Kursen som helhet ökade de närståendes känsla av trygghet och minskade deras upplevelse av stress och oro. Syfte: Syftet var att utvärdera hur närstående uppfattade kursen "Lära sig leva med demenssjukdom". Kursen avsåg att ge stöd i relationen mellan personen med demenssjukdom och den närstående. Metod: Utvärderingen genomfördes som deltagarbaserad aktionsforskning med fokusgruppssamtal. Deltagarna var närstående till personer med demenssjukdom. Resultat: Närstående upplevde en känsla av trygghet genom att få utbildning och information om demenssjukdomen. Att prata med andra i liknande situation och att känna igen sig gav dem en känsla av samhörighet.Diskussion: Upplevelse av att inte känna sig ensam i situationen och att få dela med sig av sina erfarenheter till andra. Kursen som helhet ökade de närståendes känsla av trygghet och minskade deras upplevelse av stress och oro.

    • Gruvindustrins betydelse för samhällsutveckling och infrastruktur i Sverige och inte minst i Norrbottens län är mycket stor. De geologiska förutsättningarna att hitta nya brytvärda förekomster i Norrbotten är goda. Länet är tillsammans med Västerbotten en av Europas viktigaste regioner för utvinning av metaller. Det syns också i den nyligen framtagna regionala mineralstrategin för Norrbotten och Västerbotten. Visionen för den regionala mineralstrategin: ”Genom långsiktigt hållbart nyttjande av Norrbottens och Västerbottens läns mineralresurser har ytterligare tillväxt skapats i regionen och hela Sverige. Vi har utvecklat och stärkt vår ställning som ledande gruv- och mineralnation.”Eftersom framtidspotentialen för gruvnäringen är mycket god men okunnigheten hos både allmänhet och beslutsfattare om näringens betydelse för innovation och samhällsutveckling är stor, kopplat med en utbredd oro för miljöpåverkan, måste dessa viktiga framtidsfrågor belysas. Med finansiering från Länsstyrelsen i Norrbotten bedrevs därför under första hälften av 2014 en förstudie som syftade till att sammanfatta kunskapsläget om framtidens gruvindustri i Norrbotten. Resultaten av förstudien redovisas i den här rapporten. En viktig slutsats är att det under nästa strukturfondsperiod (med start 2015) behövs ett framtidsinriktat forskningsprogram för att belysa de möjligheter som finns. Denna förstudie utgör grund för en kommande ansökan till strukturfonderna. Kompetensen som finns vid Luleå tekniska universitet, Sveriges centrum för gruvrelaterad forskning och utbildning, bör användas för att studera troliga framtidsmöjligheter och hur de ska kunna användas för att få en så positiv utveckling som möjligt för länet. Projektet bör innehålla följande tre huvudinriktningar, som naturligtvis hör ihop:Vilka malm- och mineralresurser finns det potential för i Norrbotten, och vilka kommer sannolikt att exploateras i framtiden?Vad kommer den exploateringen att ha för betydelse för innovation och samhällsutveckling?Vad kommer den exploateringen att få för miljöeffekter och hur ska man göra för att minska miljöbelastningen?En annan slutsats är att nedlagda gruvområden inte måste ses som förstörd natur. Betydande mervärden som gruvturism skulle kunna skapas om vilja, kreativitet och beslutsamhet finns. Detta är ett givet utvecklingsområde där småföretag och entreprenörer kan göra stor insats om de politiska och myndighetsmässiga förutsättningarna finns. Dessa aspekter skulle också kunna belysas i det föreslagna forskningsprogrammet eller i ett eget projekt.

    • The REMinE project is organized in five work packages that comprise: detailedcharacterization and risk assessment of the mine wastes selected (WP2), identification of new processing methods for mine waste (WP3), characterization and risk assessment of the remaining residuals (WP4), outlining business opportunities and environmental impact in a conceptual model for sustainable mining (WP5). The project comprises case studies of historical mine wastes from three different European countries, namely Portugal, Romania and Sweden. The interdisciplinary research collaboration in this project is innovative in the sense that separation of minerals and extraction of metals not only are basedon technical and economic gain but also considers the environmental perspective.

