  • Genome-wide association study identifies Sjogren's risk loci with functional implications in immune and glandular cells
  • 2022
  • Ingår i: Nature Communications. - : Springer Science and Business Media LLC. - 2041-1723. ; 13:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Sjogren's disease is a complex autoimmune disease with twelve established susceptibility loci. This genome-wide association study (GWAS) identifies ten novel genome-wide significant (GWS) regions in Sjogren's cases of European ancestry: CD247, NAB1, PTTG1-MIR146A, PRDM1-ATG5, TNFAIP3, XKR6, MAPT-CRHR1, RPTOR-CHMP6-BAIAP6, TYK2, SYNGR1. Polygenic risk scores yield predictability (AUROC = 0.71) and relative risk of 12.08. Interrogation of bioinformatics databases refine the associations, define local regulatory networks of GWS SNPs from the 95% credible set, and expand the implicated gene list to >40. Many GWS SNPs are eQTLs for genes within topologically associated domains in immune cells and/or eQTLs in the main target tissue, salivary glands. The genetic architecture underlying Sjogren's syndrome is not fully understood. Here, the authors perform a genome-wide association study to identify 10 new genetic risk regions, implicating genes involved in immune and salivary gland function.

  • Association of Genes in the NF-kappa B Pathway with Antibody-Positive Primary Sjogren's Syndrome
  • 2013
  • Ingår i: Scandinavian Journal of Immunology. - : Wiley. - 0300-9475 .- 1365-3083. ; 78:5, s. 447-454
  • Tidskriftsartikel (refereegranskat)abstract
    • Primary Sjogren's syndrome (SS) is a systemic autoimmune inflammatory disease characterized by focal lymphocytic infiltrates in the lachrymal and salivary glands and autoantibodies against the SSA/Ro and SSB/La antigens. Experimental studies have shown an activation of NF-B in primary SS. NF-B activation results in inflammation and autoimmunity and is regulated by inhibitory and activating proteins. Genetic studies have shown an association between multiple autoimmune diseases and TNFAIP3 (A20) and TNIP1 (ABIN1), both repressors of NF-B and of IKBKE (IKK epsilon), which is an NF-B activator. The aim of this study was to analyse single nucleotide polymorphisms (SNPs) in the IKBKE, NFKB1, TNIP1 and TNFAIP3 genes for association with primary SS. A total of 12 SNPs were genotyped in 1105 patients from Scandinavia (Sweden and Norway, n=684) and the UK (n=421) and 4460 controls (Scandinavia, n=1662, UK, n=2798). When patients were stratified for the presence of anti-SSA and/or anti-SSB antibodies (n=868), case-control meta-analysis found an association between antibody-positive primary SS and two SNPs in TNIP1 (P=3.4x10(-5), OR=1.33, 95%CI: 1.16-1.52 for rs3792783 and P=1.3x10(-3), OR=1.21, 95%CI: 1.08-1.36 for rs7708392). A TNIP1 risk haplotype was associated with antibody-positive primary SS (P=5.7x10(-3), OR=1.47, 95%CI: 1.12-1.92). There were no significant associations with IKBKE, NFKB1 or TNFAIP3 in the meta-analysis of the Scandinavian and UK cohorts. We conclude that polymorphisms in TNIP1 are associated with antibody-positive primary SS.

  • The effect of the polymerisation temperature on molecular weight and photoluminescence quantum yield for a phenylsubstituted PPV
  • 2001
  • Ingår i: Synthetic metals. - 0379-6779 .- 1879-3290. ; 119:1-3, s. 63-64
  • Tidskriftsartikel (refereegranskat)abstract
    • We present the synthesis and characterisation of poly(2-(2',5'-bis(2?-ethylhexyloxy)phenyl)-1, 4-phenylenevinylene) (BEHP-PPV) polymerised under different conditions. The photoluminescence efficiencies (?PL) in the solid state of BEHP-PPV obtained at 144°C and 0°C are 28% and 60%, respectively. Polymerisation temperatures below 0°C decreases the molecular weight without changing the photoluminescence efficiency to any large extent.

