  • A GWAS follow-up study reveals the association of the IL12RB2 gene with systemic sclerosis in Caucasian populations

    • A single-nucleotide polymorphism (SNP) at the IL12RB2 locus showed a suggestive association signal in a previously published genome-wide association study (GWAS) in systemic sclerosis (SSc). Aiming to reveal the possible implication of the IL12RB2 gene in SSc, we conducted a follow-up study of this locus in different Caucasian cohorts. We analyzed 10 GWAS-genotyped SNPs in the IL12RB2 region (2309 SSc patients and 5161 controls). We then selected three SNPs (rs3790567, rs3790566 and rs924080) based on their significance level in the GWAS, for follow-up in an independent European cohort comprising 3344 SSc and 3848 controls. The most-associated SNP (rs3790567) was further tested in an independent cohort comprising 597 SSc patients and 1139 controls from the USA. After conditional logistic regression analysis of the GWAS data, we selected rs3790567 [P-MH = 1.92 x 10(-5) odds ratio (OR) = 1.19] as the genetic variant with the firmest independent association observed in the analyzed GWAS peak of association. After the first follow-up phase, only the association of rs3790567 was consistent (P-MH = 4.84 x 10(-3) OR = 1.12). The second follow-up phase confirmed this finding (P-chi 2 = 2.82 x 10(-4) OR = 1.34). After performing overall pooled-analysis of all the cohorts included in the present study, the association found for the rs3790567 SNP in the IL12RB2 gene region reached GWAS-level significant association (P-MH = 2.82 x 10(-9) OR = 1.17). Our data clearly support the IL12RB2 genetic association with SSc, and suggest a relevant role of the interleukin 12 signaling pathway in SSc pathogenesis.

  • Validation of five years (2003-2007) of SCIAMACHY CO total column measurements using ground-based spectrometer observations

    • This paper presents a validation study of SCanning Imaging Absorption spectroMeter for Atmospheric CHartographY (SCIAMACHY) carbon monoxide (CO) total column measurements from the Iterative Maximum Likelihood Method (IMLM) algorithm using ground-based spectrometer observations from twenty surface stations for the five year time period of 2003-2007. Overall we find a good agreement between SCIAMACHY and ground-based observations for both mean values as well as seasonal variations. For high-latitude Northern Hemisphere stations absolute differences between SCIAMACHY and ground-based measurements are close to or fall within the SCIAMACHY CO 2 sigma precision of 0.2 x 10(18) molecules/cm(2) (similar to 10%) indicating that SCIAMACHY can observe CO accurately at high Northern Hemisphere latitudes. For Northern Hemisphere mid-latitude stations the validation is complicated due to the vicinity of emission sources for almost all stations, leading to higher ground-based measurements compared to SCIAMACHY CO within its typical sampling area of 8 degrees x 8 degrees. Comparisons with Northern Hemisphere mountain stations are hampered by elevation effects. After accounting for these effects, the validation provides satisfactory results. At Southern Hemisphere mid-to high latitudes SCIAMACHY is systematically lower than the ground-based measurements for 2003 and 2004, but for 2005 and later years the differences between SCIAMACHY and ground-based measurements fall within the SCIAMACHY precision. The 2003-2004 bias is consistent with previously reported results although its origin remains under investigation. No other systematic spatial or temporal biases could be identified based on the validation presented in this paper. Validation results are robust with regard to the choices of the instrument-noise error filter, sampling area, and time averaging required for the validation of SCIAMACHY CO total column measurements. Finally, our results show that the spatial coverage of the ground-based measurements available for the validation of the 2003-2007 SCIAMACHY CO columns is sub-optimal for validation purposes, and that the recent and ongoing expansion of the ground-based network by carefully selecting new locations may be very beneficial for SCIAMACHY CO and other satellite trace gas measurements validation efforts.

    • In this paper, the experimental flow boiling visualization results of a microchannel are presented and discussed. A series of visualization experiments have been conducted in a horizontal, circular, uniformly heated microchannel, to record the two-phase flow patterns evolved during the boiling process and to study the ebullition process. A high speed camera (REDLAKE HG50LE) with a maximum of 100000 fps together with tungsten lights was used to capture the images along the test section. Microchannel was made of circular fused silica tube having an internal diameter of 0.781 mm and a uniformly heated length of 191 mm. Outside of the test tube was coated with a thin, electrically conductive layer of Indium Tin Oxide (ITO) for direct heating of the test section. Refrigerant R134a was used as working fluid and experiments were performed at two different system pressures corresponding to saturation temperatures of 25 degrees C and 30 degrees C. Mass flux was varied from 100 kg/m(2)s to 400 kg/m(2)s and heat flux ranged from 5 kW/m(2) to 45 kW/m(2). Visualization results show that the bubble growth is restricted by the tube diameter which results in very short existence of isolated bubbly flow regime except essentially restricted to a very short length of test tube. Flow patterns observed along the length were: Isolated bubble, elongated bubble, slug flow, semi annular and annular flow. Rigorous boiling and increased coalescence rates were observed with increase in heat flux. Bubble frequency was observed to increase with both heat and mass flux. A comparison with our previous flow boiling visualization studies, carried out for a test tube of 1.33 mm internal diameter, shows that the number of active nucleation sites is less while the bubble frequency is higher for the current study. Mean bubble length and bubble velocity during elongated bubble flow pattern have also been calculated from the images obtained during the tests.