It focuses on the ambivalent role that the transnational ‘rainbow’ space plays for community building for LG- BTQ activists in in Pakistan. e rainbow ag can function as a way to mobilize an imagined transnational community of belonging, enabling people to politicize their experiences of discrimination as a demand of recognition directed at the sta- te. But it can also enable homonationalism and transnational middle class forma- tions that exclude groups of people, for example illiterates and people perceived of as traditional, such as Khwaja Siras. e article is based on auto-ethnographic re ections on encounters with activists in Pakistan, and critically discusses the problem of feeling ‘too comfortable’, as white, Western, middle-class researchers, exploring ‘imperial narratives’ dominating the feminist and LGBTQ activist trans- national imagined community of belonging. It argues for the importance of recog- nizing the transnational space as a space in its own right, with di erent positions, communities and con icts stretching around the globe.

To provide personalized treatment options for bipolar patients, it is essential to identify prediction biomarkers such as polygenic scores. In this study, we developed a polygenic score for lithium treatment response (Li+PGS) in patients with BD. To gain further insights into lithium's possible molecular mechanism of action, we performed a genome-wide gene-based analysis. Using polygenic score modeling, via methods incorporating Bayesian regression and continuous shrinkage priors, Li+PGS was developed in the International Consortium of Lithium Genetics cohort (ConLi+Gen: N=2,367) and replicated in the combined PsyCourse (N=89) and BipoLife (N=102) studies. The associations of Li+PGS and lithium treatment response - defined in a continuous ALDA scale and a categorical outcome (good response vs. poor response) were tested using regression models, each adjusted for the covariates: age, sex, and the first four genetic principal components. Li+PGS was positively associated with lithium treatment response in the ConLi+Gen cohort, in both the categorical (P=9.8×10-12, R2=1.9%) and continuous (P=6.4×10-9, R2=2.6%) outcomes. Compared to bipolar patients in the 1st decile of the risk distribution, individuals in the 10th decile had 3.47-fold (95%CI: 2.22-5.47) higher odds of responding favorably to lithium. The results were replicated in the independent cohorts for the categorical treatment outcome (P=3.9×10-4, R2=0.9%), but not for the continuous outcome (P=0.13). Gene-based analyses revealed 36 candidate genes that are enriched in biological pathways controlled by glutamate and acetylcholine. Li+PGS may be useful in the development of pharmacogenomic testing strategies by enabling a classification of bipolar patients according to their response to treatment.

    • Lithium is a first-line mood stabilizer for the treatment of bipolar affective disorder (BPAD). However, the efficacy of lithium varies widely, with a nonresponse rate of up to 30%. Biological response markers are lacking. Genetic factors are thought to mediate treatment response to lithium, and there is a previously reported genetic overlap between BPAD and schizophrenia (SCZ).To test whether a polygenic score for SCZ is associated with treatment response to lithium in BPAD and to explore the potential molecular underpinnings of this association.A total of 2586 patients with BPAD who had undergone lithium treatment were genotyped and assessed for long-term response to treatment between 2008 and 2013. Weighted SCZ polygenic scores were computed at different P value thresholds using summary statistics from an international multicenter genome-wide association study (GWAS) of 36989 individuals with SCZ and genotype data from patients with BPAD from the Consortium on Lithium Genetics. For functional exploration, a cross-trait meta-GWAS and pathway analysis was performed, combining GWAS summary statistics on SCZ and response to treatment with lithium. Data analysis was performed from September 2016 to February 2017.Treatment response to lithium was defined on both the categorical and continuous scales using the Retrospective Criteria of Long-Term Treatment Response in Research Subjects with Bipolar Disorder score. The effect measures include odds ratios and the proportion of variance explained.Of the 2586 patients in the study (mean [SD] age, 47.2 [13.9] years), 1478 were women and 1108 were men. The polygenic score for SCZ was inversely associated with lithium treatment response in the categorical outcome, at a threshold P<5×10-2. Patients with BPAD who had a low polygenic load for SCZ responded better to lithium, with odds ratios for lithium response ranging from 3.46 (95% CI, 1.42-8.41) at the first decile to 2.03 (95% CI, 0.86-4.81) at the ninth decile, compared with the patients in the 10th decile of SCZ risk. In the cross-trait meta-GWAS, 15 genetic loci that may have overlapping effects on lithium treatment response and susceptibility to SCZ were identified. Functional pathway and network analysis of these loci point to the HLA antigen complex and inflammatory cytokines.This study provides evidence for a negative association between high genetic loading for SCZ and poor response to lithium in patients with BPAD. These results suggest the potential for translational research aimed at personalized prescribing of lithium.