  • Conformational disorder of a substituted polythiophene in solution revealed by excitation transfer
  • 2001
  • Ingår i: Chemical Physics Letters. - 0009-2614 .- 1873-4448. ; 339:1-2, s. 96-102
  • Tidskriftsartikel (refereegranskat)abstract
    • Anisotropy and isotropic transient absorption decays of a polythiophene derivative in dilute solution have been measured and compared to calculated decays on Monte Carlo simulated polymer chains to obtain knowledge about the geometrical conformation of the polymer. The polymer chain conformations are generated by a partially correlated self-avoiding random walk. Excitation migration along the isolated chain is modeled assuming a hopping mechanism. This approach can be used to determine conformational disorder and possible realizations of the polymer chains in solution as well as the trapping time and migration length of the excitation. © 2001 Elsevier Science B.V.

  • Convenient synthetic route to poly(p-phenylene-1,2-diphenylvinylenes)
  • 2000
  • Ingår i: Synthetic metals. - 0379-6779 .- 1879-3290. ; 113:3, s. 293-297
  • Tidskriftsartikel (refereegranskat)abstract
    • The synthesis and characterization of two soluble poly(p-phenylenevinylene)s substituted with phenyl groups on the vinylene moiety is reported. The polymers are poly(1,4-phenylene-1,2-diphenylvinylene) (PPV-DP) and poly(1,4-phenylene-1,2-(3'-(2?-ethylhexyloxy))-diphenylvinylene) (PPV-mEHDP). The monomers containing chloromethyl groups were polymerized with excess potassium tert-butoxide in dioxane at room temperature. The polymerization conditions are optimized. The photoluminescence (PL) properties of the polymers showed an increase in the PL efficiency in film from 19% for PPV-DP to 37% for PPV-mEHDP. Both polymers emitted green light with higher intensity in film than in solution.

  • Synthesis and characterisation of polyfluorenes with light-emitting segments
  • 2001
  • Ingår i: Synthetic metals. - 0379-6779 .- 1879-3290. ; 121:1-3, s. 1761-1762
  • Tidskriftsartikel (refereegranskat)abstract
    • We present the synthesis and characterisation of two copolymers. They consists of poly(9-hexyl-9-(2'-ethylhexyl)fluorene) (HEH-PF) with a small amount of another polymer with a narrow band-gap. The synthetic route was designed to allow the incorporation of only one low band-gap segment per polymer chain. Photoluminescence measurements showed that energy transfer from the PF-segments to the low band-gap segments, did occur. In the solid state, all emissions were detected from the low band-gap segments with photoluminescence quantum yields up to 68%. One copolymer showed a low threshold for lasing (2 µJ/cm2) in a microcavity device.

  • Concomitant Ro/SSA and La/SSB antibodies are biomarkers for the risk of venous thromboembolism and cerebral infarction in primary Sjögren's syndrome
  • 2019
  • Ingår i: Journal of Internal Medicine. - : John Wiley & Sons. - 0954-6820 .- 1365-2796. ; 286:4, s. 458-468
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: To assess the risk of incident cardiovascular disease in patients with primary Sjögren's syndrome, overall and stratified by Ro/SSA and La/SSB autoantibody status. Methods: A cohort of patients with primary Sjögren's syndrome in Sweden (n = 960) and matched controls from the general population (n = 9035) were included, and data extracted from the National Patient Register to identify events of myocardial infarction, cerebral infarction and venous thromboembolism. Hazard ratios were estimated using cox proportional hazard regressions. Results: During a median follow‐up of 9.5 years, the overall hazard ratio (HR) was 1.6 (95% CI 1.2–2.1) for myocardial infarction, 1.2 (95% CI 0.9–1.7) for cerebral infarction and 2.1 (95% CI 1.6–2.9) for venous thromboembolism. Patients positive for both Ro/SSA and La/SSB autoantibodies had a substantially higher risk of cerebral infarction (HR 1.7, 95% CI 1.0–2.9) and venous thromboembolism (HR 3.1, 95% CI 1.9–4.8) than the general population. These risks were not significantly increased in Ro/SSA‐ and La/SSB‐negative patients. Among autoantibody‐positive patients, the highest HR of cerebral infarction was seen after ≥10 years disease duration (HR 2.8, 95% CI 1.4–5.4), while the HR for venous thromboembolism was highest 0–5 years after disease diagnosis (HR 4.7, 95% CI 2.3–9.3) and remained high throughout disease duration. Conclusions: Primary Sjögren's syndrome is associated with a markedly increased risk of cardiovascular disease and the presence of Ro/SSA and La/SSB autoantibodies identify the subgroup of patients carrying the highest risk. These findings suggest that monitoring and prevention of cardiovascular disease in this patient group should be considered.

