using hold-out (CNN1) and cross-validation (CNN2–4) approaches. To verify generalisability, we used two external datasets with different populations and MRI scan characteristics to evaluate the model. To demonstrate its usability, we included the external dataset’s images in the cross-validation training (CNN3). To ensure that our model used only the brain signal on the image, we also predicted brain age using skull-stripped images (CNN4). Results: The trained models achieved a mean absolute error of 2.99, 2.67, 2.67, and 3.08 years for CNN1–4, respectively. The model’s performance in the external dataset was in the typical range of mean absolute error (MAE) found in the literature for testing sets. Adding the external dataset to the training set (CNN3), overall, MAE is unaffected, but individual cohort MAE improves (5.63–2.25 years). Salience maps of predictions reveal that periventricular, temporal, and insular regions are the most important for age prediction. Discussion: We provide indicators for using biological (predicted) brain age as a metric for age correction in neuroimaging studies as an alternative to the traditional chronological age. In conclusion, using different approaches, our CNN-based model showed good performance using one T1w brain MRI preprocessing step. The proposed CNN model is made publicly available for the research community to be easily implemented and used to study ageing and age-related disorders.

    • A sophisticated design of 750 LaBr3(Ce):LaCl3(Ce) phoswich crystals (CEPA10) with a segmentation determined by the Doppler correction and an energy resolution of 5% at 1 MeV is presented. Monte Carlo simulations have been performed for high energy protons (50–500 MeV) and gamma radiation (0.5–30 MeV) to determine the length and shape of the crystals for optimum performance of the detector. In the case of protons, the two-layer detector can be used as a ΔELaBr3 − ETot telescope or, for very high energies, as a double energy loss detector (ΔELaBr3 + ΔELaCl3), in order to determine the initial energy. In addition, an experimental test with high energy protons (70–230 MeV) was performed at the cyclotron center in Krakow, Poland with a first prototype of 2 x 2 phoswich rectangular crystals (CEPA4) packed in an aluminum can (0.5 mm case). To simulate CEPA10 efficiencies and resolutions, optical pulses detected in CEPA4 by photomultiplier tubes with a DAQ system were used as energy input functions in Monte Carlo simulations.

    • High kinetic-energy photoelectron spectroscopy (HIKE) or hard x-ray photoelectron spectroscopy has been used to investigate the alloying of Ni/Cu (100) multilayers. Relative intensities of the corelevels and their chemical shifts derived from binding energy changes are shown to give precise information on physicochemical properties and quality of the buried layers. Interface roughening, including kinetic properties such as the rate of alloying, and temperature effects on the processes can be analyzed quantitatively. Using HIKE, we have been able to precisely follow the deterioration of the multilayer structure at the atomic scale and observe the diffusion of the capping layer into the multilayer structure which in turn is found to lead to a segregation in the ternary system. This is of great importance for future research on multilayered systems of this kind. Our experimental data are supplemented by first-principles theoretical calculations of the core-level shifts for a ternary alloy to allow for modeling of the influence of capping materials on the chemical shifts.

    • Pediococcus parvulus 2.6 (previously Pediococcus damnosus 2.6, here confirmed as P. parvulus by 16S DNA sequencing) displayed antibacterial activity toward several bacterial species, including isolates found as contaminants in oats, herein genetically identified as Bacillus cereus. No inhibition of Listeria monocytogenes was found under the conditions used. Antibacterial activity was retrieved after ammonium sulfate or acetone precipitation showed it to be peptide mediated. P. parvulus 2.6 has previously shown good technological properties in oat-based products. This, together with the currently found inhibition of food spoilage microorganisms like B. cereus, makes it suitable as a food protective culture. Survival trials of P. parvulus 2.6 at conditions mimicking the gastrointestinal tract were prompted by previously found cholesterol-lowering effects in humans after consumption of oat products cofermented by using P. parvulus 2.6 and Bifidobacterium spp. Viability was measured with in vitro, gutlike simulations at 37 degrees C. High survival was shown under two of three conditions (gastric juice, bile, and small intestine juice), defined as main obstacles of the gastrointestinal tract. The critical step was bile exposure. At a concentration of 20%, viability was low, but 0.3% bile (mean concentration in the intestine) did not have a major influence on growth. Viability of P. parvulus 2.6 was significantly decreased in gastric juice at pH 1.5 (with pepsin), but it was not significantly affected at pH 2.5, and was also improved at a lower pH in 20% oat milk. Viability was judged sufficient for colonization at gutlike conditions, qualifying the strain for further probiotic studies.

