    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • The Visual Object Tracking Segmentation VOTS2023 challenge is the eleventh annual tracker benchmarking activity of the VOT initiative. This challenge is the first to merge short-term and long-term as well as single-target and multiple-target tracking with segmentation masks as the only target location specification. A new dataset was created; the ground truth has been withheld to prevent overfitting. New performance measures and evaluation protocols have been created along with a new toolkit and an evaluation server. Results of the presented 47 trackers indicate that modern tracking frameworks are well-suited to deal with convergence of short-term and long-term tracking and that multiple and single target tracking can be considered a single problem. A leaderboard, with participating trackers details, the source code, the datasets, and the evaluation kit are publicly available at the challenge website1

  • Efficient CsPbBr3 Perovskite Light-Emitting Diodes Enabled by Synergetic Morphology Control

    • The development of solution-processed inorganic metal halide perovskite light-emitting diodes (PeLEDs) is currently hindered by low emission efficiency due to morphological defects and severe non-radiative recombination in all-inorganic perovskite emitters. Herein, bright PeLEDs are demonstrated by synergetic morphology control over cesium lead bromide (CsPbBr3) perovskite films with the combination of two additives. The phenethylammonium bromide additive enables the formation of mixed-dimensional CsPbBr3 perovskites featuring the reduced grain size (amp;lt;15 nm) and efficient energy funneling, while the dielectric polyethyleneglycol additive promotes the formation of highly compact and pinhole-free perovskite films with defect passivation at grain boundaries. Consequently, green PeLEDs achieve a current efficiency of 37.14 cd A(-1) and an external quantum efficiency of 13.14% with the maximum brightness up to 45 990 cd m(-2) and high color purity. Furthermore, this method can be effectively extended to realize flexible PeLEDs on plastic substrates with a high efficiency of 31.0 cd A(-1).

    • The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the cause of a rapidly spreading illness, coronavirus disease 2019 (COVID-19), affecting more than seventeen million people around the world. Diagnosis and treatment guidelines for clinicians caring for patients are needed. In the early stage, we have issued "A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus (2019-nCoV) infected pneumonia (standard version)"; now there are many direct evidences emerged and may change some of previous recommendations and it is ripe for develop an evidence-based guideline. We formed a working group of clinical experts and methodologists. The steering group members proposed 29 questions that are relevant to the management of COVID-19 covering the following areas: chemoprophylaxis, diagnosis, treatments, and discharge management. We searched the literature for direct evidence on the management of COVID-19, and assessed its certainty generated recommendations using the Grading of Recommendations, Assessment, Development and Evaluation (GRADE) approach. Recommendations were either strong or weak, or in the form of ungraded consensus-based statement. Finally, we issued 34 statements. Among them, 6 were strong recommendations for, 14 were weak recommendations for, 3 were weak recommendations against and 11 were ungraded consensus-based statement. They covered topics of chemoprophylaxis (including agents and Traditional Chinese Medicine (TCM) agents), diagnosis (including clinical manifestations, reverse transcription-polymerase chain reaction (RT-PCR), respiratory tract specimens, IgM and IgG antibody tests, chest computed tomography, chest x-ray, and CT features of asymptomatic infections), treatments (including lopinavir-ritonavir, umifenovir, favipiravir, interferon, remdesivir, combination of antiviral drugs, hydroxychloroquine/chloroquine, interleukin-6 inhibitors, interleukin-1 inhibitors, glucocorticoid, qingfei paidu decoction, lianhua qingwen granules/capsules, convalescent plasma, lung transplantation, invasive or noninvasive ventilation, and extracorporeal membrane oxygenation (ECMO)), and discharge management (including discharge criteria and management plan in patients whose RT-PCR retesting shows SARS-CoV-2 positive after discharge). We also created two figures of these recommendations for the implementation purpose. We hope these recommendations can help support healthcare workers caring for COVID-19 patients.

    • The Visual Object Tracking challenge VOT2019 is the seventh annual tracker benchmarking activity organized by the VOT initiative. Results of 81 trackers are presented; many are state-of-the-art trackers published at major computer vision conferences or in journals in the recent years. The evaluation included the standard VOT and other popular methodologies for short-term tracking analysis as well as the standard VOT methodology for long-term tracking analysis. The VOT2019 challenge was composed of five challenges focusing on different tracking domains: (i) VOT-ST2019 challenge focused on short-term tracking in RGB, (ii) VOT-RT2019 challenge focused on "real-time" short-term tracking in RGB, (iii) VOT-LT2019 focused on long-term tracking namely coping with target disappearance and reappearance. Two new challenges have been introduced: (iv) VOT-RGBT2019 challenge focused on short-term tracking in RGB and thermal imagery and (v) VOT-RGBD2019 challenge focused on long-term tracking in RGB and depth imagery. The VOT-ST2019, VOT-RT2019 and VOT-LT2019 datasets were refreshed while new datasets were introduced for VOT-RGBT2019 and VOT-RGBD2019. The VOT toolkit has been updated to support both standard short-term, long-term tracking and tracking with multi-channel imagery. Performance of the tested trackers typically by far exceeds standard baselines. The source code for most of the trackers is publicly available from the VOT page. The dataset, the evaluation kit and the results are publicly available at the challenge website(1).