  • Association of EBF1, FAM167A(C8orf13)-BLK and TNFSF4 gene variants with primary Sjögren's syndrome
  • 2011
  • Ingår i: Genes and Immunity. - : Springer Science and Business Media LLC. - 1466-4879 .- 1476-5470. ; 12:2, s. 100-109
  • Tidskriftsartikel (refereegranskat)abstract
    • We performed a candidate gene association study in 540 patients with primary Sjögren's Syndrome (SS) from Sweden (n=344) and Norway (n=196) and 532 controls (n=319 Swedish, n=213 Norwegian). A total of 1139 single-nucleotide polymorphisms (SNPs) in 84 genes were analyzed. In the meta-analysis of the Swedish and Norwegian cohorts, we found high signals for association between primary SS and SNPs in three gene loci, not previously associated with primary SS. These are the early B-cell factor 1 (EBF1) gene, P=9.9 × 10−5, OR 1.68, the family with sequence similarity 167 member A–B-lymphoid tyrosine kinase (FAM167A–BLK) locus, P=4.7 × 10−4, OR 1.37 and the tumor necrosis factor superfamily (TNFSF4=Ox40L) gene, P=7.4 × 10−4, OR 1.34. We also confirmed the association between primary SS and the IRF5/TNPO3 locus and the STAT4 gene. We found no association between the SNPs in these five genes and the presence of anti-SSA/anti-SSB antibodies. EBF1, BLK and TNFSF4 are all involved in B-cell differentiation and activation, and we conclude that polymorphisms in several susceptibility genes in the immune system contribute to the pathogenesis of primary SS.

  • Luminescence from inter-chain aggregates in polythiophene films
  • 2001
  • Ingår i: Synthetic Metals. - 0379-6779 .- 1879-3290. ; 119:1-3, s. 603-604
  • Tidskriftsartikel (refereegranskat)abstract
    • We report time-resolved photoluminescence of two polythiophene derivatives with different chain packing. Emission of inter-chain aggregates is distinguished from that of intra-chain singlet excitons. The aggregate luminescence is shifted towards lower energies relative to that of intra-chain exciton by 0.17 eV, and the Huang-Rhys factor of the coupling to the intra-chain C double bondC vibration is S = 1.5. The excitonic coupling in aggregates is of intermediate strength.