  • Health problems and work-related stress in Swedish ambulance personnel

    • Background: Previous studies have shown a high incidence of both acute and post-traumatic stress among ambulance personnel. It has been shown that ambulance personnel are at a higher risk of being affected of heart disease, high blood pressure and cancer. Studies have also seen a higher incidence of substance abuse and suicide. One cause of these health problems can be work-related stress. No previous research has focused on the body's physical reactions in the form of changes in heart rate and stress hormones in ambulance personnel in connection with work-related stress. Nor is there any overall picture of what actually affects Swedish ambulance personnel in terms of reported morbidity.Aim: The overall aim of the thesis is to investigate health problems in Swedish ambulance personnel and to study if there are any factors related to the work environment and the special conditions occurring in the prehospital environment that can be linked to the findings that appear.The questions that should be answered are: Are there health problems that affect Swedish ambulance personnel to a higher extent than other professions in Sweden? Are there any factors relating to morbidity that can be linked to the profession and can be regarded as potentially dangerous? If so, are there methods to prevent health problems that can be implemented in daily work?Methods: Studies I, II and III were all studies where stress markers (heart rate and cortisol levels) were measured during different conditions linked to the profession. Study I was a study where this was measured during physical exertion e.g. to carry a stretcher. Study II was a validated theoretical stress test to see how the ambulance personnel reacted to unknown factors. Study III meant measuring stress markers during priority-1 alarms. Study IV was carried out as a longitudinal register study where data about ICD-codes was collected from Statistics Sweden and the National Board of Health and Welfare.Results: In study I it was shown that the use of lifting aids reduced the measurable stress in the form of both reduced heart rate and decreased cortisol levels. Study II showed that personnel were stressed of the unknown test though women had the highest salivary cortisol levels before the Trier social stress test while the highest value for men occurred 10 to 20 minutes after the test. Study III showed that there was an increase in heart rate during priority-1 alarms that could not be linked to physical activity. It also indicated/showed that traffic accidents, patients with heart attacks in need of acute PCI or thrombectomy and alarms regarding children generate the highest stress onset seen as prolonged elevation of cortisol levels. This was seen regardless of gender, age, education or experience in all three studies. Study IV showed that Swedish ambulance personnel run the risk of being affected by certain diseases such as cardiac arrhythmias and injuries as arthrosis of the knee, dorsopathies and intervertebral disc disorders to a greater extent compared to other health care workers and other professions in Sweden.Conclusion: Swedish ambulance personnel have a higher incidence of certain health problems and diseases such as paroxysmal tachycardia, atrial fibrillation and flutter, other cardiac arrhythmias, high blood pressure, of the knee and dorsopathies and intervertebral disc disorders compared to other professions. These conditions can be caused by work-related stress, although they are not the only cause. Some factors that cause a stress reaction that can be linked to the ambulance profession have emerged in the studies. But the extent to which these factors alone cause the health problems that Swedish ambulance personnel suffer from is more difficult to determine with certainty based on these studies. Using shoulder straps reduces both heart rate and cortisol secretion.Clinical implications: Greater use of aids both in terms of lifting, moving and carrying heavy loads should mean that the physical load on the body would be reduced. These aids could also prevent some musculoskeletal problems that have emerged in this thesis by relieving and distributing the burden throughout the body. An important aspect in preventing work-related stress is that sufficient time is given for recovery. The organizations need to be dimensioned so that there is sufficient standby time so that there is time for recovery and reflection but also time for education and training. This is something that is far from reality in many Swedish ambulance organizations. It is also time to seriously discuss what is an actual reality in many countries, namely that employees in the ambulance service can benefit from a lower retirement age after a certain number of years of service.