    • I denna studie undersöks hur föreställningar kring normalitet kommer till uttryck och förhandlas i de vardagliga praktikerna i skolans tidigare år. I särskilt fokus är normer kring ålder, genus och sexualitet. Det empiriska materialet är skapat genom deltagande observation och intervjuer med elever och lärare i två klasser i två skolor (årskurs 2 och 3). I studien används poststrukturalistiska teorier för att belysa hur makt verkar i det vardagliga, både i att göra begriplighet och meningsfullhet men också att definiera avvikelser och normalisera utanförskap och underordning. Analysen visar hur elever och lärare iscensätter och ifrågasätter olika förväntningar på hur man ska vara och agera som barn, vuxna, elever, lärare, flicka, pojke, etcetera. Detta "görande" av normer kopplas i studien till skapandet av olika tillhörigheter, formella och informella miljöer, skillnader mellan lektioner och raster, klassrum och omklädningsrum eller mellan allvar och skratt. Studien visar att sådana växlingar ger utrymme för förhandlingar och utmaningar i vad som kan betraktas som osäkra ormråden eller gränsland. Genom att rikta fokus mot hur normer samverkar visar studien vidare att utmaningar och ifrågasättanden av normer kring genus och sexualitet riskerar att trivialiseras i relation till elevernas position som barn. Detta innebär att även om normerna verkar som gränser för det möjliga och önskvärda, kan man inte likställa utmaningar och överskridande av gränser med omvandling av normer. Studien hävdar således att pedagogiskt arbete som syftar till att problematisera hur normer upprepas och utmanas i skolan måste överväga tvetydigheter och osäkra som en del av att skapa hierarkiska ordningar.

    • Objective-To examine whether DNA microarray analysis of chromosomal susceptibility regions for allergy can help to identify candidate genes. Material and Methods-Nasal biopsies were obtained from 23 patients with allergic rhinitis and 12 healthy controls. RNA was extracted from the biopsies and pooled into three patient and three control pools. These were then analysed in duplicate with DNA microarrays containing 12626 genes. Candidate genes were further examined in nasal biopsies (real-time polymerase chain reaction) and blood samples (single nucleotide polymorphisms) from other patients with allergic rhinitis and from controls. Results-A total of 37 differentially expressed genes were identified according to criteria involving both the size and consistency of the gene expression levels. The chromosomal location of these genes was compared with the chromosomal susceptibility regions for allergic disease. Using a statistical method, five genes were identified in these regions, including serine protease inhibitor, Kazal type, 5 (SPINK5) and HLA-DRB2. The relevance of these genes was examined in other patients with allergic rhinitis and in controls; none of the genes were differentially expressed in nasal biopsies. Moreover, no association between allergic rhinitis and SPINK5 polymorphisms was found, at either the genotype or haplotype level. Conclusions-DNA microarray analysis of chromosomal susceptibility regions did not lead to identification of candidate genes that could be validated in a new material. However, because gene polymorphisms may cause differential gene expression, further studies, including validation data, are needed to examine this approach.

    • The notion of Sweden as a secular nation-state, or rather the linkage between notions of secularism and gender equality, is strong in public discourse. Within this frame, religion is located in a traditional past and often understood as a hindrance to liberal and modern values.In this chapter we focus on our own situatedness as feminist researchers living in Sweden and thereby explore how, where and why ideologies of secularism entangled with notions of European values and superiority become dominant. Inspired by the feminist tradition of memory work, an aim is to explore the boundary between the secular and the religious through our own experiences and from our location in Sweden. The aim is also to search for counter-memories, both in the doing of secular (gendered) selves as well as the ongoing production of the “religious other”.