    • Experimental results of two-phase pressure drop in a horizontal circular microchannel are reported in this paper. A test tube was made of fused silica having an internal diameter of 781 mu m with a total length of 261 mm and a heated length of 191 mm. The outer surface of the test tube was coated with an electrically conductive thin layer of ITO (indium tin oxide) for direct heating of the test section. Refrigerants R134a and R245fa were used as the working fluids, and mass flux during the experiments was varied between 100 and 650 kg/m(2)-s. Experiments were performed at two different system pressures corresponding to saturation temperatures of 25 degrees C and 30 degrees C for R134a and at three different system pressures corresponding to saturation temperatures of 30 degrees C, 35 degrees C, and 40 degrees C for R245fa. Two-phase frictional pressure drop characteristics with variation of mass flux, vapor fraction, saturation temperature, and heat flux were explored in detail. Finally, the prediction capability of some well-known correlations available in the literature, some developed for macrochannels and others especially developed for microchannels, was assessed.

    • In this paper the experimental flow boiling heat transfer results of a minichannel are presented. A series of experiments was conducted to measure the heat transfer coefficients in a minichannel made of stainless steel (AISI 316) having an internal diameter of 1.7mm and a uniformly heated length of 220mm. R134a was used as working fluid and experiments were performed at two different system pressures corresponding to saturation temperatures of 27 degrees C and 32 degrees C. Mass flux was varied from 50 kg/m(2) s to 600 kg/m(2) s and heat flux ranged from 2kW/m(2) to 156 kW/m(2). The test section was heated directly using a DC power supply. The direct heating of the channel ensured uniform heating and heating was continued until dry out was reached. The experimental results show that the heat transfer coefficient increases with imposed wall heat flux while mass flux and vapour quality have no considerable effect. Increasing the system pressure slightly enhances the heat transfer coefficient. The heat transfer coefficient is reduced as dryout is reached. It is observed that dryout phenomenon is accompanied with fluctuations and a larger standard deviation in outer wall temperatures.

    • Micro-channel-based evaporators are a promising option for high heat flux cooling applications. Micro-channels offer several advantages, including a smaller coolant inventory, superior heat transfer performance, compactness, lightness of weigh. Despite being attractive, the governing phenomena in micro-channels, especially during phase change, are less understood. This article reports the experimental flow boiling heat transfer results of refrigerants R134a and R245fa in a horizontal micro-channel. A series of experiments was conducted to measure the heat transfer coefficients in a circular micro-channel made of fused silica having an internal diameter of 781 mu m and a uniformly heated length of 191 mm. The outer surface of the test tube was coated with a thin, electrically conductive layer of indium-tin-oxide. The surface coating with the electrically conductive layer of indium-tin-oxide made it possible to visualize the flow boiling process simultaneously with uniform heating of the test section. R134a and R245fa were used as working fluids and experiments were performed at a system pressure of 7.7 bar for R134a and at 1.8 bar for R245fa, corresponding to saturation temperature of 30 degrees C. Mass flux was varied from 175 kg/m(2)s to 500 kg/m(2)s, and heat flux ranged from 5 kW/m(2) to 60 kW/m(2). A high-speed camera was used to capture the images in the case of flow boiling of R134a. The experimental results indicated that the heat transfer coefficient increased with heat flux while the mass flux proved to have a negligible effect on heat transfer coefficient.

    • Dense packaging of electronic components generates very high heat fluxes and therefore results in challenges for proper thermal management of such components. Microchannel based evaporators with phase changing liquids are regarded as a promising solution for such high heat flux cooling applications. Due to confinement of flow and differences in the relative importance of governing phenomena, the two-phase flow and heat transfer characteristics of microchannels have been shown to be different from those of conventional sized channels. The fact that microchannel is an attractive cooling option but at the same time there is a clear lack of understanding of related hydrodynamic and thermal transport phenomena which provides an impetus for microchannel research. This paper presents the flow patterns and flow pattern maps obtained for an experimental study of R134a during flow boiling in a horizontal microchannel. The microchannel was a fused silica tube, the outer surface of which was coated with thin, transparent and electrically conductive layer of Indium-Tin-Oxide (ITO). The microchannel was 781 m in internal diameter and 191 mm in heated length. Operating parameters during the experiments were: mass flux 100-400 kg/m2 s, heat flux 5-45 kW/m2, saturation temperature 25 and 30 °C. A High speed camera was used with a close up lens to capture the flow patterns evolved along the channel. Flow pattern maps are presented in terms of superficial gas and liquid velocity and in terms of Reynolds number and vapor quality plots. The results are compared with some flow pattern maps for conventional and micro scale channels available in literature.

