    • Congenital heart block (CHB) may develop in fetuses of Ro/SSA autoantibody-positive women. Given the rarity of CHB, information on comorbidity and complications later in life is difficult to systematically collect for large groups of patients. We therefore used nation-wide healthcare registers to investigate comorbidity and outcomes in patients with CHB and their siblings.MethodsData from patients with CHB (n= 119) and their siblings (n= 128), all born to anti-Ro/SSA-positive mothers, and from matched healthy controls (n= 1,190) and their siblings (n= 1,071), were retrieved from the Swedish National Patient Register. Analyses were performed by Cox proportional hazard modelling.ResultsIndividuals with CHB had a significantly increased risk of cardiovascular comorbidity, with cardiomyopathy and/or heart failure observed in 20 (16.8%) patients versus 3 (0.3%) controls, yielding a HR of 70.0 (95% CI 20.8 to 235.4), and with a HR for cerebral infarction of 39.9 (95% CI 4.5 to 357.3). Patients with CHB also had a higher risk of infections. Pacemaker treatment was associated with a decreased risk of cerebral infarction but increased risks of cardiomyopathy/heart failure and infection. The risk of systemic connective tissue disorder was also increased in patients with CHB (HR 11.8, 95% CI 4.0 to 11.8), and both patients with CHB and their siblings had an increased risk to develop any of 15 common autoimmune conditions (HR 5.7, 95% CI 2.83 to 11.69 and 3.6, 95% CI 1.7 to 8.0, respectively).ConclusionsThe data indicate an increased risk of several cardiovascular, infectious and autoimmune diseases in patients with CHB, with the latter risk shared by their siblings.

  • Multiplexing DNA methylation markers to detect circulating cell-free DNA derived from human pancreatic β cells

    • It has been proposed that unmethylated insulin promoter fragments in plasma derive exclusively from β cells, reflect their recent demise, and can be used to assess β cell damage in type 1 diabetes. Herein we describe an ultrasensitive assay for detection of a β cell–specific DNA methylation signature, by simultaneous assessment of 6 DNA methylation markers, that identifies β cell DNA in mixtures containing as little as 0.03% β cell DNA (less than 1 β cell genome equivalent). Based on this assay, plasma from nondiabetic individuals (N = 218, aged 4–78 years) contained on average only 1 β cell genome equivalent/mL. As expected, cell-free DNA (cfDNA) from β cells was significantly elevated in islet transplant recipients shortly after transplantation. We also detected β cell cfDNA in a patient with KATP congenital hyperinsulinism, in which substantial β cell turnover is thought to occur. Strikingly, in contrast to previous reports, we observed no elevation of β cell–derived cfDNA in autoantibody-positive subjects at risk for type 1 diabetes (N = 32), individuals with recent-onset type 1 diabetes (<4 months, N = 92), or those with long-standing disease (>4 months, N = 38). We discuss the utility of sensitive β cell cfDNA analysis and potential explanations for the lack of a β cell cfDNA signal in type 1 diabetes.

    • The legal regime applicable to groups of companies in the European Union has been discussed for many years. National legislations have been adopted in a certain number of Member States, and new initiatives are being considered by the European Commission and in academic writing. The central issues in groups of companies is the relationship between the controlling shareholder, often the parent company and the subsidiaries, and the potential for abuse to the detriment of the latter's minority shareholders and creditors. Several answers have been formulated, ranging from a duty of the parent to indemnify the subsidiary for the charges imposed by the parent, to the acceptance of these charges provided they result in some benefit to the subsidiary and do not endanger the subsidiary's solvency. In another approach, these issues may be solved by other common company law, e.g., on the basis of the unfair prejudice provisions. With respect to shareholder and creditor protection, a comparative analysis concludes that there is no need for additional regulatory safeguards. The present approaches indicate that group relations are often characterised by conflicts of interest. Therefore, it is proposed to develop a standard for dealing with these, especially under the form of related party transactions. The specific conditions for dealing with intragroup related party transactions are submitted for further discussion.

