  • Foraminiferal Mn/Ca as Bottom-Water Hypoxia Proxy: An Assessment of Nonionella stella in the Santa Barbara Basin, USA

    • Hypoxia is of increasing concern in marine areas, calling for a better understanding of mechanisms leading to decreasing dissolved oxygen concentrations ([O2]). Much can be learned about the processes and implications of deoxygenation for marine ecosystems using proxy records from low-oxygen sites, provided proxies, such as the manganese (Mn) to calcium (Ca) ratio in benthic foraminiferal calcite, are available and well calibrated. Here we report a modern geochemical data set from three hypoxic sites within the Santa Barbara Basin (SBB), USA, where we study the response of Mn/Caforam in the benthic foraminifer Nonionella stella to variations in sedimentary redox conditions (Mn, Fe) and bottom-water dissolved [O2]. We combine molecular species identification by small subunit rDNA sequencing with morphological characterization and assign the SBB N. stella used here to a new phylotype (T6). Synchrotron-based scanning X-ray fluorescence (XRF) imaging and Secondary Ion Mass Spectrometry (SIMS) show low Mn incorporation (partition coefficient DMn < 0.05) and limited proxy sensitivity of N. stella, at least within the range of dissolved [O2] (2.7–9.6 μmol/l) and Mnpore-water gradients (2.12–21.59 μmol/l). Notably, even though intra- and interspecimen Mn/Ca variability (33% and 58%, respectively) was only partially controlled by the environment, Mn/Caforam significantly correlated with both pore-water Mn and bottom-water [O2]. However, the prevalent suboxic bottom-water conditions and limited dissolved [O2] range complicate the interpretation of trace-elemental trends. Additional work involving other oxygenation proxies and samples from a wider oxygen gradient should be pursued to further develop foraminiferal Mn/Ca as an indicator for hypoxic conditions.

  • A micrometeorite from a stony asteroid identified in Luna 16 soil

    • Despite the intense cratering history of the Moon, very few traces of meteoritic material have been identified in the more than 380 kg of samples returned to Earth by the Apollo and Luna missions. Here we show that an ~200-µm-sized fragment collected by the Luna 16 mission has extra-lunar origins and probably originates from an LL chondrite with similar properties to near-Earth stony asteroids. The fragment has not experienced temperatures higher than 400 °C since its protolith formed early in the history of the Solar System. It arrived on the Moon, either as a micrometeorite or as the result of the break-up of a bigger impact, no earlier than 3.4 Gyr ago and possibly around 1 Gyr ago, an age that would be consistent with impact ages inferred from basaltic fragments in the Luna 16 sample and of a known dynamic upheaval in the Flora asteroid family, which is thought to be the source of L and LL chondrite meteorites. These results highlight the importance of extra-lunar fragments in constraining the impact history of the Earth–Moon system and suggest that material from LL chondrite asteroids may be an important component.

  • Genetic association study of childhood aggression across raters, instruments, and age

    • Childhood aggressive behavior (AGG) has a substantial heritability of around 50%. Here we present a genome-wide association metaanalysis (GWAMA) of childhood AGG, in which all phenotype measures across childhood ages from multiple assessors were included. We analyzed phenotype assessments for a total of 328 935 observations from 87 485 children aged between 1.5 and 18 years, while accounting for sample overlap. We also meta-analyzed within subsets of the data, i.e., within rater, instrument and age. SNP-heritability for the overall meta-analysis (AGGoverall) was 3.31% (SE= 0.0038). We found no genome-wide significant SNPs for AGG(overall). The gene-based analysis returned three significant genes: ST3GAL3 (P= 1.6E-06), PCDH7 (P= 2.0E-06), and IPO13 (P= 2.5E-06). All three genes have previously been associated with educational traits. Polygenic scores based on our GWAMA significantly predicted aggression in a holdout sample of children (variance explained = 0.44%) and in retrospectively assessed childhood aggression (variance explained = 0.20%). Genetic correlations (rg) among rater-specific assessment of AGG ranged from r(g)= 0.46 between self- and teacher-assessment to r(g)d= 0.81 between mother- and teacher-assessment. We obtained moderate-to-strong rgs with selected phenotypes from multiple domains, but hardly with any of the classical biomarkers thought to be associated with AGG. Significant genetic correlations were observed with most psychiatric and psychological traits (range r(g): 0.19-1.00), except for obsessive-compulsive disorder. Aggression had a negative genetic correlation (r(g)=-0.5) with cognitive traits and age at first birth. Aggression was strongly genetically correlated with smoking phenotypes (range |r(g)| : 0.46-0.60). The genetic correlations between aggression and psychiatric disorders were weaker for teacher-reported AGG than for mother- and self-reported AGG. The current GWAMA of childhood aggression provides a powerful tool to interrogate the rater-specific genetic etiology of AGG.

  • Single-Cell Measurements of Fixation and Intercellular Exchange of C and N in the Filaments of the HeterocystForming Cyanobacterium Anabaena sp. Strain PCC 7120