    • Digital LLRF technology has been widely used in new generation particle accelerators. IF quadrature sampling is a common method for amplitude and phase detection. Many strategies, which obey the same rule of f(sample) = (M/N)f(IF) (M/N is a rational number), have been proposed to reduce the effects of spectrum aliasing. However, we found that M/N does not need to be a rational number according to Shannon's theorem. Therefore, we propose a new IQ detection method in this paper. This method is based on a special IIR filter which is derived from an RLC circuit. The unique characteristic of the method is that the value of f(IF) is independent of the value of, f(sample). We have set up an experimental platform to verify our method. A 122.88 MHz sampling clock is used to sample a 3 MHz IF signal. The DOS and PI control techniques are used to realize the closed-loop control. Results show that the stability of the system is within +/- 0.05% (peak to peak) for the amplitude, and with +/- 0.03 degrees (peak to peak) for the phase in 5 h. (C) 2012 Elsevier B.V.   • Quartet DNA reference materials and datasets for comprehensively evaluating germline variant calling performance

    • BACKGROUND: Genomic DNA reference materials are widely recognized as essential for ensuring data quality in omics research. However, relying solely on reference datasets to evaluate the accuracy of variant calling results is incomplete, as they are limited to benchmark regions. Therefore, it is important to develop DNA reference materials that enable the assessment of variant detection performance across the entire genome.RESULTS: We established a DNA reference material suite from four immortalized cell lines derived from a family of parents and monozygotic twins. Comprehensive reference datasets of 4.2 million small variants and 15,000 structural variants were integrated and certified for evaluating the reliability of germline variant calls inside the benchmark regions. Importantly, the genetic built-in-truth of the Quartet family design enables estimation of the precision of variant calls outside the benchmark regions. Using the Quartet reference materials along with study samples, batch effects are objectively monitored and alleviated by training a machine learning model with the Quartet reference datasets to remove potential artifact calls. Moreover, the matched RNA and protein reference materials and datasets from the Quartet project enables cross-omics validation of variant calls from multiomics data.CONCLUSIONS: The Quartet DNA reference materials and reference datasets provide a unique resource for objectively assessing the quality of germline variant calls throughout the whole-genome regions and improving the reliability of large-scale genomic profiling.

    • The deformation mechanisms and associated microstructure changes during tensile loading of an annealed twinning-induced plasticity steel with chemical composition Fe-20Mn-3Si-3Al-0.045C (wt.%) were systematically investigated using in situ time-of-flight neutron diffraction in combination with post mortem transmission electron microscopy (TEM). The initial microstructure of the investigated alloy consists of equiaxed gamma grains with the initial alpha'-phase of similar to 7% in volume. In addition to dislocation slip, twinning and two types of martensitic transformations from the austenite to alpha'- and epsilon-martensites were observed as the main deformation modes during the tensile deformation. In situ neutron diffraction provides a powerful tool for establishing the deformation mode map for elucidating the role of different deformation modes in different strain regions. The critical stress is 520 MPa for the martensitic transformation from austenite to alpha'-martensite, whereas a higher stress (>600 MPa) is required for actuating the deformation twin and/or the martensitic transformation from austenite to epsilon-martensite. Both epsilon- and alpha'-martensites act as hard phases, whereas mechanical twinning contributes to both the strength and the ductility of the studied steel. TEM observations confirmed that the twinning process was facilitated by the parent grains oriented with < 1 1 1 > or < 1 1 0 > parallel to the loading direction. The nucleation and growth of twins are attributed to the pole and self-generation formation mechanisms, as well as the stair-rod cross-slip mechanism.

    • Metallic coatings are widely employed to improve the oxidation resistance of superalloys. However, the interdiffusion between the metallic coatings and the superalloys leads to microstructural degradation in both. Some of the underlying degradation mechanisms are still elusive, e.g., the γ′ (Ni3Al) phase depletion in superalloys, where a large amount of γ′ precipitates are dissolved in the γ matrix even though the incoming Al from coatings indeed increases the Al content. Here, we investigated the interdiffusion behavior between the Amdry365 coating and the IN792 superalloy at 1100 °C, using multiple microscopic techniques and thermodynamics calculations. Our results showed an excellent agreement between experiments and thermodynamics simulations, indicating the dominant role of Al on the initial diffusion-induced phase transitions. We proposed the Al-Cr interference effect to account for the pile-up behavior of Cr and the reduced Al content near the coating/superalloy interface. The local phase equilibrium calculations revealed that the γ′ depletion in the superalloy is primarily attributed to the loss of γ′-forming elements, such as Ta and Ti. Our findings opened up an avenue for studies on the superalloy/coating interdiffusion, contributing to reducing this damaging impact.

    • Uranium as the heaviest naturally occurring element plays important roles in nuclear industries. Hydrogen-caused corrosions and irradiation-caused structural damages are two critical degradations that threaten the safe storage and practical applications of uranium. Through alloying with transition metals like Nb, the gamma-phase of U can be stabilized at room temperature, which shows better performance against hydrogen-caused corrosions than the ground-state alpha-U. The underlying mechanisms have not been fully understood yet. To explain the preferential hydriding phenomenon observed on a specially fabricated double-phase U-2.5 wt % Nb alloy, we perform multiscale ab initio calculations and kinetic Monte Carlo (KMC) simulations. We find that because of different diffusion mechanisms, intrinsic alpha-U and gamma-U already show different hydrogen accumulation behaviors. The existence of random Nb atoms further inhibits hydrogen accumulation in gamma-U. Our work declares its contribution by pointing out the important role of crystal lattice architectures on hydrogen accumulations in metals.

  • How many microplastics do we ingest when using disposable drink cups?

    • Microplastics in food are receiving increasing attention because they are directly ingested by humans and pose a health threat. However, little attention has been paid to the microplastics resulting from the disposable drink cups. Here, we analyzed the microplastic contamination in beverages in three commonly used plastic cups (PP, PET, and PE cups). Effects of the cup materials, the residence time of the drink in the cup, shaking, and water temperatures on the microplastic release were systematically investigated. The results showed that the micro -plastic abundance in water reached 723???1,489 particles cup??? 1 after 5 min exposure and increased significantly with increasing residence time. The size of released microplastics was mainly in the range of < 50 ??m. The shaking applied during transport promoted the release of microplastics from plastic cups. Besides, the results showed that cups released more microplastics when serving hot drinks than serving cold drinks. Based on the results, we estimated that people may unconsciously ingest 37,613???89,294 microplastics a year due to the use of one plastic cup every 4???5 days. Considering the potential harm of microplastics, the contamination of micro -plastics resulting from the use of plastic cups for drinks needs to be taken seriously.