    • Currently, the Council of the European Union is negotiating the European Commission's recent proposal on cross-border mobility. This paper provides an overall assessment based on the proposal's central pillars: freedom of establishment and protection of the interests of creditors, shareholders, and employees. The proposed directive meets a real necessity for regulation on a European level and pursues an ambitious agenda. While the general approach is excellent, there is room for improvement on some issues of importance.

    • Neonatal lupus erythematosus (NLE) develops in foetuses of mothers with Ro/SSA and La/SSB antibodies and may include foetal atrioventricular block and dermatologic manifestations. In this study, we investigated postnatal Ro and La IgG, IgA and IgM antibody levels up to 1 year of age in 32 children born to Ro/SSA positive mothers. Antibody levels were correlated with NLE manifestations, and the role of breast feeding in transfer of autoantibodies from mother to child was evaluated. Ro52, Ro60 and La IgG antibodies all transferred from the mothers to their foetus in utero and were present in the infant at birth as detected by enzyme-linked immunosorbent assay using recombinant antigens and a synthetic peptide. A significant decrease in Ro52, Ro60 and La IgG autoantibody levels of the infants was observed from birth to 4–5 weeks of age ( P < 0.05, P < 0.05 and P < 0.01). Ro- and La-specific IgA and IgM antibodies were detected in the serum from a subset of mothers. However, Ro- and La-specific IgA and IgM antibody levels were low or non-detectable in children raised both with and without breastfeeding. Furthermore, NLE skin lesions developed independently of breastfeeding. Our findings support a role for placental materno-foetal transfer of IgG autoantibodies in the pathogenesis of NLE and indicate that refraining from breastfeeding does not protect from NLE skin involvement.

    • Adeno-associated virus (AAV) vectors have shown remarkable efficiency for gene delivery to cultured cells and in animal models of human disease. However, limitations to AAV vectored gene transfer exist after intravenous transfer, including off-target gene delivery (e.g., liver) and low transduction of target tissue. Here, we show that during production, a fraction of AAV vectors are associated with microvesicles/exosomes, termed vexosomes (vector-exosomes). AAV capsids associated with the surface and in the interior of microvesicles were visualized using electron microscopy. In cultured cells, vexosomes outperformed conventionally purified AAV vectors in transduction efficiency. We found that purified vexosomes were more resistant to a neutralizing anti-AAV antibody compared to conventionally purified AAV. Finally, we show that vexosomes bound to magnetic beads can be attracted to a magnetized area in cultured cells. Vexosomes represent a unique entity which offers a promising strategy to improve gene delivery.

Humans are continuously exposed to low levels of beta-carbolines through cooked food, coffee, alcoholic beverages and tobacco smoke. beta-Carbolines have previously been detected in higher levels in the pigmented substantia nigra than in the cortex of humans. The distribution of H-3-labelled harman and norharman in the brain of pigmented and albino mice and in frogs (a species having neuromelanin) was studied by tape-section and light-microscopic autoradiography. Furthermore, the binding of these beta-carbolines to dopamine-melanin and melanin granules from Sepia officinalis was examined. The results revealed a high affinity binding to melanin and a long-term retention (up to 30 days) in pigmented tissues, including neuromelanin-containing neurons of frogs after a single injection. The role of long-term exposure to food-related beta-carbolines and a retention of these compounds in pigment-containing neurons in the induction of idiopathic Parkinson's disease should be further considered.

    • The Swedish OCTO and NONA immune longitudinal studies were able to identify and confirm an immune risk profile (IRP) predictive of an increased 2-year mortality in very old individuals, 86–94 years of age. The IRP, was associated with persistent cytomegalovirus infection and characterized by inverted CD4/CD8 ratio and related to expansion of terminally differentiated effector memory T cells (TEMRA phenotype). In the present HEXA immune longitudinal study, we have examined a younger group of elderly individuals (n = 424, 66 years of age) in a population-based sample in the community of Jönköping, Sweden, to examine the relevance of findings previously demonstrated in the very old. Immunological monitoring that was conducted included T cell subsets and CMV-IgG and CMV-IgM serology. The result showed a prevalence of 15 % of individuals with an inverted CD4/CD8 ratio, which was associated with seropositivity to cytomegalovirus and increases in the level of TEMRA cells. The proportion of individuals with an inverted CD4/CD8 ratio was significantly higher in men whereas the numbers of CD3+CD4+ cells were significantly higher in women. In conclusion, these findings are very similar to those previously found by us in the Swedish longitudinal studies, suggesting that an immune profile previously identified in the very old also exists in the present sample of hexagenerians. Therefore, it will be important to examine clinical parameters, including morbidity and mortality, to assess whether the immune profile also is a risk profile associated with higher mortality in this sample of hexagenerians.