    • Under diazotrophic conditions, the model filamentous, heterocystforming cyanobacterium Anabaena sp. strain PCC 7120 develops a metabolic strategy based on the physical separation of the processes of oxygenic photosynthesis, in vegetative cells, and N-2 fixation, in heterocysts. This strategy requires the exchange of carbon and nitrogen metabolites and their distribution along the filaments, which takes place through molecular diffusion via septal junctions involving FraCD proteins. Here, Anabaena was incubated in a time course (up to 20 h) with [C-13]bicarbonate and N-15(2) and analyzed by secondary ion mass spectrometry imaging (SIMS) (large-geometry SIMS [LG- SIMS] and NanoSIMS) to quantify C and N assimilation and distribution in the filaments. The C-13/C-12 and N-15/N-14 ratios measured in wild-type filaments showed a general increase with time. The enrichment was relatively homogeneous in vegetative cells along individual filaments, while it was reduced in heterocysts. Heterocysts, however, accumulated recently fixed N at their poles, in which the cyanophycin plug [multi-L-arginyl-poly(L-aspartic acid)] is located. In contrast to the rather homogeneous label found along stretches of vegetative cells, C-13/C-12 and N-15/N-14 ratios were significantly different between filaments both at the same and different time points, showing high variability in metabolic states. A fraC fraD mutant did not fix N-2, and the C-13/C-12 ratio was homogeneous along the filament, including the heterocyst in contrast to the wild type. Our results show the consumption of reduced C in the heterocysts associated with the fixation and export of fixed N and present an unpredicted heterogeneity of cellular metabolic activity in different filaments of an Anabaena culture under controlled conditions. IMPORTANCE Filamentous, heterocyst- forming cyanobacteria represent a paradigm of multicellularity in the prokaryotic world. Physiological studies at the cellular level in model organisms are crucial to understand metabolic activities and qualify specific aspects related to multicellularity. Here, we used stable isotopes (C-13 and N-15) coupled to LG-SIMS and NanoSIMS imaging to follow single-cell C and N-2 fixation and metabolic dynamics along the filaments in the model heterocyst-forming cyanobacterium Anabaena sp. strain PCC 7120. Our results show a close relationship between C and N fixation and distribution in the filaments and indicate that wild-type filaments in a culture can exhibit a substantial variability of metabolic states. This illustrates how some novel properties can be appreciated by studying microbial cultures at the single-cell level.

  • U-Pb zircon geochronology from Haag Nunataks, Coats Land and Shackleton Range (Antarctica) : Constraining the extent of juvenile Late Mesoproterozoic arc terranes

    • Understanding the accretionary stages of Rodinia evolution and the arrangement of cratons and arc terranes is dependent upon high-precision geochronology from key piercing points of Mesoproterozoic rocks. U-Pb zircon dating is presented here from the Mesoproterozoic Haag Nunataks gneiss complex of West Antarctica where the dominant granodiorite protolith was emplaced at 1238 ± 4 Ma, aplite/pegmatite sheets were intruded at 1064 ± 4 Ma and the final intrusive phase of microgranite sheets were emplaced at 1056 ± 8 Ma. A separate magmatic event at ~1170 Ma is recorded as inherited zircons in the later stage intrusions. Based on field relationships, the main phase of deformation at Haag Nunataks is thought to have developed prior to the emplacement of the microgranite sheet at ~1056 Ma but after the ~1064 Ma aplite/pegmatite intrusive phase. Potentially correlative units from the Shackleton Range and Coats Land of East Antarctica are also dated to test supposed correlations with arc terranes and crustal blocks at the margins of Laurentia and the proto-Kalahari craton. An ice-transported granite pegmatite sample recovered from the Brunt Ice Shelf is used as a partial proxy for unexposed rocks of the ice-covered Coats Land block and has been dated at ~1100 Ma. A diorite gneiss from the Shackleton Range was also analysed as it forms part of a magnetic domain shared with the Haag Nunataks crustal block. Core zircon ages of ~2470 Ma were determined, and the age of migmatisation is interpreted at ~1740 Ma and rules out any potential correlation with the Haag Nunataks gneiss complex. The magmatic precursors of the Haag Nunataks orthogneisses were emplaced in a juvenile arc setting. We argue that this arc was located in the Natal Embayment region, contiguous with the Namaqua-Natal-Maud belt of arc terranes typified by enhanced magmatism at ~1240 Ma and ~1170 Ma not associated with any significant deformation events. The later magmatic events at Haag Nunataks at ~1060 Ma are more closely associated with collision of Laurentia with the proto-Kalahari craton and the associated deformation is correlated with the Ottawan phase of the Grenville orogeny.

    • Carbonaceous matter (CM) in Archean rocks represents a valuable archive for the reconstruction of early life. Here we investigate the nature of CM preserved in ∼ 3.5 Ga old black bedded barites from the Dresser Formation (Pilbara Carton, Western Australia). Using light microscopy and high-resolution Raman mapping, three populations of CM were recognized: (i) CM at the edges of single growth bands of barite crystals (most frequent), (ii) CM within the barite matrix, and (iii) CM in 50–300 µm wide secondary quartz veins that cross-cut the black bedded barite. Raman spectra of CM inside black bedded barite indicated peak metamorphic temperatures of ∼ 350 °C, consistent with those reached during the main metamorphic event in the area ∼ 3.3 Ga ago. By contrast, CM in quartz veins yielded much lower temperatures of ∼ 220 °C, suggesting that quartz-vein associated CM entered the barite after 3.3 Ga. Near edge X-ray absorption fine structure (NEXAFS) and solid-state nuclear magnetic resonance (NMR) revealed a highly aromatic nature of the CM with a lower aliphatic content, which is in line with the relatively elevated thermal maturity. Catalytic hydropyrolysis (HyPy) did not yield any hydrocarbons detectable with gas chromatography–mass spectrometry (GC–MS). Secondary ion mass spectrometry (SIMS) based δ13C values of individual CM particles ranged from − 33.4 ± 1.2 ‰ to − 16.5 ± 0.6 ‰ and are thus in accordance with a biogenic origin, which is also consistent with stromatolitic microbialites associated with the black bedded barite. Based on these results we conclude that CM at growth bands and inside the barite matrix is syngenetic and only the CM inside quartz veins, which represents a minor portion of the total CM, is a later addition to the system. Furthermore, we discuss different pathways for the input of CM into the barite-forming environment, including the cycling of biological organic material within the hydrothermal system.