    • This paper outlines a general 2-loop model of action research in order to account for two important facets of action research that traditional cycle models do not explicitly account for. One facet concerns the interplay between the processes of action research (ie diagnosing, action planning, acting, evaluating, and learning) on one hand, and their conceptual outcomes (ie the perceived real world situation, the theoretical understanding, and the observed organizational response) on the other. The second facet concerns the dynamic exchange of knowledge between the world theory and the world of practice. Both types of knowledge are necessary, but none of them are sufficient to shape the result of an action research project. We argue that the dialectic between these two worlds manifests itself in a certain point which we call “the moment of truth”, where the theoretical understanding is challenged by practical facts, and where practice is scrutinized by theoretical knowledge. We believe that by acknowledging these facets, it is possible to enhance the quality of action research.

    • Purpose: The purpose of this paper is to contribute to the advancement of research methods in management spirituality by proposing a novel and innovative approach for generating empirical material on issues relevant for practitioners. Design/methodology/approach: The approach introduced is designed as a one-year group process based on Schein’s concept of clinical inquiry, primarily aimed at helping the participating managers to address challenging issues that they face in real life. When this helping process is successful, empirical material on these issues is revealed, as the participants share information to receive help themselves. Findings: Findings illustrate how the approach generates extensive, rich and broad data, guided by what becomes important for the participating managers as the process unfolds. A strong holding environment allows for urgent matters to be shared straight from the heart. Research limitations/implications: As this activity is based on the belief that the managers know best the issues relevant to them, a deviation from the assumed research direction should be seen as help to focus on relevant issues, rather than an obstacle of following an original plan. Practical implications: The proposed approach may be of particular interest to researchers who not only want to contribute to academic knowledge but also wish to work with people and facilitate transformation. Originality/value: This paper advances understandings of generating empirical material in the field of management spirituality through an innovative, client-centered research approach, rooted in well-established theories. © 2017, © Emerald Publishing Limited.

Upper limits are set at 95% confidence level on the production cross section times branching fractions for quantum black holes decaying into a lepton and a quark in a search region with invariant mass above 2.0 TeV.     • Quantifying the degree of atrophy is done clinically by neuroradiologists following established visual rating scales. For these assessments to be reliable the rater requires substantial training and experience, and even then the rating agreement between two radiologists is not perfect. We have developed a model we call AVRA (Automatic Visual Ratings of Atrophy) based on machine learning methods and trained on 2350 visual ratings made by an experienced neuroradiologist. It provides fast and automatic ratings for Scheltens' scale of medial temporal atrophy (MTA), the frontal subscale of Pasquier's Global Cortical Atrophy (GCA-F) scale, and Koedam's scale of Posterior Atrophy (PA). We demonstrate substantial inter-rater agreement between AVRA's and a neuroradiologist ratings with Cohen's weighted kappa values of kappa(w) = 0.74/0.72 (MTA left/right), kappa(w) = 0.62 (GCA-F) and kappa(w) = 0.74 (PA). We conclude that automatic visual ratings of atrophy can potentially have great scientific value, and aim to present AVRA as a freely available toolbox.