  • 2000
  • Ingår i: Chemical Physics Letters. - 0009-2614 .- 1873-4448. ; 322:1-2, s. 136-142
  • Tidskriftsartikel (refereegranskat)abstract
    • Photoexcitation dynamics in films of polythiophenes with different side groups were studied by transient absorption spectroscopy using 80 fs pulses. We show that inter-chain charge pairs (CP) are generated with similar to 20% efficiency in a polymer with short inter-chain distance (similar to 4 Angstrom) within 100 fs after photoexcitation. We discuss two mechanisms for inter-chain charge separation: (i) electron or hole transfer from an initially excited intra-chain singlet state or (ii) optical excitation of mixed exciton-charge transfer states, which quickly evolve to inter-chain CPs. • A Population-based Investigation of the Autoantibody Profile in Mothers of Children with Atrioventricular Block
  • 2011
  • Ingår i: Scandinavian Journal of Immunology. - Oxford : Blackwell Publishing. - 0300-9475 .- 1365-3083. ; 74:5, s. 511-517
  • Tidskriftsartikel (refereegranskat)abstract
    • The objective of the study was to investigate the antigen specificity and occurrence of individual autoantibodies in mothers of children diagnosed with atrioventricular (AV) block in a nation-wide setting. Patients with AV block detected before 15 years of age were identified using national quality registries as well as a network of pediatric and adult cardiologists and rheumatologists at the six university hospitals in Sweden. Patients with gross heart malformations, surgically or infectiously induced blocks were excluded. Blood samples were obtained from the mothers and maternal autoantibody profile, including the occurrence of antibodies against Ro52, Ro60, La, SmB, SmD, RNP-70k, RNP-A, RNP-C, CENP-C, Scl-70, Jo-1, ribosomal RNP and histones was investigated in 193 mothers of children with AV block by immunoblotting and ELISA. Autoantibody reactivity was detected in 48% (93/193) of the mothers of children with AV block. In autoantibody-positive mothers, the vast majority, 95% (88/93), had antibodies against Ro52, while 63% (59/93) had autoantibodies to Ro60 and 58% (54/93) had autoantibodies to La. In addition, 13% (12/93) of the autoantibody-positive mothers had antibodies to other investigated antigens besides Ro52, Ro60 and La, and of these anti-histone antibodies were most commonly represented, detected in 8% (7/93) of the mothers. In conclusion, this Swedish population-based study confirms that maternal autoantibodies may associate with heart block in the child. Further, our data demonstrate a dominant role of Ro52 antibodies in association with AV block.

  • 2001
  • Ingår i: Chemical Physics Letters. - 0009-2614 .- 1873-4448. ; 337:4-6, s. 277-283

    • Steady-state and rime-resolved photoluminescence (PL) is used to probe different states of order in highly regular poly(3-(2'-methoxy-5'-octylphenyl)thiophene) (POMeOPT). In a good solvent (chloroform) the polymer has a PL yield of 0.33 and in a solvent with low solubility (mixture of chloroform and toluene) a new phase appears with higher PL yield (0.50). The new phase is attributed to micro-crystals formed by the regular part of the polymer which have low solubility in toluene. The crystallites have highly structured and redshifted electronic transitions compared with chains which are dissolved. (C) 2001 Published by Elsevier Science B.V.

  • Interchain photoluminescence in substituted polyfluorenes
  • 2001
  • Ingår i: Synthetic metals. - 0379-6779 .- 1879-3290. ; 119:1-3, s. 615-616
  • Tidskriftsartikel (refereegranskat)abstract
    • We have characterised the interchain emitting states of two substituted polyfluorenes with steady-state and time-resolved photoluminescence (PL). Site selective excitation of the interchain state shows that physical dimers are formed in the film. These dimers have low concentration in the films but exciton diffusion to the dimers is reducing the intrachain exciton lifetime. With asymmetrical substituents, the PL quantum yield is higher and the PL lifetime is longer due to lower concentration of dimers. Slow cooling from the melt, gives a new red shifted emission band and less intrachain emission. A discrepancy in the calculated radiative lifetimes suggest a fast formation of this new band.