    • This study proposes a method and apparatus to estimate shelf stability of nanofluids. Nanofluids are fabricated by dispersion of solid nanoparticles in base fluids, and shelf stability is a key issue for many practical applications of these fluids. In this study, shelf stability is evaluated by measuring the weight of settled solid particles on a suspended tray in a colloid versus time and correlated with the performance change of some nanofluid systems. The effects of solid particle concentration and bath sonication time were investigated for selected nanofluids. The results show the applicability of this simple method and the apparatus to evaluate nanofluid shelf stability. Furthermore, it shows that Stokes' law is not valid for determining the settling time of the tested nanoparticles probably due to their complicated shape and presence of surface modifiers. The effect of shelf stability on thermal conductivity and viscosity was illustrated for some nanofluids. Experimental results show that water-based Al2O3 nanofluids have quite good shelf stability and can be good candidates for industrial applications.

    • Objectives The aim of this study was to confirm the influence of TNFSF4 polymorphisms on systemic sclerosis (SSc) susceptibility and phenotypic features. Methods A total of 8 European populations of Caucasian ancestry were included, comprising 3014 patients with SSc and 3125 healthy controls. Four genetic variants of TNFSF4 gene promoter (rs1234314, rs844644, rs844648 and rs12039904) were selected as genetic markers. Results A pooled analysis revealed the association of rs1234314 and rs12039904 polymorphisms with SSc (OR 1.15, 95% CI 1.02 to 1.31; OR 1.18, 95% CI 1.08 to 1.29, respectively). Significant association of the four tested variants with patients with limited cutaneous SSc (lcSSc) was revealed (rs1234314 OR 1.22, 95% CI 1.07 to 1.38; rs844644 OR 0.91, 95% CI 0.83 to 0.99; rs844648 OR 1.10, 95% CI 1.01 to 1.20 and rs12039904 OR 1.20, 95% CI 1.09 to 1.33). Association of rs1234314, rs844648 and rs12039904 minor alleles with patients positive for anti-centromere antibodies (ACA) remained significant (OR 1.23, 95% CI 1.10 to 1.37; OR 1.12, 95% CI 1.01 to 1.25; OR 1.22, 95% CI 1.07 to 1.38, respectively). Haplotype analysis confirmed a protective haplotype associated with SSc, lcSSc and ACA positive subgroups (OR 0.88, 95% CI 0.82 to 0.96; OR 0.88, 95% CI 0.80 to 0.96; OR 0.86, 95% CI 0.77 to 0.97, respectively) and revealed a new risk haplotype associated with the same groups of patients (OR 1.14, 95% CI 1.03 to 1.26; OR 1.20, 95% CI 1.08 to 1.35; OR 1.23, 95% CI 1.07 to 1.42, respectively). Conclusions The data confirm the influence of TNFSF4 polymorphisms in SSc genetic susceptibility, especially in subsets of patients positive for lcSSc and ACA.

  • Identification of Novel Genetic Markers Associated with Clinical Phenotypes of Systemic Sclerosis through a Genome-Wide Association Strategy

    • The aim of this study was to determine, through a genome-wide association study (GWAS), the genetic components contributing to different clinical sub-phenotypes of systemic sclerosis (SSc). We considered limited (IcSSc) and diffuse (dcSSc) cutaneous involvement, and the relationships with presence of the SSc-specific auto-antibodies, anti-centromere (ACA), and anti-topoisomerase I (ATA). Four GWAS cohorts, comprising 2,296 SSc patients and 5,171 healthy controls, were meta-analyzed looking for associations in the selected subgroups. Eighteen polymorphisms were further tested in nine independent cohorts comprising an additional 3,175 SSc patients and 4,971 controls. Conditional analysis for associated SNPs in the HLA region was performed to explore their independent association in antibody subgroups. Overall analysis showed that non-HLA polymorphism rs11642873 in IRF8 gene to be associated at GWAS level with lcSSc (P = 2.32x10(-12), OR = 0.75). Also, rs12540874 in GRB10 gene (P = 1.27 x 10(-6), OR = 1.15) and rs11047102 in SOX5 gene (P = 1.39x10(-7), OR = 1.36) showed a suggestive association with lcSSc and ACA subgroups respectively. In the HLA region, we observed highly associated allelic combinations in the HLA-DQB1 locus with ACA (P = 1.79x10(-61), OR = 2.48), in the HLA-DPA1/B1 loci with ATA (P = 4.57x10(-76), OR = 8.84), and in NOTCH4 with ACA P = 8.84x10(-21), OR = 0.55) and ATA (P = 1.14x10(-8), OR = 0.54). We have identified three new non-HLA genes (IRF8, GRB10, and SOX5) associated with SSc clinical and autoantibody subgroups. Within the HLA region, HLA-DQB1, HLA-DPA1/B1, and NOTCH4 associations with SSc are likely confined to specific auto-antibodies. These data emphasize the differential genetic components of subphenotypes of SSc.