By studying Swedish contemporary debates about freedom of speech, veils, gender equality, and the in/tolerant society, we aim to unpack how secularism is done in a Swedish neoliberal contemporary context. Inspired by anthropologist of secularism, Talal Asad, we wish to contribute to the undermining of the tightly knit weave of secularism, reason and critique in Western discourse.

    • BACKGROUND: Depression is a common cause of sickness absence (SA) and also highly associated with stigma. Few studies have addressed the role of stigma in relation to SA. OBJECTIVE: To investigate if attitudes to depression were associated with the public's opinion of depression as a valid reason of SA. METHODS: The study population (n = 2413) originated from a web-based panel of citizens. The survey included a short vignette describing a person with symptoms of depression and the person's work tasks, followed by a question on recommendation of SA. Negative attitudes were measured by the Depression Stigma Scale. Logistic regressions were used to estimate the odds ratios (OR) for the likelihood of not recommending SA, controlling for individual and work-related co-variates. RESULTS: The crude association between negative attitudes and not recommending SA was OR 2.15 (95% CI, 1.76-2.62). In the fully adjusted model the OR was 1.76 (95% CI, 1.40 - 2.21) for not recommending SA. CONCLUSIONS: Participants with negative attitudes to depression were more likely to not consider depression as a valid reason of sickness absence. The study supports theories on layered stigma; attitudes from one arena are related to other arenas. Future studies are needed to confirm our findings.

    • ObjectiveThe Internet plays a major role for pregnant women in seeking knowledge and for getting in touch with like-minded women. The information is available at all hours and can be accessed anywhere. The information provides the women with a sense of control and confidence but the large amount of information available can also be overwhelming. The aim of this study was to identify how women use the Internet as a source of information during their pregnancy and how it affects them.Design and settingA descriptive cross-sectional study was conducted.Data were collected through a questionnaire at antenatal clinics in the southern Sweden. The data were analyzed descriptivelyParticipantsA total of 193 Swedish women, pregnant at least 34 weeks, participated in the study. The response rate was 94%.FindingsAlmost all (95%) of the women in the study used the Internet as a source of information. The main reason was to find information and read about people in the same situation. Reading pregnancy-related information on the Internet was seen as positive. However, a majority of the woman experienced feelings of worry due to something they read online. These feelings were most commonly coped with by talking to a partner, relatives, and friends or by asking the midwife at their next appointment. Eleven per cent of the women contacted the general healthcare services because of their feelings of worry.ConclusionAlmost all women in this study searched the Internet to find pregnancy-related information, despite being satisfied with the information they received from the ANC. Using the Internet was seen as complementary to the information from professionals. It also caused feelings of worry, which could lead to the woman contacting healthcare services for support. ANC could help to reduce these feelings for some women by informing about the advantages and disadvantages with online information and recommending suitable web pages.

    • This article employs a holistic understanding of environmental, social, and economic sustainability to explore the interaction between neoliberalism, climate crisis, digitalization, and academic work with a focus on its everyday aspects. Drawing our experience of organizing an online conference during the Covid-19 pandemic and our dialogue with conference participants, we first problematize the presumed disembodiment of digital exchange and suggest a nuanced understanding of physicality’s role in knowledge production. We then explore the impact of the changing times and spaces of academic work on bodies and minds and the boundaries between private and public realms. Finally, we challenge the notion of digital solutionism by highlighting the implications of inhabiting digital platforms as spaces for knowledge production. While there is no simple solution to the problems around the digital shift in academic work and conferencing, we argue, downsizing can be a counteraction to platform capitalism in times of the climate crisis.