  • Variability in the analysis of a single neuroimaging dataset by many teams

    • Data analysis workflows in many scientific domains have become increasingly complex and flexible. Here we assess the effect of this flexibility on the results of functional magnetic resonance imaging by asking 70 independent teams to analyse the same dataset, testing the same 9 ex-ante hypotheses(1). The flexibility of analytical approaches is exemplified by the fact that no two teams chose identical workflows to analyse the data. This flexibility resulted in sizeable variation in the results of hypothesis tests, even for teams whose statistical maps were highly correlated at intermediate stages of the analysis pipeline. Variation in reported results was related to several aspects of analysis methodology. Notably, a meta-analytical approach that aggregated information across teams yielded a significant consensus in activated regions. Furthermore, prediction markets of researchers in the field revealed an overestimation of the likelihood of significant findings, even by researchers with direct knowledge of the dataset(2-5). Our findings show that analytical flexibility can have substantial effects on scientific conclusions, and identify factors that may be related to variability in the analysis of functional magnetic resonance imaging. The results emphasize the importance of validating and sharing complex analysis workflows, and demonstrate the need for performing and reporting multiple analyses of the same data. Potential approaches that could be used to mitigate issues related to analytical variability are discussed. The results obtained by seventy different teams analysing the same functional magnetic resonance imaging dataset show substantial variation, highlighting the influence of analytical choices and the importance of sharing workflows publicly and performing multiple analyses.

    • Background: Tumor metastasis is one of the leading causes of poor prognosis for colorectal cancer (CRC) patients. Loss of Smad4 contributes to aggression process in many human cancers. However, the underlying precise mechanism of aberrant Smad4 expression in CRC development is still little known. Results: miR-20a-5p negatively regulated Smad4 by directly targeting its 3UTR in human colorectal cancer cells. miR-20a-5p not only promoted CRC cells aggression capacity in vitro and liver metastasis in vivo, but also promoted the epithelial-to-mesenchymal transition process by downregulating Smad4 expression. In addition, tissue microarray analysis obtained from 544 CRC patients clinical characters showed that miR-20a-5p was upregulated in human CRC tissues, especially in the tissues with metastasis. High level of miR-20a-5p predicted poor prognosis in CRC patients. Methods: Five miRNA target prediction programs were applied to identify potential miRNA(s) that target(s) Smad4 in CRC. Luciferase reporter assay and transfection technique were used to validate the correlation between miR-20a-5p and Smad4 in CRC. Wound healing, transwell and tumorigenesis assays were used to explore the function of miR-20a-5p and Smad4 in CRC progression in vitro and in vivo. The association between miR-20a-5p expression and the prognosis of CRC patients was evaluated by Kaplan-Meier analysis and multivariate cox proportional hazard analyses based on tissue microarray data. Conclusions: miR-20a-5p, as an onco-miRNA, promoted the invasion and metastasis ability by suppressing Smad4 expression in CRC cells, and high miR-20a-5p predicted poor prognosis for CRC patients, providing a novel and promising therapeutic target in human colorectal cancer.

    • In this paper, InGaAs p-i-n photodetectors (PDs) on an InP/SiO2/Si (InPOI) substrate fabricated by ion-slicing technology are demonstrated and compared with the identical device on a commercial InP substrate. The quality of epitaxial layers on the InPOI substrate is similar to that on the InP substrate. The photo responsivities of both devices measured at 1.55 µm are comparable, which are about 0.808–0.828 A W−1. Although the dark current of PD on the InPOI substrate is twice as high as that of PD on the InP substrate at 300 K, the peak detectivities of both PDs are comparable. In general, the overall performance of the InPOI-based PD is comparable to the InP-based PD, demonstrating that the ion-slicing technology is a promising route to enable the high-quality Si-based InP platform for the full photonic integration on a Si substrate.

    • We report the effect of cell structures on the fatigue behavior of additively manufactured (AM) 316L stainless steel (316LSS). Compared with the cell-free samples, the fatigue process of fully cellular samples only consists of steady and overload stages, without an initial softening stage. Moreover, the fully cellular sample possesses higher strength, lower cyclic softening rate and longer lifetime. Microscopic analyses show no difference in grain orientations, dimensions, and shapes. However, the fully cellular samples show planar dislocation structures, whereas the cell-free samples display wavy dislocation structures. The existence of cell structures promotes the activation of planar slip, delays strain localization, and ultimately enhances the fatigue performance of AM 316LSS.

    • Anisotropic ductility in additively manufactured (AM) alloys, namely better ductility along the building direction (BD) has been extensively studied and traditionally attributed to the crystallographic texture. However, recent studies have shown significant ductility anisotropy in weakly or non-textured AM alloys, indicating that other factors may also play critical roles. To explore this, AM Inconel 718 with weak crystallographic texture was selected as the model material, and the in-situ high-energy X-ray diffraction tests together with multiscale microstructural characterization techniques were performed to explore the deformation micromechanisms. The results of this study, for the first time, revealed that the better ductility in the vertical specimen (loading parallel to BD) was partially due to the negative stress triaxiality factor (TF) of the {220} grains during plastic deformation, which results in the shrinkage or even healing of the microvoids. Furthermore, the & delta;-phase alignment in conjunction with grain boundary orientation were also proved to have a pronounced impact on the anisotropic ductility of AM alloys. On the other hand, though in the overall weak-textured microstructure, the proportion of 101 grains were marginally over other grains. Thus, the positive effect of {220} grains on ductility was stronger than the negative effect of {200} and {311} grains, contributing to the excellent failure elongation exceeding 12% for both samples. The findings of this study shed new light on the mechanisms underlying the anisotropic ductility of AM alloys and provide insight into strategies for enhancing their performance.

    • We have investigated the low cycle fatigue (LCF) properties and the extent of strengthening in a dense additively manufactured stainless steel containing different volume fractions of cell structures but having all other microstructure characteristics the same. The samples were produced by laser powder bed fusion (L-PBF), and the concentration of cell structures was varied systematically by varying the annealing treatments. Load-controlled fatigue experiments performed on samples with a high fraction of cell structures reveal an up to 23 times increase in fatigue life compared to an essentially cell-free sample of the same grain configuration. Multiscale electron microscopy characterizations reveal that the cell structures serve as the soft barriers to the dislocation propagation and the partials are the main carrier for cyclic loading. The cell structures, stabilized by the segregated atoms and misorientation between the adjacent cells, are retained during the entire plastic deformation, hence, can continuously interact with dislocations, promote the formation of nanotwins, and provide massive 3D network obstacles to the dislocation motion. The compositional micro-segregation caused by the cellular solidification features serves as another non-negligible strengthening mechanism to dislocation motion. Specifically, the cell structures with a high density of dislocation debris also appear to act as dislocation nucleation sites, very much like coherent twin boundaries. This work indicates the potential of additive manufacturing to design energy absorbent alloys with high performance by tailoring the microstructure through the printing process.