  • High-Dose Radioimmunotherapy Combined With Extracorporeal Depletion in a Syngeneic Rat Tumor Model

    • BACKGROUND: The aim of the current study was to investigate the possibility of increasing the maximal tolerated dose (MTD) of a tumor-selective radiolabeled antibody when radioimmunotherapy (RIT) is combined with extracorporeal depletion of radioimmunoconjugates from the circulation. Furthermore, the authors evaluated whether this increase in dose improved the therapeutic effect on solid manifest tumors in an immunocompetent animal model. METHODS: Rats were injected with high activities/body weight of lutetium (Lu-177)- or yttrium (Y-90)-labeled antibody conjugates (monoclonal antibody tetraazacyclododecanetetraacetic acid-biotin) and subjected to removal of the conjugate from the circulation by extracorporeal affinity adsorption treatment 24 hours postinjection. Myelotoxicity was assessed by analysis of blood parameters for 12 weeks. The effect of increased doses in combination with extracorporeal affinity adsorption treatment was evaluated with respect to myelotoxicity and therapeutic effect in a syngeneic rat colon cancer model. RESULTS: The MTD of Lu-177- or Y-90-labeled immunoconjugates could be increased 2.0x or 1.5x, respectively, when RIT was combined with extracorporeal affinity adsorption treatment. All animals treated with Lu-177- or Y-90-labeled antibodies showed persistent complete response of manifest tumors (approximately 10 x 15 mm) within 16 days postinjection. However, several animals showed disseminated disease 1.5 to 3 months postinjection. CONCLUSIONS: Extracorporeal affinity adsorption treatment is a method that safely and efficiently reduces myelotoxicity associated with RIT. Extracorporeal affinity adsorption treatment allows increased administered activity without increased toxicity, with the aim of increasing the absorbed dose to the tumor. However, because tumor/normal tissue radiosensitivity ratios are more favorable in rodents, it is not possible to draw any conclusions concerning the therapeutic efficacy of increased administered activity in combination with extracorporeal affinity adsorption treatment in this study. Targeted RIT with beta-emitting radionuclides seems not to be effective in microscopic disease, because metastases developed at sites without previously known disease.     • Purpose: Evaluation of the possibilities of reducing the accumulation of radiolabeled streptavidin in radiosensitive organs by extracorporeal affinity adsorption (ECAT). Experimental Design: Rats were injected with biotinylated antibody and subjected to removal of the antibodies from the circulation by ECAT 24 h after injection (avidin column). Animals were then injected with In-111-1,4,7,10-tetra-azacylododecane N,N',N '',N'''-tetraacetic acid (DOTA) streptavidin. In a third step, animals were subjected to a second ECAT 8 h after injection to remove the DOTA-streptavidin from the circulation (biotin column). Biodistribution and tumor targeting of DOTA-streptavidin 24 h after injection was determined. Results: Elimination of biotinylated antibody by ECAT before injection of DOTA-streptavidin increased the tumor targeting by 50%. In addition, the levels of DOTA-streptavidin in liver and lymph nodes were reduced by 60%, which implied a 4.3- and 3.8-fold increase of tumor-to-liver and tumor-to-lymph node ratios, respectively. By doing a second ECAT to remove DOTA-streptavidin from the circulation, accumulation in normal tissues was reduced. However, this latter ECAT also reduced tumor accumulation by 25% (mostly corresponding to radioactivity in the circulation). Conclusions: ECAT was efficient as a means of removing biotinylated antibodies and would probably also be efficient for the clearance of streptavidin-conjugated antibodies. Conversely, the use of ECAT for removal of radiolabeled streptavidin seems not to offer any advantage.

    • A fermented, oat-based product with properties resembling those of the traditional, Swedish, fermented milk product "filmjoelk" (sour milk) was developed. Com., mixed-strain, mesophilic starter cultures were grown in the oatbases M30, G40, and Oatly, which differed in the compn. of their carbohydrates. The exopolysaccharide-producer Pediococcus damnosus 2.6 was included to increase the viscosity of the G40 product. The CHN-19 culture chosen for the final product formulation produced a pleasant, buttery aroma, but a less sour taste than ordinary "filmjoelk". The addn. of glucose to M30 increased the acidification rate and gave a better aroma. A sensory anal. of two flavored products based on the G40 and M30 media, showed a higher acceptance of the M30 product. This study shows a potential for producing an oat-based "filmjoelk", but further work on consistency and taste is needed.