    • Tumor infiltration by lymphocytes has been linked to improved clinical outcome in children with neuroblastoma (NB) but T-cell activation has never been demonstrated to occur within the NB microenvironment. Here we show that tumor-associated lymphocytes (TALs) obtained from lesions representing all genetic subsets of NB and autologous peripheral blood lymphocytes (PBLs) analyzed on the day of tumor excision differed in composition, phenotype and functional characteristics. The NB microenvironment appeared to promote the accumulation of CD3(+)CD8(+) T cells and contained a larger proportion of T cells expressing the interleukin-2 receptor α chain (CD25) and manifesting an effector memory (CCR7(-)CD45RA(-)) phenotype. Accordingly, the stimulation of PBLs with autologous tumor cells in short-term cultures increased the proportion of effector memory T cells, upregulated CD25, stimulated the expression of the TH1 cytokines interferon γ and tumor necrosis factor α, and reduced the expression of transforming growth factor β. In situ proliferation as well as a characteristic pattern of T-cell receptor aggregation at the contact sites with malignant cells was revealed by the immunohistochemical staining of TALs in primary tumors, indicating that the NB milieu is compatible with the activation of the immune system. Our results are compatible with the hypothesis that CD8(+) T cells are specifically activated within the NB microenvironment, which appears to be permissive for effector memory responses.

    • Response to lithium varies widely between individuals with bipolar disorder (BD). Polygenic risk scores (PRSs) can uncover pharmacogenomics effects and may help predict drug response. Patients (N = 2,510) with BD were assessed for long-term lithium response in the Consortium on Lithium Genetics using the Retrospective Criteria of Long-Term Treatment Response in Research Subjects with Bipolar Disorder score. PRSs for attention-deficit/hyperactivity disorder (ADHD), major depressive disorder (MDD), and schizophrenia (SCZ) were computed using lassosum and in a model including all three PRSs and other covariates, and the PRS of ADHD (β = -0.14; 95% confidence interval [CI]: -0.24 to -0.03; p value = 0.010) and MDD (β = -0.16; 95% CI: -0.27 to -0.04; p value = 0.005) predicted worse quantitative lithium response. A higher SCZ PRS was associated with higher rates of medication nonadherence (OR = 1.61; 95% CI: 1.34-1.93; p value = 2e-7). This study indicates that genetic risk for ADHD and depression may influence lithium treatment response. Interestingly, a higher SCZ PRS was associated with poor adherence, which can negatively impact treatment response. Incorporating genetic risk of ADHD, depression, and SCZ in combination with clinical risk may lead to better clinical care for patients with BD.

  • Harnessing the immunoregulatory capacity of single-stranded oligonucleotides to modulate innate immunity

    • As the first line of defense, the innate immune system is exceptionally efficacious against invading pathogens but must be tightly regulated to avoid immune-related pathogenesis. Accordingly, the innate immune system recognizes highly conserved components associated with infections and damaged cells using a limited number of pattern-recognition receptors (PRRs), which are differentially expressed across cell types. Importantly, tissue damage and viral infections can induce the release and production of nucleic acids, which can be sensed by nucleic-acid sensing Toll-like-receptors (TLRs), a subset of PRRs located in cellular endosomes that encompass the receptors TLR3,7,8 or 9. It is imperative that these receptors can differentiate “self” nucleic acids from “non-self” in order to avoid autoimmune reactions. However, increasing data has shown that over-active signaling from these receptors can contribute to TLR-mediated inflammatory and auto-immune consequences. Previously, a 35 bases long single-stranded oligonucleotide (ssON) was revealed to inhibit TLR3 activation in dendritic cells (DCs). Hence, the research presented in this thesis aims to harness the capacity of certain immunosuppressive oligonucleotides to modulate the innate immune response to inflammation and viral infection. In Paper I we demonstrated that ssON inhibits certain receptor-mediated endocytosis, thereby preventing activation from TLR3/4/7 signaling endosomes Additionally, ssON modulated TLR3-mediated activation in vivo in the skin. Therefore, in Paper II, we further assessed ssON’s effect on prominent symptoms of inflammatory skin disorders. We demonstrated that ssON inhibits non-IgE-mediated degranulation of mast cells induced by certain ligands of the pseudo-allergic receptor MRGPRX2, thereby alleviating the respective downstream itch and inflammation.  There is currently an urgent need to develop new broad-spectrum antivirals against acute respiratory tract infections, which at present remain the deadliest communicable diseases. Since ssON inhibits clathrin-mediated and caveolin-dependent endocytosis, which are cellular entry pathways utilized by many viruses, we investigated ssON’s potential as an anti-viral agent against two major respiratory viruses. In Paper III we aimed to assess the effect of ssON on influenza A virus (IAV) infection. We demonstrated that ssON inhibits the infection of IAV, and more specifically the pandemic H1N1, in vitro in DCs and reduces viral load and disease in murine models of IAV infection. In Paper IV we aimed to assess the anti-viral capacity of ssON on respiratory syncytial virus (RSV) infection. We discovered that ssON acts as an efficient antiviral agent against RSV in vitro, seemingly by competing with the virus for binding to the entry receptor nucleolin. Additionally, ssON inhibits viral infection in vivo in a murine model of RSV infection, and the combination of ssON treatment and RSV infection leads to an upregulation of immune-related genes in the lungs, which most likely aid in viral clearance.In summary, the research presented in this thesis uncovers novel immunomodulatory functions of synthetic, noncoding ssONs to modulate the innate immune responses in the context pseudo- allergic itch and inflammation as well as in IAV and RSV infection.  Furthermore, these studies unveil prospective therapeutic possibilities for ssON as an antiviral agent or as a treatment for certain inflammatory dermatoses.