    • Background Determination of the role of steroid hormones in expression and regulation of endometrial glucose transport 4 (GLUT4) in humans is important for understanding endometrial disorders such as polycystic ovary syndrome (PCOS), a common hormone-imbalance disease. Methods Endometrial biopsy samples were collected from non-PCOS patients with regular menstrual cycles or with hyperplasia and from PCOS patients with or without hyperplasia. In addition, endometrial tissues from postmenopausal women were incubated with human chorionic gonadotropin (hCG, 10 IU/ml), 17β-estradiol (E2, 10 nM), progesterone (P4, 100 nM), or a combination of E2 and P4 for 24 h. The expression of GLUT4 was measured at the mRNA level using quantitative real-time polymerase chain reaction (qRT-PCR) and at the protein level using Western blot analysis and immunohistochemistry. Results A cyclical change in GLUT4 expression pattern was observed in non-PCOS patients, and a high level of GLUT4 expression was seen in the proliferative phase compared to the secretory phase. Low levels of GLUT4 expression were found in PCOS patients compared to menstrual cycle phase-matched non-PCOS patients, and there was no significant change in GLUT4 expression in PCOS patients during the menstrual cycle. GLUT4 was localized in both epithelial and stromal cells, with notable changes in epithelial cells. We postulate that decreased GLUT4 expression might be regulated by steroid hormones. In support of this, we showed that in cultured endometrial tissues hCG and E2 alone had no effect on GLUT4 expression. However, P4 alone and P4 in combination with E2 decreased GLUT4 expression. Compared with non-PCOS controls, PCOS patients with endometrial hyperplasia exhibited decreased GLUT4 expression in particular in the epithelial cells. Conclusion We conclude that P4 can induce changes in endometrial GLUT4 expression during the menstrual cycle and that abnormal hormonal conditions such as PCOS disrupt normal patterns of GLUT4 expression in endometrial cells.

    • Women with polycystic ovary syndrome (PCOS) are at higher risk for metabolic disorders compared to healthy women, and about 51 % of women with PCOS suffer from non-alcoholic fatty liver disease (NAFLD). Investigation into the pathological mechanism behind this association will provide insights for the prevention and treatment of this complication.Dihydrotestosterone (DHT), a nonaromatic androgen, was used to mimic the pathological conditions of hyperandrogenism and insulin resistance. Hematoxylin and eosin staining, Oil Red O staining, immunofluorescent staining, Western blots, and qRT-PCR were used to verify the hepatic steatosis and inflammation, and the latter two methods were also used for energy and mitochondrion-related assays. ELISA was used to measure the level of reactive oxygen species.Twelve weeks of DHT exposure led to obesity and insulin resistance as well as hepatic steatosis, lipid deposition, and different degrees of inflammation. The expression of molecules involved in respiratory chain and aerobic respiration processes, such as electron transfer complex II, pyruvate dehydrogenase, and succinate dehydrogenase complex subunit A, was inhibited. In addition, molecules associated with apoptosis and autophagy were also abnormally expressed, such as increased Bak mRNA, an increased activated caspase-3 to caspase-3 ratio, and increased Atg12 protein expression. All of these changes are associated with the mitochondria and lead to lipid deposition and inflammation in the liver.Long-term androgen excess contributes to insulin resistance and hepatic steatosis by affecting mitochondrial function and causing an imbalance in apoptosis and autophagy, thus suggesting the pathogenesis of NAFLD in women with PCOS.

    • To identify susceptibility genes for amyotrophic lateral sclerosis (ALS), we conducted a genome-wide association study (GWAS) in 506 individuals with sporadic ALS and 1,859 controls of Han Chinese ancestry. Ninety top SNPs suggested by the current GWAS and 6 SNPs identified by previous GWAS were analyzed in an independent cohort of 706 individuals with ALS and 1,777 controls of Han Chinese ancestry. We discovered two new susceptibility loci for ALS at 1q32 (CAMK1G, rs6703183, P-combined = 2.92 x 10(-8), odds ratio (OR) = 1.31) and 22p11 (CABIN1 and SUSD2, rs8141797, P-combined = 2.35 x 10(-9), OR = 1.52). These two loci explain 12.48% of the overall variance in disease risk in the Han Chinese population. We found no association evidence for the previously reported loci in the Han Chinese population, suggesting genetic heterogeneity of disease susceptibility for ALS between ancestry groups. Our study identifies two new susceptibility loci and suggests new pathogenic mechanisms of ALS.

This work seeks to develop an improved technique for characterizing this brain ultrashort- (Formula presented.) component in vivo accounting for (Formula presented.) weighting. Methods: Data from ultrashort echo time scans from 16 healthy volunteers with variable flip angles (VFA) were collected and fitted into an advanced regression model to quantify signal fraction, relaxation time, and frequency shift of the ultrashort- (Formula presented.) component. Results: The fitted components show intra-subject differences of different white matter structures and significantly elevated ultrashort- (Formula presented.) signal fraction in the corticospinal tracts measured at 0.09 versus 0.06 in other white matter structures and significantly elevated ultrashort- (Formula presented.) frequency shift in the body of the corpus callosum at (Formula presented.) 1.5 versus (Formula presented.) 2.0 ppm in other white matter structures. Conclusion: The significantly different measured components and measured (Formula presented.) relaxation time of the ultrashort- (Formula presented.) component suggest that this method is picking up novel signals from phospholipid membrane-bound protons.