    • This paper presents the cultural and archaeological context of the human fossil bones from Muierii Cave, dated by us to the age of 30 150 ± 800 14C years BP (Before Present) or 34 810 ± 927 cal years BP (calibrated years Before Present), and from Cioclovina Cave, dated to the age of 29 000 ± 700 14C years BP or 33 540 ± 832 cal years BP, in the Southern Carpathians. These are among the most ancient dated human fossil remains from Central and South-Eastern Europe and are described in conjunction with other sites with Mousterian assemblages of the recent Neanderthal population, and sites with Aurignacian assemblage of early modern humans, from Romanian region, for the interval of time 34,000–26,000, the transitional period from the Middle Paleolithic to the Upper Paleolithic.

    • Eco neighborhoods with green infrastructure solutions are increasingly being prescribed today in Swedish cities for reducing and attenuating storm water runoff, increasing biodiversity, having a temperature moderating effect and for energy saving. Thus, contractors are simply required to build with green gardens on concrete decks, such as green roofs, green terraces, green courtyards and green complete neighborhoods. A lot of knowledge and experience is in fact lacking today, and consequences may therefore be devastating. If green system solutions are to be seen as an obvious choice in future settlements, and not as a problem, there must be clear guidelines and specifications that ensure a sustainable outcome. This is missing today. This paper reports a project aiming at bringing together researchers, government and industry to the collaborative development of new and attractive solutions for green roof gardens with consideration to the environment and high requirements for durability, materials, construction and energy efficiency. These solutions must also be adaptable to similar types of facilities, specific needs and environments. One such area concerns public land such as parks, streets and squares on concrete decks. Certification and tailored guidelines for different types of systems are being developed. The initiating part of this project clearly indicates that there is a need for better understanding, more research and long term monitoring/follow up of green roofs. Furthermore, a holistic approach is introduced to ensure that one good green roof function will not have severe negative effects on other functions.

    • A comprehensive performance evaluation of a cross-layer solution to increase users' downlink data rates over HSDPA is provided. The solution consists of a proxy entity between a server and the Radio Network Controller, and cross-layer signalling from the base station to the proxy. The performance of the solution is evaluated though a detailed ns-2 simulator environment, which includes all HSDPA features, as well as some existing TCP enhancing protocols widely adopted for internet traffic over wireless links. Numerical results show that the proxy significantly increases the users' throughput, while also improving the utilization of the radio resources.

    • This study explores the emergence and expansion of data network effects (DNEs) in AI platforms. Previous research has focused on direct and indirect network effects. However, the rise of AI platforms necessitates understanding DNEs for platforms’ learning and improvement. Through a longitudinal case study of a Conversational AI (CAI) platform's 12-year evolution, the study identifies generative feedback loops as the mechanism for DNEs. These loops are initiated by adding functions that enhance the platform's generative capacity, resulting in more diverse data that improves platform learning. DNEs develop through interactions with different ecosystem actors, including clients and external developers, and rely on various data sources beyond user data to enhance AI platform capabilities. This study contributes to IS literature, specifically digital platform literature, following recent calls to empirically examine DNEs to better understand how AI platforms grow and improve their algorithmic capabilities over time.