  • 2001
  • Ingår i: Macromolecules. - : American Chemical Society (ACS). - 0024-9297 .- 1520-5835. ; 34:21, s. 7294-7299
  • Tidskriftsartikel (refereegranskat)abstract
    • The synthesis and characterization of poly(pyridine vinylene) (PPyV) via the nonionic sulfinyl precursor route is presented. Starting from an unsymmetrical monomer, precursor polymers were prepared in various solvents, which led to polymers with variable molecular weights. The thermal conversion to the conjugated structure, as well as its stability, was studied with different techniques such as FT-IR, UV-vis, TGA, and direct insertion probe mass spectroscopy (DIP-MS). From these results we were able to derive the most suitable conditions to perform the conversion. The fully conjugated PPyV was further characterized with photoluminescence (PL) and cyclic voltammetry (CV) measurements. The PL efficiency was found to be as high as 14%. The CV measurements showed that the polymer can be reduced (n-doped).

  • Gender differences in symptoms of depression among adolescents in Eastern Norway : Results from a cross-sectional study
  • 2019
  • Ingår i: Scandinavian Journal of Public Health. - : SAGE Publications Ltd. - 1403-4948 .- 1651-1905. ; 47:2, s. 157-165
  • Tidskriftsartikel (refereegranskat)abstract
    • Aim: The aim of this study was to compare and describe gender differences and the associations between symptoms of depression and family conflict and economics, lifestyle habits, school satisfaction and the use of health-care services among adolescents. Methods: Data were retrieved from Ungdata which is a cross-sectional study. Adolescents (n=8052) from secondary school grades 8, 9 and 10 (age 13–16 years) participated in the study from 41 municipal schools in four counties. Results: Girls reported a higher prevalence of symptoms of depression than boys. Gender differences were seen on all items related to symptoms of depression, family conflict and economics, lifestyle habits, school satisfaction and health-care services. Multiple regressions showed that family conflicts and economics contributed to 19.2% of the variance in symptoms of depression in girls and 12.4% in boys. School satisfaction made a strong contribution: 21.5% in girls and 15.4% in boys. The total model explained 49% of the total variance in symptoms of depression in girls and 32.5% in boys. Conclusions: Gender demonstrated a pattern through a higher proportion of girls reporting symptoms of depression, family conflict and economics, lifestyle habits, school satisfaction and use of health-care services. Even though the adolescents reported symptoms of depression, few used the school health-care services and public health nurses. This indicates that they need a person-centered approach for symptoms of depression. The findings may have important implications for planning for adolescents in school health services.

  • No association of primary Sjogren's syndrome with Fc gamma receptor gene variants
  • 2013
  • Ingår i: Genes and Immunity. - : Springer Science and Business Media LLC. - 1476-5470 .- 1466-4879. ; 14:4, s. 234-237
  • Tidskriftsartikel (refereegranskat)abstract
    • The genetic background of primary Sjogren's syndrome (pSS) is partly shared with systemic lupus erythematosus (SLE). Immunoglobulin G Fc receptors are important for clearance of immune complexes. Fcg receptor variants and gene deletion have been found to confer SLE risk. In this study, four Fc gamma receptor single-nucleotide polymorphisms (SNPs) and one copy number variation (CNV) were studied. Swedish and Norwegian pSS patients (N = 527) and controls (N = 528) were genotyped for the Fc gamma receptor gene variant FCGR2A H131R (rs1801274) by the Illumina GoldenGate assay. FCGR3A F158V (rs396991) was analysed in 488 patients and 485 controls, FCGR3B rs447536 was analysed in 471 patients and 467 controls, and FCGR3B rs448740 was analysed in 478 cases and 455 controls, using TaqMan SNP genotyping assays. FCGR3B CNV was analysed in 124 patients and 139 controls using a TaqMan copy number assay. None of the SNPs showed any association with pSS. Also, no FCGR3B CNV association was detected. The lack of association of pSS with Fc gamma receptor gene variants indicates that defective immune complex clearance may not be as important in pSS pathogenesis as in SLE, and may point to important differences between SLE and pSS.