    • As one of the major causes of antimicrobial resistance, beta-lactamase develops rapidly among bacteria. Detection of beta-lactamase in an efficient and low-cost point-of-care testing (POCT) way is urgently needed. However, due to the volatile environmental factors, the quantitative measurement of current POCT is often inaccurate. Herein, we demonstrate an artificial intelligence (AI)-assisted mobile health system that consists of a paper-based beta-lactamase fluorogenic probe analytical device and a smartphone-based AI cloud. An ultrafast broad-spectrum fluorogenic probe (B1) that could respond to beta-lactamase within 20 s was first synthesized, and the detection limit was determined to be 0.13 nmol/L. Meanwhile, a three-dimensional microfluidic paper-based analytical device was fabricated for integration of B1. Also, a smartphone-based AI cloud was developed to correct errors automatically and output results intelligently. This smart system could calibrate the temperature and pH in the beta-lactamase level detection in complex samples and mice infected with various bacteria, which shows the problem-solving ability in interdisciplinary research, and demonstrates potential clinical benefits.

    • Thermal barrier coating (TBC) systems are commonly used in gas turbines for protection against high-temperature degradation. Penetration of the ceramic top coat by corrosive species may cause corrosion damage on the underlying NiCoCrAlY bond coat and cause failure of the TBC system. In the current study, four oxidation/corrosion conditions were tried: (i) lab air, (ii) water vapor, (iii) sodium sulfate deposited on the specimens, and (iv) water vapor + sodium sulfate. The test was done at 750 °C in a cyclic test rig with 48 h cycles. The corrosion damage was studied on NiCoCrAlY-coated specimens, thin APS TBC specimens, and thick APS TBC specimens. Water vapor was found to have very minor influence on the oxidation, while sodium sulfate increased the TGO thickness both for NiCoCrAlY specimens and TBC-coated specimens; the influence of the TBC thickness was found to be very small. Sodium sulfate promoted thicker TGO; more Cr-rich TGO; the formation of Y oxides, and internally, Y sulfides; pore formation in the coating as well as in the substrate; and the formation of a Cr-depleted zone in the substrate.

    • Thermal barrier coatings (TBC) are deposited on superalloys in gas turbines to prolong component life. Electro-discharge machining (EDM) provides a convenient way of machining superalloys prior to coating deposition. Wire-EDM may leave residues from the wire material on the cut surface. A study was performed to investigate if brass residues from the EDM wire affected TBC oxidation behavior and spallation life. Isothermal oxidation at 1050 ℃ and thermal cycling at 1100 ℃ were performed on plasma sprayed TBCs on two different EDM cut substrates. No decrease in TBC life was detected due to the brass residues on the cut surfaces.

It was found that a -depletion based lifecriterion could not be used for the studied coating composition and temperature as it would be tooconservative. A 0-depletion based model was instead suggested and supported by interdiffusion sim-ulation.

    • Coatings from MCrAlY-type alloys are commonly used for oxidation and corrosion protection in gas turbines. As coated components are exposed to high temperature, the coating provides oxidation protection by the formation of an alumina scale, thus depleting the coating of Al which, eventually, will cause the coating to fail. The present study deals with MCrAlY alloy design from a lifing perspective. A previously developed coupled oxidation-diffusion model was used to study the influence of coating composition, substrate composition and oxidation temperature on the expected life of MCrAlY coatings. Eight model coatings, covering the wide range of MCrAlY compositions used industrially, and two model substrates, corresponding to a blade material and a combustor material, were evaluated by the oxidation-diffusion model. Three life criteria were tried: 1) beta-phase-depletion, 2) critical Al content at the coating surface, and 3) a critical TGO thickness. It was shown that the critical TGO thickness was the most conservative life criterion for high-Al coatings on high-Al substrates. For low-Cr and low-Co coatings, the beta-depletion criterion was usually the most conservative. For cases where beta-stability was high (such as at low temperatures and for coatings high in Cr, Co and Al) the critical-Al criterion was often the most conservative.

    • The limited light absorption capacity for most polymer acceptors hinders the improvement of the power conversion efficiency (PCE) of all-polymer solar cells (all-PSCs). Herein, by simultaneously increasing the conjugation of the acceptor unit and enhancing the electron-donating ability of the donor unit, a novel narrow-bandgap polymer acceptor PF3-DTCO based on an A–D–A-structured acceptor unit ITIC16 and a carbon–oxygen (C–O)-bridged donor unit DTCO is developed. The extended conjugation of the acceptor units from IDIC16 to ITIC16 results in a red-shifted absorption spectrum and improved absorption coefficient without significant reduction of the lowest unoccupied molecular orbital energy level. Moreover, in addition to further broadening the absorption spectrum by the enhanced intramolecular charge transfer effect, the introduction of C–O bridges into the donor unit improves the absorption coefficient and electron mobility, as well as optimizes the morphology and molecular order of active layers. As a result, the PF3-DTCO achieves a higher PCE of 10.13% with a higher short-circuit current density (Jsc) of 15.75 mA cm−2 in all-PSCs compared with its original polymer acceptor PF2-DTC (PCE = 8.95% and Jsc = 13.82 mA cm−2). Herein, a promising method is provided to construct high-performance polymer acceptors with excellent optical absorption for efficient all-PSCs.

    • The jiuding Ridge (Sichuan, China) has been explored during the past several years by a team from chengdu Institute of Biology. Chinese Academy of Sciences, and three undescribed species of Corydalis DC. (Fumariaceae) have been revealed and are apparently endemic to this mountain. The new species were observed in forest (C. capitata X.F. Cao, Liden & Y.W. Wang), and alpine rocky limestone areas (C. schistostigma X.F. Cao, Liden, Y.W. Wang & Y.L. Peng). The two first species have their closest relatives in the Wolong-Balangshan District in the Wolong-Balangshan District (Wenchuan. Sichuan). Corydalis aeaeae differs from C. panada Liden & Y.W. Wang in its small size, few-flowered racemes, and broadly obtuse outer petals with low, short erests. Corydalis capitata differs from the C. flexuosa Franchet complex in the capitate racemes, small flowers with deeply serrate petals, and the peculiar, thin, strictly ecect lateral branches. corydalis schistostinma is unique in the C. curviflora Maximowicz ex Hemsley group, in the deeply cleft stigma and very forward-projecting crests to the inner petals, and possibly has iits affinities more to the north and northwest in the Hengduan Mountains.