    • The formation of acrylamide in potato crisps was fitted by empirical mathematical models. Potato slices were fried under the same experimental conditions for different times. Besides the content of precursors in the raw potato slices, acrylamide and water content in the potato crisps were quantified after predetermined times (2-6 min). The temperature developments in the surrounding oil and outer cell layer of the potato slices were monitored, giving more insight in the frying process and making future comparisons between studies possible. The pattern found for the formation of acrylamide, which was similar to earlier studies, was fitted to three empirical models. Statistical methods were used to compare the performance of the models, with the "Logistic-Exponential" and "Empirical" model performing equally well. The obtained model parameters were in the range of earlier reported studies, although this comparison is not unequivocal as the experimental conditions differed between studies. The precision of parameter estimates was problematic; this should be improved by better experimental design. Nevertheless, the approach of this study will make it possible to truly compare acrylamide formation patterns and model parameters in the future, with the ability to develop a tool to predict acrylamide formation in potato crisps.

    • This letter investigates relationships between iterated filtering algorithms based on statistical linearization, such as the iterated unscented Kalman filter (IUKF), and filtering algorithms based on quasi-Newton (QN) methods, such as the QN iterated extended Kalman filter (QN-IEKF). Firstly, it is shown that the IUKF and the iterated posterior linearization filter (IPLF) can be viewed as QN algorithms, by finding a Hessian correction in the QN-IEKF such that the IPLF iterate updates are identical to that of the QN-IEKF. Secondly, it is shown that the IPLF/IUKF update can be rewritten such that it is approximately identical to the QN-IEKF, albeit for an additional correction term. This enables a richer understanding of the properties of iterated filtering algorithms based on statistical linearization.

  • Levels of C-14 in the terrestrial environment in the vicinity of two European nuclear power plants

    • Radiocarbon is produced in all types of nuclear reactors. Most of the C-14 released into the environment is in the form of gaseous emissions. Recent data on the C-14 concentration found in terrestrial samples taken in the vicinity of nuclear power plants in Romania and Lithuania are presented. We found increased C-14 levels in the surroundings of both power plants. At the Romanian power plant Cernavoda, we found excess levels of C-14 in grass within a distance of about 1000 in, the highest C-14 specific activity being 311 Bq/kg C (approximately 28% above the contemporary C-14 background) found at a distance of 200 in from the point of release (nearest sampling location). At the Lithuanian power plant Ignalina, samples of willow, pine, and spruce showed a C-14 excess of similar magnitude, while significantly higher values were found in moss samples. The samples were analyzed at the accelerator mass spectrometry facility in Lund, Sweden.

    • Diagnostic platforms providing biomarkers that are highly predictive for diagnosing, monitoring, and stratifying cancer patients are key instruments in the development of personalized medicine. We demonstrate that tumor cells transfer (mutant) RNA into blood platelets in vitro and in vivo, and show that blood platelets isolated from glioma and prostate cancer patients contain the cancer-associated RNA biomarkers EGFRvIII and PCA3, respectively. In addition, gene-expression profiling revealed a distinct RNA signature in platelets from glioma patients compared with normal control subjects. Because platelets are easily accessible and isolated, they may form an attractive platform for the companion diagnostics of cancer.

    • A set of fossil bones from Romania has been analysed by accelerator mass spectrometry and by neutron activation analysis in order to estimate their age. The temporal attributing of Malu Rosu archaeological settlement has been extensively analyzed. The radiocarbon age, determined by accelerator mass spectrometry, for this site is 5510+/-200 BP. This is in agreement with the age of 6000+/-2000 BP, obtained by the dating method based on fluorine content, determined by neutron activation analysis.

    • Pig meat shows natural variations in the concentrations of precursors of heterocyclic amines (HCAs), which may affect formation of HCAs in cooked pig meat. To study this, 26 pigs with an inherent genetic variation (carriers and noncarriers of the RN- allele) were subjected to different feeding regimes (conventional feed compared with feed composed according to organic standards). In addition, the effect of sex (castrated males or females) was considered when assessing chemical and technological meat quality parameters. Concentrations of precursors of HCAs, i.e., creatine, residual glycogen, dipeptides, and free amino acids, were analyzed in the raw meat, and the levels of some HCAs (4,8-DiMelQx, MelQx, PhIP, harman, and norharman) were then determined in fried meat patties prepared from these pigs. The FIN genotype most affected technological meat quality parameters and the level of precursors of HCAs, especially the level of residual glycogen, where carriers of the RN- allele showed levels four times as high as those of noncarriers (75.3 +/- 2.6 compared with 17.2 +/- 2.4,mumol/g meat, least-squares means +/- SE). The increased level of residual glycogen resulted in about 50% lower amounts of total mutagenic HCAs in cooked meat compared with cooked meat from normal pigs. Fried meat from carriers of the RN- allele obtained darker crust color than meat from noncarriers. Feeding regime and sex did not significantly affect the chemical composition of the meat or the formation of HCAs.