    • Psoriasis is an inflammatory skin disorder affecting approximately 3% of the population. Genetic studies published so far have shown a complex genetic inheritance with heterogeneity and a putative major susceptibility locus in the HLA region on chromosome 6. We have collected a large amount of material consisting mostly of small nuclear families in order to perform a genome-wide scan for psoriasis-associated genes. In order to focus the scan properly on possible candidate regions, we performed a cytogenetic analysis of 477 unrelated psoriatics. We divided our findings into sporadic, affecting a minor fraction of the cells, and constitutional, i.e. they were present in all cells examined. We found three cases of balanced translocation, all of which involved chromosome 11q. Two of these had a breakpoint in q12-13, whilst one involved the telomeric part of chromosome 11q. In order to characterise further the breakpoint on 11q12-13, we used bacterial artificial chromosomes (BACs) analysed by fluorescent in situ hybridisation (FISH). We were able to show that the persons had a close, but not identical breakpoints; they were separated by at least 5 cM. The major atopy locus is located in this region, as well as a locus for insulin-dependent diabetes mellitus, both being conditions with a pathogenetic mechanism involving antigen presentation.

    • Psoriasis is a heterogeneous disease in which several reports suggest the presence of a susceptibility gene in or in the proximity of the human leukocyte antigen complex in chromosome 6p. There is an association between HLA-Cw6 and young onset of the disease. The S gene (corneodesmosin), located 160 kb telomeric of HLA-C, is a strong candidate for psoriasis due to its reportedly exclusive expression in differentiating keratinocytes. We have studied this gene in a large Swedish psoriasis population and we report a strikingly high degree of polymorphism in the coding parts of the gene, 1 every 100 base pairs. We used a stratified approach to compare the polymorphic variants in patients and controls. A single nucleotide polymorphism in the coding region leading to an amino acid exchange (Ser-->Phe) that differed significantly between patients and controls was identified (position 619). Owing to a high allele frequency in a larger control group, however, and an insignificant influence of the variant on the age at onset distribution curve based on a large psoriasis population, we could not confirm that this coding single nucleotide polymorphism was involved in disease etiology. We also examined the single nucleotide polymorphism in position 1243, recently proposed to have an influence on the pathogenesis of the disease. This polymorphism showed less association to the disease as compared with the single nucleotide polymorphism at positions 619 and 722. Such a high degree of variation present also in an HLA gene which is not involved in immune response indicates the difficulty involved in assessing the role of a specific allele in the pathogenesis of a complex disease in this region. A strong association effect due to linkage disequilibrium in an extended region in the HLA complex is also a complicating factor.