    • Development of thermal barrier coating systems (TBCs) for gas turbine applications allowing higher combustion temperatures is of high interest since it results in higher fuel efficiency and lower emissions. TBCs produced by suspension plasma spraying (SPS) have been shown to exhibit significantly lower thermal conductivity as compared to conventional systems due to their very fine porosity microstructure. However they have not been commercialised yet due to low reliability and life expectancy of the coatings. In addition to the initial topcoat microstructure and its sintering resistance, lifetime of a TBC system is highly dependent on bondcoat chemistry as it influences the growth rate of thermally grown oxide (TGO) layer. To enhance the lifetime of SPS TBCs, fundamental understanding of relationships between topcoat microstructure and its evolution with time, bondcoat chemistry, TGO growth rate, and lifetime is essential. The objective of this work was to study the effect of topcoat microstructure evolution and TGO growth rate on lifetime in SPS TBC systems. Experimental MCrAlY bondcoat powders with different aluminium activities were investigated and compared to a commercial bondcoat powder. High velocity air fuel spraying was used for bondcoat deposition while axial-SPS was used for yttria stabilized zirconia topcoat deposition. Lifetime was examined by thermal cyclic fatigue testing. Isothermal heat treatment was performed to study TGO evolution with time. The changes in microstructure of SPS coatings due to sintering under long term exposure at high temperatures were investigated. Different failure modes in SPS TBCs were also examined. The bondcoat with higher aluminium activity resulted in a significantly higher thermal cyclic lifetime of the corresponding TBC as it could have promoted protective alumina layer growth for a longer period of time. The results indicate that the significant changes in topcoat microstructure due to sintering as observed in this work could have a detrimental effect on TBC lifetime. © 2017 Elsevier B.V.

    • Growing evidence suggests that epithelial-mesenchymal transition (EMT) and its regulator mitogen-activated protein kinase (MAPK) contribute to endometria-related reproductive disorders. However, the regulation of EMT and MAPK signaling components in the endometrium from polycystic ovary syndrome (PCOS) patients has not been systematically investigated and remains elusive. In humans, how metformin induces molecular alterations in the endometrial tissues under PCOS conditions is not completely clear. Here, we recruited 7 non-PCOS patients during the proliferative phase (nPCOS), 7 non-PCOS patients with endometrial hyperplasia (nPCOSEH), 14 PCOS patients during the proliferative phase (PCOS), and 3 PCOS patients with endometrial hyperplasia (PCOSEH). Our studies demonstrated that compared with nPCOS, PCOS patients showed decreased Claudin 1 and increased Vimentin and Slug proteins. Similar to increased Slug protein, nPCOSEH and PCOSEH patients showed increased N-cadherin protein. Western blot and immunostaining revealed increased epithelial phosphorylated Cytokeratin 8 (p-CK 8) expression and an increased p-CK 8:CK 8 ratio in PCOS, nPCOSEH, and PCOSEH patients compared to nPCOS patients. Although nPCOSEH and PCOSEH patients showed increased p-ERK1/2 and/or p38 protein levels, the significant increase in p-ERK1/2 expression and p-ERK1/2:ERK1/2 ratio was only found in PCOS patients compared to nPCOS patients. A significant induction of the membrane ERβ immunostaining was observed in the epithelial cells of PCOS and PCOSEH patients compared to nPCOS and nPCOSEH patients. While in-vitro treatment with metformin alone increased Snail and decreased Claudin 1, N-cadherin and α-SMA proteins, concomitant treatment with metformin and E2 increased the expression of CK 8 and Snail proteins and decreased the expression of Claudin 1, ZO-1, Slug and α-SMA proteins. Our findings suggest that the EMT contributes to the switch from a healthy state to a PCOS state in the endometrium, which might subsequently drive endometrial injury and dysfunction. We also provide evidence that metformin differentially modulates EMT protein expression in PCOS patients depending on estrogenic stimulation.

    • Low-grade chronic inflammation is commonly seen in polycystic ovary syndrome (PCOS) patients with elevated levels of inflammatory cytokines in the endometrium. However, our understanding of the mechanisms underlying cytokine synthesis and increased endometrial inflammation in PCOS patients remains limited.Endometrial biopsy samples were collected from non-PCOS (n = 17) and PCOS (n = 22) patients either during the proliferative phase of the menstrual cycle or with hyperplasia. Endometrial explants were prepared from PCOS patients and subjected to pharmacological manipulation in vitro. The expression and localization of TLR2/4, key elements of innate immune signal transduction and NFκB signaling pathways, and multiple cytokines were comprehensively evaluated by Western blotting, immunohistochemistry, and immunofluorescence in endometrial tissues.We demonstrated the distribution of protein expression and localization associated with the significantly increased androgen receptor, TLR2, and TLR4-mediated activation of IRF-7 and NFkB signaling, cytokine production, and endometrial inflammation in PCOS patients compared to non-PCOS patients with and without endometrial hyperplasia. In vitro experiments showed that 5α-dihydrotestosterone (DHT) enhanced androgen receptor, TLR4, IRF-7, and p-NFκB p65 protein expression along with increased IFNα and IFNɣ abundance. The effects of DHT on IRF-7, p-NFκB p65, and IFN abundance were abolished by flutamide, an anti-androgen. Although 17β-estradiol (E2) decreased p-IRF-7 expression with little effect on TLR-mediated IRF7 and NFκB signaling or on cytokine protein levels, exposure to metformin alone or in combination with E2 suppressed IRAK4, p-IRF-7, IRF-7, IKKα, p-NFκB p65, IFNɣ, and TNFα protein expression.Cytokine synthesis and increased endometrial inflammation in PCOS patients is coupled to androgen-induced TLR4/IRF-7/NFkB signaling, which is be inhibited by metformin treatment.