    • The current view of type 1 diabetes (T1D) is that it is an immune-mediated disease where lymphocytes infiltrate the pancreatic islets, promote killing of beta cells and cause overt diabetes. Although tissue resident immune cells have been demonstrated in several organs, the composition of lymphocytes in human healthy pancreatic islets have been scarcely studied. Here we aimed to investigate the phenotype of immune cells associated with human islets of non-diabetic organ donors. A flow cytometry analysis of isolated islets from perfused pancreases (n = 38) was employed to identify alpha, beta, T, natural killer (NK) and B cells. Moreover, the expression of insulin and glucagon transcripts was evaluated by RNA sequencing. Up to 80% of the lymphocytes were CD3(+) T cells with a remarkable bias towards CD8(+) cells. Central memory and effector memory phenotypes dominated within the CD8(+) and CD4(+) T cells and most CD8(+) T cells were positive for CD69 and up to 50-70% for CD103, both markers of resident memory cells. The frequency of B and NK cells was low in most islet preparations (12 and 3% of CD45(+) cells, respectively), and the frequency of alpha and beta cells varied between donors and correlated clearly with insulin and glucagon mRNA expression. In conclusion, we demonstrated the predominance of canonical tissue resident memory CD8(+) T cells associated with human islets. We believe that these results are important to understand more clearly the immunobiology of human islets and the disease-related phenotypes observed in diabetes.

  • Analysis of heterocyclic amines in food products: interlaboratory studies

    • A feasibility study and two interlaboratory exercises on the determination of selected heterocyclic amines (HAs) in beef extract, organised in the framework of a European project, are presented. The aim of these exercises was to improve the quality of the laboratories and to evaluate the performance of a standardised analytical method and also the methods currently used by each of the participants for the analysis of these compounds. Three lyophilised portions of a commercial beef material previously spiked with HAs at different concentration levels ranging from 10 to 75 ng g(-1) were used as laboratory reference materials (lot A, B and C). Firstly, a feasibility study was carried out using a test standard solution and the beef extract (lot A), which contained only five HAs. Then, two interlaboratory exercises were carried out using the laboratory reference materials lot B and lot C, containing 10 selected HAs at two different concentration levels, 75 and 10ng/g, respectively. The results obtained by all participant laboratories using the proposed method showed satisfactory agreement and the CV(%) between-laboratories obtained were from 8.3 to 24.1% for lot B and from 8.7 to 44.5% for lot C. The standardised method evaluated in these collaborative studies is therefore proposed for the analysis of HAs in food material. Moreover, LC-MS is recommended as the most suitable technique for the analysis of a large number of HAs in food samples. (C) 2003 Elsevier B.V All rights reserved.

    • The guiding of highly charged ions through nanocapillaries in different insulating materials, such as polyethylene terephthalate, SiO2, and Al2O3 has been investigated by our group, using 7 keV Ne7+ ions. We find transmission of ions incident at angles larger than the angle given by the capillary aspect ratio in all these materials. The measured angular distributions, however, vary with the membrane material. In this report we compare the experimental findings with the different membranes.

  • Digital innovation diffusion : a case study of an X-reality innovation diffusion

    • Digital innovation (DI) has enabled businesses to enhance their existing market offerings by integrating digital features. Despite advanced technologies, substantial marketing efforts, and global recognition, businesses can still struggle to convince customers to adopt their digital market offerings. This process of spreading novel innovation is known as diffusion. In the fast-growing digital world, due to the unique characteristics of DI, traditional diffusion theories and models show limited explanatory power, creating challenges for researchers and practitioners alike. With the aim of exploring these challenges, we position our research within the IS literature with the following research question:"How and why do diffusion enablers and barriers emerge during digital innovation?". We conducted an interpretive case study of Company X, one of the world's largest consulting firms and an active DI practitioner. Our findings suggest that digital innovation diffusion can be enabled or hindered by several understudied interdependencies in its technological architecture. Furthermore, for successful diffusion, how DI distributes the division of labor between actors and subsequent layers must be effectively embedded and aligned for value in the clients' context in order to diffuse successfully. This study provides novel insights and compelling research avenues.