  • Associations of cigarette smoking with disease phenotype and type I interferon expression in primary Sjogren's syndrome
  • 2019
  • Ingår i: Rheumatology International. - : Springer Science and Business Media LLC. - 0172-8172 .- 1437-160X. ; 39:9, s. 1575-1584
  • Tidskriftsartikel (refereegranskat)abstract
    • Several studies have shown a negative association between smoking and primary Sjogren's syndrome (pSS), and smoking may interfere with the immune response. The purpose of this study was to investigate if smoking affects disease activity and disease phenotype in pSS. In this cross-sectional study, consecutive pSS patients filled out the EULAR Sjogren's Syndrome Patient Reported Index (ESSPRI) form and a structured questionnaire regarding smoking habits. EULAR Sjogren's Syndrome Disease Activity Index (ESSDAI) scores were calculated and blood samples were analysed for type I interferon signature using RT-PCR. Of 90 patients (93% women, median age 66.5 years), 72% were type I IFN signature positive and 6, 42 and 53% were current, former and never smokers, respectively. No significant differences by smoking status were found regarding ESSDAI total score, activity in the ESSDAI domains or type I IFN signature. Patients with a higher cumulative cigarette consumption (>= median) had higher scores in ESSPRI total [5.0 (3.0-6.3) vs 8.0 (6.0-8.3); p < 0.01] and ESSPRI sicca and pain domains. Comparing type I IFN signature negative and positive patients, the latter had significantly lower activity in ESSDAI articular domain (7/25 vs 3/64; p < 0.01) and lower scores in ESSPRI total [7.7 (5.2-8.2) vs 6.0 (4.0-7.7); p = 0.04]. Smoking was not associated with disease phenotype although patients with a higher cumulative cigarette consumption had worse symptoms in some disease domains. Current smokers were few making it difficult to draw any firm conclusions about associations to current smoking.

  • Intra- and interchain luminescence in amorphous and semicrystalline films of phenyl-substituted polythiophene
  • 2001
  • Ingår i: JOURNAL OF PHYSICAL CHEMISTRY B. - : American Chemical Society (ACS). - 1089-5647 .- 1520-6106 .- 1520-5207. ; 105:32, s. 7624-7631
  • Tidskriftsartikel (refereegranskat)abstract
    • We report time-resolved photoluminescence (PL) measurements of spin-cast amorphous films of a regioregular polythiophene derivative poly[3-(4-octylphenyl)thiophene] and of annealed semicrystalline films of the same polymer with an interchain stacking distance of 5 Angstrom. Red-shifted PL appears at long delay times in both pristine and annealed films, which we assign to interchain aggregates populated by excitation energy migration. Aggregate luminescence in annealed films exhibits a pronounced vibronic structure indicating the coupling to a C=C bond stretch with a Huang-Rhys factor S = 2. Two types of aggregates are distinguished in annealed films: in a few picoseconds most excitations are trapped by aggregates with a large energy gap (about 1.83 eV) between the lowest excited singlet state and the ground state. Excitation energy is transferred or the aggregates relax structurally with a time constant of about 200 ps to aggregates with a smaller energy gap (about 1.75 eV). The radiative lifetime of aggregates with smaller energy gap is estimated to be about 35 ns, two times longer than that of aggregates with the larger energy gap. In spin-cast amorphous films, emission from intrachain singlet excitons has a longer lifetime than in annealed films and only high-energy-gap aggregates are populated.

  • Photoluminescence quenching at a polythiophene/C-60 heterojunction

  • Ingår i: Physical Review B (Condensed Matter and Materials Physics). - 1098-0121 .- 1550-235X. ; 61:19, s. 12957-12963
  • Tidskriftsartikel (refereegranskat)abstract
    • Quenching of photoluminescence in a substituted polythiophene in the presence of a deposited C-60 layer is studied by steady-state and time-resolved photoluminescence (PL). The steady-state PL is evaluated by con -sidering the interference of the absorbed and emitted electro-optical field in the thin film coupled to exciton diffusion in the conjugated polymer. PL quenching occurs for excitons generated within 5 nm from the heterojunction. A blueshift of the polymer emission spectrum is observed when C-60 is deposited on top of a polymer thin film. The blueshift is shown to be caused by PL quenching before the excitation is transferred to the lowest-energy sites.