Three different exopolysaccharide (EPS) producing lactic acid bacteria strains were used to ferment the non-dairy oat-base (Adavena(R) G40) (Ceba Foods AB, Lund; Sweden). Two commercial non-dairy products based on oats (Mill Milk(TM)) (Ceba Foods AB, Lund, Sweden) and rice (Rice Dream(R)) (Imagine Foods, London, UK) were used as non-ropy and unfermented controls. All the standardized feeds were sterilized before being fed to the animals. Adult, germfree-and conventional AGUS rats, were fed the above sterile diets ad libitum for 21 days. Blood samples and faecal samples were collected and the animals' weight gain was monitored throughout the study. No significant change in serum lipids or faecal excretion of cholesterol was observed between the groups on the different diets. A difference in faecal SCFA pattern was observed in conventional rats fed on the oat-based diets in comparison. to the group fed on the rice-based diet. More evidence is needed to support the effect of fermented, ropy, oat-based products and their potential effect on serum lipids, faecal cholesterol/coprostanol levels and amounts of short chain fatty acids.

    • This investigation determined the effects of fermented oat-based products containing both native and microbial beta-glucans on plasma lipids and on fecal total bacterial count and Bifidobacterium ssp. The study was randomized, double blind with 3 parallel groups. Sixty-two free-living volunteers with moderately increased plasma cholesterol levels were recruited. In the final analysis, 56 subjects remained, as 6 subjects had left the study either due to lack of time (n = 2), unwillingness to continue the regimen (n = 2), or for other reasons (n = 2). During the first 3 weeks, all subjects received a fermented dairy-based product (control product, run-in period). On the following 5 weeks, I group continued with the control product, whereas the other 2 groups were given fermented oat-based products (intervention period, 3-3.5 g native beta-glucans per day). One of the oat products (ropy) was cofermented with an exopolysaccharide-producing strain, Pediococcus damnosus 2.6. A significant (P = .022) reduction in total cholesterol by 6% was observed in volunteers who had eaten the fermented, ropy, oat-based product compared with the control group. No other significant changes in plasma lipids were found. A significant increase in total bacterial count (P = .001) and Bifidobacterium ssp (P = .012) was observed in fecal samples from volunteers in the group who had eaten the fermented, ropy, oat-based product. This study shows that a fermented, ropy, oat-based product, containing both native and microbial glucans, can reduce the blood cholesterol level and also stimulate the bifidobacteria flora in the gastrointestinal tract.     • This thesis deals with the fermentation characteristics of lactic acid bacteria (LAB) in oat-based suspensions, with formulation work of fermented products based on oat and with nutritional studies of these products. Changes in structure in terms of viscosity and ropiness were studied when exopolysaccharide (EPS)-producing LAB strains, namely, Lactobacillus delbrueckii subsp. bulgaricus NCFB 2772, Lactobacillus brevis G-77 and Pediococcus damnosus 2.6 were grown in these oat-based suspensions. Differences in structure were seen after growth of EPS-producing bacteria, resulting in higher viscosity and ropiness in these products. The formulation work of fermented products was carried out by using commercial yoghurt cultures with and without the presence of an EPS-producing strain. The products developed were found to have sensory acceptance and good bacterial survival during storage. The bacterial survival of three probiotic strains, Lactobacillus reuteri ATCC 55730, Lactobacillus acidophilus DSM 20079 and Bifidobacterium bifidum DSM 20456, was also studied in different oat-based suspensions with and without the influence of a yoghurt culture during 30 days of storage. The highest survival was seen for the Lactobacillus reuteri strain. The co-fermentation in the presence of a yoghurt culture gave a lower pH in the final products and decreased the survival rates for all strains. The nutritional effects of fermented, oat-based products were investigated by using germfree and conventional rats and by performing a clinical study on 56 healthy subjects. Physiological parameters such as changes in serum cholesterol, faecal excretion of cholesterol, and amount of faecal short chain fatty acids (SCFA) were studied. In addition, changes in the faecal flora were investigated in the human subjects. No changes in the serum cholesterol levels or faecal excretion of cholesterol were seen in the rat models when the animals were fed on different, fermented oat-based products. A diffeent feacal SCFA pattern was seen in the conventional rats fed on the oat-based diets than in the group fed on rice. A lipid lowering effect was seen in the human subjects eating a fermented, ropy, oat-based product for five weeks, which gave a dietary intake of 3.5 g native beta-glucans from oat per day. An increase in total bacterial count and Bifidobacterium subsp. was also seen in faecal samples from these subjects. In conclusion, there is a potential for the development of fermented, non-dairy, oat-based products with a "ropy" structure containing soluble fibres of both native and microbial origin and with the lipid lowering effect that is generally associated with an intake of oat.