From our analysis of the Spotify music streaming service, we have identified three strategic objectives that service platform providers need to pursue as they establish and scale their services. Achieving each objective will require trade-offs, and we described the tactics Spotify used to manage these trade-offs. We conclude by providing recommendations on how other service platform providers can apply these tactics

    • Cluster evolution research suggests that maintaining an optimal technological heterogeneity that is exploitable by cluster actors is key to sustainable cluster development. This paper argues that exploring this optimal span and its influence on local synergy creation calls for understanding the interaction between cluster actions, local conditions for collaboration, and heterogeneity requirements over time. For this purpose, a longitudinal case study is conducted, tracing the development of a digital creative cluster that has experienced the initiation, rise, and decline of local technological heterogeneity exploitation. By applying institutional logics as a sensitising device, the analysis explores how actors interact with local and theme structures in this process. Findings show how hub-firms draw on creative norms and technologies to produce situated heterogeneity requirements. These are assessed with co-location factors and accumulated experience of local collaboration to produce local organising rationales that guides decisions to engage in local collaboration.

    • In order to face the challenges derived from an increasingly competitive and disruptive environment, firms often engage in collaborative arrangements with other firms. While it is argued that inter-firm networks can serve as a way to catalyze innovation, to manage risks involved in R&D and to enable the creation of new value through co-creation, the causes and reasons for inter-firm collaboration are well-known. However, little effort has been focused at critically examining the challenges that co-creation brings on a network and firm-level. This research addresses this issue by taking a process perspective on the formation and development of an inter-firm network in relation to its technologically turbulent environment. Building on a case study involving firms from that network, this research shows that such arrangements may also involve challenges for participating firms. These challenges relates to a paradoxical tension between exploitation of relation-specific assets and success in the long- and short-term, but also a challenge in terms of positioning the firm within the network.

  • The Dynamics of Digital Transformation : the Role of Digital Innovation, Ecosystems and Logics in Fundamental Organizational Change

    • This dissertation investigates digital transformation, understood here as processes where organizational actors engage in digital innovation and transform their organizations in order to respond to change in their business and technology environments. Specifically, it examines the dynamics of digital transformation, seeking to understand the key sociotechnical elements and their relationships that drive digital transformation processes and influence how they unfold over time. To theorize the dynamics of digital transformation, I synthesize extant knowledge with contributions from four appended research papers.The outset for theorizing in this dissertation is a body of literature that has begun to accumulate knowledge on digital transformation as a distinct phenomenon. Within this literature, I identify three main areas that are vital to understanding digital transformation processes, yet have so far not been sufficiently theorized. First, research on digital transformation often describe it as a complex and longitudinal process that involves several sequences of digital innovation, yet it has primarily been studied in the form of discrete instances of innovation decoupled in time and space. As a result, current knowledge on digital transformation as a longitudinal process is limited. Second, the literature on digital transformation emphasize that interactions between digital business and technology environments and organizations are crucial for explaining why and how digital transformation unfolds. At the same time, however, the literature has so far not been able to offer a conceptualization of these interactions in ways that make formative influence over time visible. Third, existing research on digital transformation has remained dominantly focused on the role of managers and paid limited attention to other organizational actors in digital transformation.Addressing the limitations identified in existing digital transformation research, I draw upon established theoretical concepts and the four appended research papers to theorize a conceptual framework on digital transformation dynamics. The conceptual framework contributes to research by clarifying a set of theoretical concepts and relationships that are instrumental for addressing digital transformation as a sequential and cumulative process, and the actors, agency and actions that realize digital transformation over time. It is supportive of future theorizing of digital transformation as a subject matter related yet distinct from other forms of organizational change enabled by technology use.