  • Sex-based differences in association between circulating T cell subsets and disease activity in untreated early rheumatoid arthritis patients
  • 2018
  • Ingår i: Arthritis Research & Therapy. - : Springer Science and Business Media LLC. - 1478-6354 .- 1478-6362. ; 20:1
  • Tidskriftsartikel (refereegranskat)abstract
    • Background: It is not known if sex-based disparities in immunological factors contribute to the disease process in rheumatoid arthritis (RA). Hence, we examined whether circulating T cell subset proportions and their association with disease activity differed in male and female patients with untreated early rheumatoid arthritis (ueRA). Methods: Proportions of T cell subsets were analyzed in peripheral blood from 72 ueRA DMARD-and corticosteroid-naive patients (50 females and 22 males) and in 31 healthy age-and sex-matched controls. Broad analysis of helper and regulatory CD4(+) T cell subsets was done using flow cytometry. Disease activity in patients was assessed using DAS28, CDAI, swollen joint counts, tender joint counts, CRP, and ESR. Results: Multivariate factor analyses showed that male and female ueRA patients display distinct profiles of association between disease activity and circulating T cell subset proportions. In male, but not female, ueRA patients Th2 cells showed a positive association with disease activity and correlated significantly with DAS28-ESR, CDAI, and swollen and tender joint counts. Likewise, proportions of non-regulatory CTLA-4(+) T cells associated positively with disease activity in male patients only, and correlated with DAS28-ESR. In contrast, there was a negative relation between Th1Th17 subset proportions and disease activity in males only. The proportions of Th17 cells correlated positively with DAS28-ESR in males only, while proportions of Th1 cells showed no relation to disease activity in either sex. There were no significant differences in proportions of T cell subsets between the sexes in patients with ueRA. Conclusions: Our findings show sex-based differences in the association between T cell subsets and disease activity in ueRA patients, and that Th2 helper T cells may have a role in regulating disease activity in male patients.

  • Association Between Bone Mineral Density and Autoantibodies in Patients With Rheumatoid Arthritis
  • 2021
  • Ingår i: Arthritis and Rheumatology. - : Wiley. - 2326-5191 .- 2326-5205. ; 73:6, s. 921-930
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: Autoantibodies, such as anti–citrullinated protein antibodies (ACPAs), have been described as inducing bone loss in rheumatoid arthritis (RA), which can also be reflected by bone mineral density (BMD). We therefore examined the association between osteoporosis and autoantibodies in two independent RA cohorts. Methods: Dual x-ray absorptiometry (DXA) of the lumbar spine and left hip was performed in 408 Dutch patients with early RA during 5 years of follow-up and in 198 Swedish patients with early RA during 10 years of follow-up. The longitudinal effect of ACPAs and other autoantibodies on several BMD measures was assessed using generalized estimating equations. Results: In the Dutch cohort, significantly lower BMD at baseline was observed in ACPA-positive patients compared to ACPA-negative patients, with an estimated marginal mean BMD in the left hip of 0.92 g/cm2 (95% confidence interval [95% CI] 0.91–0.93) versus 0.95 g/cm2 (95% CI 0.93–0.97) (P = 0.01). In line with this, significantly lower Z scores at baseline were noted in the ACPA-positive group compared to the ACPA-negative group (estimated marginal mean Z score in the left hip of 0.18 [95% CI 0.08–0.29] versus 0.48 [95% CI 0.33–0.63]) (P < 0.01). However, despite clear differences at baseline, ACPA positivity was not associated with greater decrease in absolute BMD or Z scores over time. Furthermore, there was no association between BMD and higher levels of ACPAs or other autoantibodies (rheumatoid factor and anti–carbamylated protein antibodies). In the Swedish cohort, ACPA-positive patients tended to have a higher prevalence of osteopenia at baseline (P = 0.04), but again, ACPA positivity was not associated with an increased prevalence of osteopenia or osteoporosis over time. Conclusion: The presence of ACPAs is associated with significantly lower BMD at baseline, but not with greater BMD loss over time in treated RA patients. These results suggest that ACPAs alone do not appear to contribute to bone loss after disease onset when disease activity is well-managed. © 2020 The Authors. • Metabolic Profiling of Systemic Lupus Erythematosus and Comparison with Primary Sjögren’s Syndrome and Systemic Sclerosis
  • 2016
  • Ingår i: PLOS ONE. - : Public Library of Science (PLoS). - 1932-6203. ; 11:7
  • Tidskriftsartikel (refereegranskat)abstract
    • Systemic lupus erythematosus (SLE) is a chronic inflammatory autoimmune disease which can affect most organ systems including skin, joints and the kidney. Clinically, SLE is a heterogeneous disease and shares features of several other rheumatic diseases, in particular primary Sjögrens syndrome (pSS) and systemic sclerosis (SSc), why it is difficult to diag- nose The pathogenesis of SLE is not completely understood, partly due to the heterogeneity of the disease. This study demonstrates that metabolomics can be used as a tool for improved diagnosis of SLE compared to other similar autoimmune diseases. We observed differences in metabolic profiles with a classification specificity above 67% in the comparison of SLE with pSS, SSc and a matched group of healthy individuals. Selected metabolites were also significantly different between studied diseases. Biochemical pathway analysis was conducted to gain understanding of underlying pathways involved in the SLE pathogenesis. We found an increased oxidative activity in SLE, supported by increased xanthine oxidase activity and an increased turnover in the urea cycle. The most discriminatory metabolite observed was tryptophan, with decreased levels in SLE patients compared to control groups. Changes of tryptophan levels were related to changes in the activity of the aromatic amino acid decarboxylase (AADC) and/or to activation of the kynurenine pathway.