    • Psoriasis vulgaris is strongly associated with certain human leukocyte antigens, especially in early onset. The purpose of this study was to study the HLA-Cw6 allele and its contribution to disease susceptibility in a set of 104 families with at least two affected siblings. A sequencing method was utilized to examine the two exons that build up the antigen binding site of the C locus receptor. DNA from patients homozygous for Cw6 based on haplotype information were sequenced. The results confirmed the identity of the Cw6 allele in affected individuals with the consensus sequence for Cw*0602. We screened the set of families for psoriasis patients homozygous for Cw6 and found 11 individuals with a mean age at onset of 16.1 years. The corresponding figure for the Cw6 heterozygotes was 18.45 years and for the Cw6-negatives 22.36 years. This is indicative of a gene dose effect. We performed a transmission disequilibrium test (TDT) on the Cw6 allele per se, used as a biallelic marker. The analysis resulted in a P-value of 5.3 x 10(-17) (t167/nt45). This greatly exceeds our previous results of a TDT in the region, including microsatellite markers and single nucleotide polymorphisms (SNPs) in the coding part of the S gene (corneodesmosin), which is a suggested candidate gene in the region. The maximum nonparametric linkage (NPL) value was also reached using HLA-C as a marker. We conclude that Cw6 is the allele which shows the highest degree of association with psoriasis in our set of families and we propose that it directly influences the age at onset of the disease rather than increasing the genetic load in accordance with a polygenic theory.

    • Psoriasis is known to be a heterogeneous disease with so far three reported major psoriasis susceptibility loci on chromosome 4q, 6p and 17q. In this study we investigated three reported gene locations by nonparametric and parametric linkage analysis in a large family set consisting of 104 families (153 sib pairs) from Sweden. We could confirm linkage to chromosome 6p. A maximum heterogeneous lod score of 2.78 was reached at locus D6S276 (alpha = 0.60). Allelic association studies within the HLA region indicated linkage disequilibrium at locus TNFbeta with a significant p value of 0.0009. Furthermore, we obtained weak evidence of linkage to the locus on chromosome 17q while no evidence of linkage could be found to the chromosome 4q region.

    • We have performed a pair-wise linkage study in the search for psoriasis susceptibility regions. A preliminary scan was performed on 20 families. In this set we obtained indications of linkage on chromosome 3q21. This region was further investigated using material from a total of 104 families (set 1B) resulting in a non-parametric linkage (NPL) of 1.77. The material was stratified in families whose parental origin is in southwest Sweden (set 1C). A maximum NPL value of 2.77 was obtained in this group. A transmission disequilibrium test (TDT) was performed on the stratified material (set 1C) and a significant P value of 0.005 was obtained, at marker D3S1269. The locus was confirmed with TDT in replicate material consisting of 148 families in which a single member was affected (P value 0.0007) at marker D3S1551. Thus, we have observed a significant P value using TDT in the vicinity of markers D3S1269/D3S1551, suggesting a novel psoriasis susceptibility region.

    • In this article, we focus on a Swedish upper class digital space called The Good Society, where texts, videos and discussions are published. In this digital space the elite comes out publicly as underdogs in relation to the supposed hegemony of political correctness and the Swedish welfare ideology. The upper class take the position of protectors against the feminization of society and the crisis of masculinity. We argue that this space can be understood in terms of what Foucault described as a heterotopia, a form of peculiar counter-space. Using multi-sited digital ethnography we discern a playful mimicking of the state, the public, the media and public conversation. The article concludes that it is important to acknowledge and critically examine such playful and peculiar heterotopias as counter hegemonic strategies, in order to understand how digital spaces are political and serve right-wing, masculinist populist mobilization.

    • This thesis is a study of the working life of fashion models in the Swedish Fashion Industry based on 18 months of ethnographic fieldwork among a network of female and male models in Stockholm. It charts the development of the modelling business in Sweden and locates it within the changing labour market of late capitalism. The study highlights the paradoxes and contradictions of the modelling profession, which provides both extraordinary career opportunities but is also full of risks and temptations, especially for young female models. Unlike many other studies of the model business, the working lives of male models and the particular challenges they face also receive attention. Two main theoretical areas are examined in this book. One deals with the fact that a model’s body is a sexualized commodity in the market. The ability to project a sexual image at work can enhance a model’s career, but this sexualization and objectification is experienced as problematic by male and female models alike, although for different reasons. In the case of women, it involves the need to appear respectable; while for men attempting to live up to heteronormative expectations of masculinity is continually challenged in a profession coded as ‘female’. A solution employed by many models is to create a professional mask or job-persona, that is detached from what they regard as their inner or authentic self. The issue of authenticity emerges as an important concern for many models and is a theme that runs through the entire thesis. A second theoretical area is the power relations that structure the modelling business, including the relationship between fashion models and model agencies, stylists, and photographers. How models manage these power relations in the form of emotional labour, compromises, or even by occasional challenges to authority is illuminated in the study.