    • Four strains of lactic acid bacteria were used as pure or mixed cultures to investigate the texture-promoting capacity and exopolysaccharides (EPS) formation in three different oat-based, non-dairy milk products, Adavena G40, Adavena M40 (both with a dry matter content of 20%) and Mill Milk (with a dry matter content of 10%). Viscosity was measured at two different shear rates during 2 min of shear thinning. The highest viscosity was measured at a shear rate of 129 s(-1) when a mixed culture consisting of Lactobacillus delbrueckii subsp. bulgaricus NCFB 2772 and Lactobacillus brevis DSM 1269 was grown at 30degreesC in the medium containing mostly glucose as the carbohydrate source (Adavena G40). The mixed culture consisting of Lactobacillus delbrueckii subsp. bulgaricus NCFB 2772 and Streptococcus thermophilus DSM 20259 gave the highest viscosity when using the medium with a dry matter content of 20% and maltose as the main carbohydrate source (Adavena M40). The EPS formation in the Adavena G40 medium was confirmed by isolating the soluble polymer fraction after fermentation. A higher yield of polymer dry mass was obtained from the samples with higher viscosity. The study shows that the eo-operative growth that occurs when using mixed cultures also influences the EPS formation and final viscosity in the different oat-based media. This knowledge is of importance when strains are selected for the development of new kinds of fermented, oat-based, non-dairy products.

    • The survival of the probiotic strains Lactobacillus reuteri ATCC 55730, Lactobacillus acidophilus DSM 20079 and Bifidobacterium bifidum DSM 20456, all of human origin, were investigated in three different oat-based, non-dairy products (Adavena M40, MG20 and G40). The products were fermented by the three strains with and without the presence of a commercial yoghurt culture (V2). Samples were stored at 6 degreesC up to 30 days. In general, the oat-based products were shown to be a suitable support for these intestinal bacteria. L. reuteri ATCC 55730 had the highest viability in all of the products investigated. After 30 days the cell viability of L. reuteri ATCC 55730 was 10(8) CFU ml(-1) in all three oat-based products. Lower viability was seen when the strains were grown in the presence of the yoghurt culture compared to when they were grown as pure cultures. These products also exhibited a lower pH value in comparison to products fermented with the pure cultures. The milisation of the main fermentable carbohydrates in the products varied across the bacterial strains. A decrease in beta-glucan content was seen for the products fermented by the B. bifidum DSM 20456 strain. This work shows that these oat-bases with different mono- and disaccharide composition can be used to support the growth of human intestinal bacteria and also maintain high cell viability during cold storage.

    • The ultrafast timescale of electron transfer processes is crucial to their role in many biological systems and technological devices. In dye-sensitized solar cells(1-4), the electron transfer from photoexcited dye molecules to nanostructured semiconductor substrates needs to be sufficiently fast to compete effectively against loss processes and thus achieve high solar energy conversion efficiencies(4). Time-resolved laser techniques indicate an upper limit of 20 to 100 femtoseconds(5-9) for the time needed to inject an electron from a dye into a semiconductor, which corresponds to the timescale on which competing processes such as charge redistribution(10,11) and intramolecular thermalization of excited states(12-14) occur. Here we use resonant photoemission spectroscopy, which has previously been used to monitor electron transfer in simple systems with an order-of-magnitude improvement in time resolution(15,16), to show that electron transfer from an aromatic adsorbate to a TiO2 semiconductor surface can occur in less than 3 fs. These results directly confirm that electronic coupling of the aromatic molecule to its substrate is sufficiently strong to suppress competing processes(17).