  • Strukturbiblioteket
  • 2020
• Epstein-Barr virus early antigen diffuse (EBV-EA/D)-directed immunoglobulin A antibodies in systemic lupus erythematosus patients
  • 2012
  • Ingår i: Scandinavian Journal of Rheumatology. - : Informa UK Limited. - 1502-7732 .- 0300-9742. ; 41:4, s. 280-289
  • Tidskriftsartikel (refereegranskat)abstract
    • Objective: We sought to determine whether the serological response towards lytic cycle antigens of Epstein Barr virus (EBV) is altered in systemic lupus erythematosus (SLE) patients. Method: We used enzyme-linked immunosorbent assay (ELISA) to investigate the prevalence of EBV early antigen diffuse (EBV-EA/D) antibodies in sera from 60 patients with SLE, 40 with scleroderma (SSc), 20 with primary Sjogren's syndrome (pSS), 20 with rheumatoid arthritis (RA), 20 healthy controls, and also subjects with various circulating autoantibodies. Samples from patients were obtained from clinics specialized within the diseases in Denmark and Sweden and samples from healthy controls were obtained from volunteers. Results: A significant elevated titre of immunoglobulin (Ig)A, IgG, and IgM EBV-EA/D antibodies was found in SLE patients compared to healthy controls, a finding not explained by immunosuppressive treatment or disease activity. The largest difference was observed for IgA EBV-EA/D antibodies (p = 0.0013) with a seropositive rate of 58% in SLE patients and 0% in healthy controls. RA and SSc patients and individuals seropositive for anti-Scl-70 were additionally found to have elevated titres of IgA EBV-EA/D antibodies (40%, p = 0.014; 60%, p = 0.015; and 38.5%, p = 0.045, respectively). However, the titres were generally lower than in SLE patients. Conclusion: Our findings support an association between EBV and SLE. The elevated titre of EBV-EA/D-directed IgA antibodies found in SLE patients could suggest reactivation of EBV in epithelial cells or reinfection of epithelial cells after reactivation in B cells, indicating lack of control of the latent infection.