    • The social-democratic-inspired "Nordic model", with its agenda for gender equality, has been an important example for the development of political interventions to transform society but at the same time, it has functioned as an emerging gender normalising and stabilising structure. In the last decade it has also become the focus of antigender movements and ethno-nationalistic parties both as emblematic for the Nordic nations as well as a threat that must be destroyed to save the nation. This issue will elaborate further on gender equality as a node, a floating signifier in powerful and often contradictory discourses. We are inspired by scholarships of hope in a dialogue with articles that search for realistic utopias that might be considered to be "beyond gender equality". The included articles engage with the messiness and crossroads of gender equality in relation to the work-line, territories, neo-liberalism, religion, the crisis of solidarity and the success of anti-genderism agenda.

    • Tumors with Anaplastic Lymphoma Kinase (ALK) fusion rearrangements, including non-small cell lung cancer and anaplastic large cell lymphoma, are highly sensitive to ALK tyrosine kinase inhibitors (TKIs), underscoring the notion that such cancers are addicted to ALK activity. While mutations in ALK are heavily implicated in childhood neuroblastoma, response to the ALK TKI crizotinib has been disappointing. Embryonal tumors in patients with DNA repair defects such as Fanconi anemia (FA) often have a poor prognosis, due to lack of therapeutic options. Here we report a child with underlying FA and ALK mutant high-risk neuroblastoma responding strongly to precision therapy with the ALK TKI ceritinib. Conventional chemotherapy treatment caused severe, life-threatening toxicity. Genomic analysis of the initial biopsy identified germ-line FANCA mutations as well as a novel ALK-I1171T variant. ALK-I1171T generates a potent gain-of-function mutant, as measured in PC12 cell neurite outgrowth and NIH3T3 transformation. Pharmacological inhibition profiling of ALK-I1171T in response to various ALK TKIs identified an 11-fold improved inhibition of ALK-I1171T with ceritinib when compared with crizotinib. Immunoaffinity-coupled LC-MS/MS phosphoproteomics analysis indicated a decrease in ALK signaling in response to ceritinib. Ceritinib was therefore selected for treatment in this child. Mono-therapy with ceritinib was well tolerated and resulted in normalized catecholamine markers and tumor shrinkage. After 7.5 months treatment, residual primary tumor was surgically removed and exhibited hallmarks of differentiation together with reduced Ki67 levels. Clinical follow-up after 21 months treatment revealed complete clinical remission including all metastatic sites. Therefore, ceritinib presents a viable therapeutic option for ALK-positive neuroblastoma.

    • Avhandlingen undersöker bisexuella kvinnors praktiker i de maktrelationer som reglerar föreställningar om kön/genus, kroppar och sexualitet. Det är den första fördjupade studien om bisexuella liv i Sverige och fyller ett glapp inom kritisk queerforskning och feministiska studier. Syftet är att dokumentera och undersöka föreställningar om kön och sexualitet i vardagslivet och i kunskapsproduktionen från ett bisexuellt perspektiv och lyfta fram bisexualitet som en analytisk kategori.Utifrån bisexuella kvinnors intervjuberättelser belyser studien deras olika perspektiv och subjektspositioner i svensk samtid. Flera av kvinnorna var också engagerade i dragkingperformance, vilket inspirerade till att söka samband mellan bisexualitet och dragkings.Med avstamp i de teman som framkommer i intervjuerna, t ex monogama och icke-monogam relationer, kroppens materialitet, bisexuell feminism och dragkingframträdanden undersöks hur ett normativt bisexuellt mellangrund omförhandlas och hur bisexualitet opererar som en alternativ strategi i den sexuella och könade geografin. Jämfört med den etablerade kunskapen, både äldre och samtida, både approprierar och avvisar kvinnorna det bisexuella medelgrundet. Studien diskuterar nödvändigheten av ett perspektiv som härrör från bisexuella subjektspositioner och att ett bisexuellt perspektiv är användbart som analytisk kategori inom feminismen och i queerstudier.