  • Effect of Substrate on Bottom-Up Fabrication and Electronic Properties of Graphene Nanoribbons

    • Taking into account the technological demand for the controlled preparation of atomically precise graphene nanoribbons (GNRs) with well-defined properties, the present thesis is focused on the investigation of the role of the underlying metal substrate in the process of building GNRs using bottom-up strategy and on the changes in the electronic structure of GNRs induced by the GNR-metal interaction. The combination of surface sensitive synchrotron-radiation-based spectroscopic techniques and scanning tunneling microscopy with in situ sample preparation allowed to trace evolution of the structural and electronic properties of the investigated systems.Significant impact of the substrate activity on the growth dynamics of armchair GNRs of width N = 7 (7-AGNRs) prepared on inert Au(111) and active Cu(111) was demonstrated. It was shown that unlike inert Au(111) substrate, the mechanism of GNRs formation on Ag(111) and Cu(111) includes the formation of organometallic intermediates based on the carbon-metal-carbon bonds. Experiments performed on Cu(111) and Cu(110), showed that a change of the balance between molecular diffusion and intermolecular interaction significantly affects the on-surface reaction mechanism making it impossible to grow GNRs on Cu(110).It was demonstrated that deposition of metals on spatially aligned GNRs prepared on stepped Au(788) substrate allows to investigate GNR-metal interaction using angle-resolved photoelectron spectroscopy. In particular intercalation of one monolayer of copper beneath 7-AGNRs leads to significant electron injection into the nanoribbons, indicating that charge doping by metal contacts must be taken into account when designing GNR/electrode systems. Alloying of intercalated copper with gold substrate upon post-annealing at 200°C leads to a recovery of the initial position of GNR-related bands with respect to the Fermi level, thus proving tunability of the induced n-doping. Contrary, changes in the electronic structure of 7-AGNRs induced by the deposition of Li are not reversible.  It is demonstrated that via lithium doping 7-AGNRs can be transformed from a semiconductor into a metal state due to the partial filling of the conduction band. The band gap of Li-doped GNRs is reduced and the effective mass of the conduction band carriers is increased.

    • Here, we investigate induction of immunological tolerance by lentiviral based gene therapy in a mouse model of rheumatoid arthritis, collagen II-induced arthritis (CIA). Targeting the expression of the collagen type II (CII) to antigen presenting cells (APCs) induced antigen-specific tolerance, where only 5% of the mice developed arthritis as compared with 95% of the control mice. In the CII-tolerized mice, the proportion of Tregs as well as mRNA expression of SOCS1 (suppressors of cytokine signaling 1) increased at day 3 after CII immunization. Transfer of B cells or non-B cell APC, as well as T cells, from tolerized to naive mice all mediated a certain degree of tolerance. Thus, sustainable tolerance is established very early during the course of arthritis and is mediated by both B and non-B cells as APCs. This novel approach for inducing tolerance to disease specific antigens can be used for studying tolerance mechanisms, not only in CIA but also in other autoimmune diseases.

    • Service blueprints are usually included in listings of standard methods within service design. Still; little research has been conducted on service blueprints. The case study at hand explores how blueprints can be applied in a situation with three key actors; all with different motives and wishes. The case study is within the domain of car parking; a service which at a first glance may seem simple; but is rather complex when scrutinized. Three ways of blueprinting the situation are presented and discussed in the paper. Finally issues which arose from the blueprinting process are discussed in regard to implications for people creating blueprints.