    • Impaired progesterone (P4) signaling is linked to endometrial dysfunction and infertility in women with polycystic ovary syndrome (PCOS). Here, we report for the first time that elevated expression of progesterone receptor (PGR) isoforms A and B parallels increased estrogen receptor (ER) expression in PCOS-like rat uteri. The aberrant PGR-targeted gene expression in PCOS-like rats before and after implantation overlaps with dysregulated expression of Fkbp52 and Ncoa2, two genes that contribute to the development of uterine P4 resistance. In vivo and in vitro studies of the effects of metformin on the regulation of the uterine P4 signaling pathway under PCOS conditions showed that metformin directly inhibits the expression of PGR and ER along with the regulation of several genes that are targeted dependently or independently of PGR-mediated uterine implantation. Functionally, metformin treatment corrected the abnormal expression of cell-specific PGR and ER and some PGR-target genes in PCOS-like rats with implantation. Additionally, we documented how metformin contributes to the regulation of the PGR-associated MAPK/ERK/p38 signaling pathway in the PCOS-like rat uterus. Our data provide novel insights into how metformin therapy regulates uterine P4 signaling molecules under PCOS conditions.

  • Genetic variants and Expression of Cytochrome p450 Oxidoreductase Predict Postoperative Survival in Patients with Hepatitis B Virus-Related Hepatocellular Carcinoma

    • Our current study investigates the prognostic values of genetic variants and mRNA expression of cytochrome p450 oxidoreductase (POR) in hepatitis B virus (HBV)-related hepatocellular carcinoma (HCC). A total of 19 candidate single nucleotide polymorphisms (SNPs) located in the exons of POR were genotyped using Sanger sequencing from 476 HBV-related HCC patients who underwent hepatectomy between 2003 and 2013. The mRNA expression of POR in 212 patients with HBV-related HCC was obtained from GSE14520 dataset. Survival analysis was performed to investigate the association of POR variants and mRNA expression with overall survival (OS) and recurrence-free survival (RFS). Nomograms were used to predict the prognosis of HBV-related HCC patients. Gene set enrichment analysis (GSEA) was used to investigate the mechanism of POR in HBV-related HCC prognosis. The polymorphism POR-rs1057868 was significantly associated with HBV-related HCC OS (CT/TT vs. CC, hazard ratio [HR] = 0.69, 95% confidence interval [CI] = [0.54, 0.88], P = 0.003), but not significantly associated with RFS (CT/TT vs. CC, P = 0.378). POR mRNA expression was also significantly associated with HBV-related HCC OS (high vs. low, HR = 0.61, 95% CI = [0.38, 0.97], P= 0.036), but not significantly associated with the RFS (high vs. low, P = 0.201). Two nomograms were developed to predict the HBV-related HCC OS. Furthermore, GSEA suggests that multiple gene sets were significantly enriched in liver cancer survival and recurrence, as well as POR-related target therapy in the liver. In conclusion, our study suggests that POR-rs1057868 and mRNA expression may serve as a potential postoperative prognosis biomarker in HBV-related HCC.

    • PURPOSE: Available evidence indicates that dipyridamole enhances the anti-thrombotic effects of aspirin for the prevention of secondary strokes. Aspirin is a well-known non-steroid anti-inflammatory drug. This anti-inflammatory property has turned aspirin into a potential drug for inflammation-related cancers such as colorectal cancer (CRC). Here, we aimed to explore whether the anti-cancer effect of aspirin against CRC could be improved by combined administration with dipyridamole.METHODS: Population-based clinical data analysis was conducted to assess a possible therapeutic effect of combined dipyridamole and aspirin treatment in inhibiting CRC compared with either monotherapy. This therapeutic effect was further verified in different CRC mouse models, i.e. an orthotopic xenograft mouse model, an AOM/DSS mouse model, an Apc min/+ mouse model and a patient derived xenograft (PDX) mouse model. The in vitro effects of the drugs on CRC cells were tested using CCK8 and flow cytometry assays. RNA-Seq, Western blotting, qRT-PCR and flow cytometry were used to identify the underlying molecular mechanisms. RESULTS: We found that dipyridamole combined with aspirin had a better inhibitory effect on CRC than either monotherapy alone. The enhanced anti-cancer effect of the combined use of dipyridamole with aspirin was found to rely on the induction of an overwhelmed endoplasmic reticulum (ER) stress and subsequent pro-apoptotic unfolded protein response (UPR), which was different from the anti-platelet effect.CONCLUSIONS: Our data indicate that the anti-cancer effect of aspirin against CRC may be enhanced by combined administration with dipyridamole. In case further clinical studies confirm our findings, these may be repurposed as adjuvant agents.

  • To which world regions does the valence-dominance model of social perception apply?

    • Over the past 10 years, Oosterhof and Todorov's valence-dominance model has emerged as the most prominent account of how people evaluate faces on social dimensions. In this model, two dimensions (valence and dominance) underpin social judgements of faces. Because this model has primarily been developed and tested in Western regions, it is unclear whether these findings apply to other regions. We addressed this question by replicating Oosterhof and Todorov's methodology across 11 world regions, 41 countries and 11,570 participants. When we used Oosterhof and Todorov's original analysis strategy, the valence-dominance model generalized across regions. When we used an alternative methodology to allow for correlated dimensions, we observed much less generalization. Collectively, these results suggest that, while the valence-dominance model generalizes very well across regions when dimensions are forced to be orthogonal, regional differences are revealed when we use different extraction methods and correlate and rotate the dimension reduction solution.     • This work concerns the study of damage evolution in a newly developed high purity nano 8YSZ thermal barrier coating during thermal cyclic fatigue tests (TCF). TCF tests were conducted between 100 degrees C-1100 degrees C with a hold time of 1 hat 1100 degrees C, first till failure and later for interrupted tests. Cross section analysis along the diameter of the interrupted test samples revealed a mixed-type failure and that the most of the damage occurred towards the end of the coatings life. To understand the most likely crack growth mechanism leading to failure, different crack growth paths have been modelled using finite element analysis. Crack growing from an existing defect in the top coat towards the top coat/TGO interface has been identified as the most likely mechanism. Estimated damage by the model could predict the rapid increase in the damage towards the end of the coatings life. (C) 2017 Elsevier Ltd. All rights reserved.

    • Thermal barrier coatings (TBCs), consisting of a ceramic top coat and a metallic bond coat, offer resistance against high temperature degradation of turbine components. Cyclic oxidation of the bond coat, thermal stresses due to their thermal mismatches during cyclic operations, and sintering of the top coat are considered to be the common ways by which thermal barrier coatings fail. To reduce sintering, a nano structured high purity yttria stabilized zirconia (YSZ) was developed. The focus of this work is to compare the damage development of such high purity nano YSZ TBC during thermal cycling with a conventional YSZ TBC. Thermal cyclic fatigue (TCF) tests were conducted on both the TBC systems between 100 °C and 1100 °C with a 1 h hold time at 1100 °C. TCF test results showed that conventional YSZ TBC exhibited much higher life compared to the high purity nano YSZ TBC. The difference in the lifetime is explained by the use of microstructural investigations, crack length measurements along the cross-section and the difference in the elastic modulus. Furthermore, stress intensity factors were calculated in order to understand the difference(s) in the damage development between the two TBC systems.

    • Thermal Barrier Coatings (TBCs) are advanced material systems that are being used in the hot sections of gas turbines such as combustor, turbine blades, and vanes. The top ceramic coating in TBCs provides insulation against the hot gases and the intermediate metallic bond coat provides oxidation and corrosion resistance to the underlying turbine components.Durability of thermal barrier coatings is very important for the overall performance of the gas turbine. TBCs can fail in several different ways and there is a combination of more than one failure mechanism in most situations. One of the most widely used TBC is atmospheric plasma sprayed (APS) yttria stabilized zirconia (YSZ). Both the deposition technique and the TBC material have certain limitations. The main aim of this research is to study new TBC materials and/or new deposition techniques and compare with the conventional YSZ and understand their failure mechanisms during cyclic oxidation and hot corrosion.Thermal cyclic oxidation of a newly developed high purity nano YSZ thermal barrier coating has been studied. Cross sectional analysis of exposed as well as completely failed samples showed a mixed-type failure caused by crack propagation parallel to the bond coat/top coat interface. The majority of the damage occurred towards the end of the coating life. A finite element model has been developed to study the probability of crack growth along different paths that leads to the final failure.Hot corrosion mechanism in suspension plasma sprayed two-layer gadolinium zirconate/YSZ, three-layer dense gadolinium zirconate/gadolinium zirconate/YSZ, and a single-layer YSZ has been studied in the presence of sodium sulfate and vanadium pentoxide. The test results showed that gadolinium zirconate coatings were more susceptible to corrosion compared to YSZ coatings despite gadolinium zirconate coatings having lower reactivity with the corrosive salts.Thermal cycling behavior of a high chromium bond coat has been studied. Cross-sectional analysis showed formation of sandwich type microstructure with chromium rich oxide and alumina as the top and the bottom layers.Inter-diffusion of minor elements between different MCrAlY coatings – substrate systems has been studied using, diffusion simulation software, DICTRA. The simulation results showed that the diffusion of minor elements in the coatings is dependent on the rate of β phase depletion in the beginning. After the depletion of β phase there was no clear dependence of the coating composition on the diffusion of minor elements.

    • The current investigation focuses on understanding the influence of a columnar microstructure and a sealing layer on the corrosion behavior of suspension plasma sprayed thermal barrier coatings (TBCs). Two different TBC systems were studied in this work. First is a double layer made of a composite of gadolinium zirconate + yttria stabilized zirconia (YSZ) deposited on top of YSZ. Second is a triple layer made of dense gadolinium zirconate deposited on top of gadolinium zirconate + YSZ over YSZ. Cyclic corrosion tests were conducted between 25 and 900 °C with an exposure time of 8 h at 900 °C. 75 wt.% Na2SO4 + 25 wt.% NaCl were used as the corrosive salts at a concentration of 6 mg/cm2. Scanning electron microscopy analysis of the samples’ cross sections showed that severe bond coat degradation had taken place for both the TBC systems, and the extent of bond coat degradation was relatively higher in the triple-layer system. It is believed that the sealing layer in the triple-layer system reduced the number of infiltration channels for the molten salts which resulted in overflowing of the salts to the sample edges and caused damage to develop relatively more from the edge.

    • Models that can predict the life of thermal barrier coatings (TBCs) during thermal cycling fatigue (TCF) tests are highly desirable. The present work focuses on developing and validating a simplified model based on the relation between the energy release rate and the TCF cycles to failure. The model accounts for stresses due to thermal mismatch, influence of sintering, and the growth of TGO (alumina and other non-protective oxides). The experimental investigation of TBCs included; 1) TCF tests at maximum temperatures of 1050 °C, 1100 °C, 1150 °C and a minimum temperature of 100 °C with 1 h and 5 h (1100 °C) hold times. 2) Isothermal oxidation tests at 900, 1000 and 1100 °C for times up to 8000 h. The model was calibrated and validated with the experimental results. It has been shown that the model is able to predict the TCF life and effect of hold time with good accuracy.

    • This study investigates the corrosion resistance of double layer Gd2Zr2O7/YSZ, triple layer dense Gd2Zr2O7 / Gd2Zr2O7/YSZ and a reference single layer YSZ coating with a similar overall top coat thickness of 300-320 ÎŒm. All the coatings were manufactured by suspension plasma spraying (SPS), resulting in a columnar structure. Corrosion tests were conducted at 900°C for 8 hours using vanadium pentoxide and sodium sulphate as corrosive salts at a concentration of 4 mg/cm2. SEM investigations after the corrosion tests show that Gd2Zr2O7 coatings exhibited lower corrosion resistance than the reference material, YSZ. Reaction between the corrosive salts and Gd2Zr2O7 results in the formation of gadolinium vanadate ( GdVO4) along the top surface and between the columns. While the stresses due to phase transformation of zirconia can be relieved to some extent by realigning of the columns in the top coat, it is believed that GdVO4 formation between the columns, along with low fracture toughness of Gd2Zr2O7 had resulted in lower corrosion resistance. Furthermore, the presence of a relatively dense layer of Gd2Zr2O7 on the top, as a preventive layer for salt infiltration, did not improve the corrosion resistance.