  • Effects of the antioxidant drug tempol on renal oxygenation in mice with reduced renal mass

    • We tested the hypothesis that reactive oxygen species (ROS) contributed to renal hypoxia in C57BL/6 mice with ⅚ surgical reduction of renal mass (RRM). ROS can activate the mitochondrial uncoupling protein 2 (UCP-2) and increase O(2) usage. However, UCP-2 can be inactivated by glutathionylation. Mice were fed normal (NS)- or high-salt (HS) diets, and HS mice received the antioxidant drug tempol or vehicle for 3 mo. Since salt intake did not affect the tubular Na(+) transport per O(2) consumed (T(Na/)Q(O2)), further studies were confined to HS mice. RRM mice had increased excretion of 8-isoprostane F(2α) and H(2)O(2), renal expression of UCP-2 and renal O(2) extraction, and reduced T(Na/)Q(O2) (sham: 20 ± 2 vs. RRM: 10 ± 1 μmol/μmol; P < 0.05) and cortical Po(2) (sham: 43 ± 2, RRM: 29 ± 2 mmHg; P < 0.02). Tempol normalized all these parameters while further increasing compensatory renal growth and glomerular volume. RRM mice had preserved blood pressure, glomeruli, and patchy tubulointerstitial fibrosis. The patterns of protein expression in the renal cortex suggested that RRM kidneys had increased ROS from upregulated p22(phox), NOX-2, and -4 and that ROS-dependent increases in UCP-2 led to hypoxia that activated transforming growth factor-β whereas erythroid-related factor 2 (Nrf-2), glutathione peroxidase-1, and glutathione-S-transferase mu-1 were upregulated independently of ROS. We conclude that RRM activated distinct processes: a ROS-dependent activation of UCP-2 leading to inefficient renal O(2) usage and cortical hypoxia that was offset by Nrf-2-dependent glutathionylation. Thus hypoxia in RRM may be the outcome of NADPH oxidase-initiated ROS generation, leading to mitochondrial uncoupling counteracted by defense pathways coordinated by Nrf-2.

    • The kidneys account for about 10% of the whole body oxygen consumption, whereas only 0.5% of the total body mass. It is known that intrarenal hypoxia is present in several diseases associated with development of kidney disease, including diabetes, and when renal blood flow is unaffected. The importance of deranged oxygen metabolism is further supported by deterioration of kidney function in patients with diabetes living at high altitude. Thus, we argue that reduced oxygen availability alters renal energy metabolism. Here, we introduce a novel magnetic resonance imaging (MRI) approach to monitor metabolic changes associated with diabetes and oxygen availability. Streptozotocin diabetic and control rats were given reduced, normal, or increased inspired oxygen in order to alter tissue oxygenation. The effects on kidney oxygen metabolism were studied using hyperpolarized [1-C-13]pyruvate MRI. Reduced inspired oxygen did not alter renal metabolism in the control group. Reduced oxygen availability in the diabetic kidney altered energy metabolism by increasing lactate and alanine formation by 23% and 34%, respectively, whereas the bicarbonate flux was unchanged. Thus, the increased prevalence and severity of nephropathy in patients with diabetes at high altitudes may originate from the increased sensitivity toward inspired oxygen. This increased lactate production shifts the metabolic routs toward hypoxic pathways.

Here they suffer high ratesof fishery exploitation, while hydropower regulation and the re-engineering of riversfor floating timber has led to the loss of spawning and rearing habitat and to a lossof connectivity among habitats (McKinnell 1998) .To remain viable in the face of demographic and environmental stochasticity,salmonid populations require a certain level of abundance, positive growth rates,adequate spatial structure, and access to (connectivity among) habitats of sufficientquantity and quality to express their life history and genetic diversity (McElhanyet al . 2000) . To understand what is limiting their productivity and viability anddevelop conservation actions for these threatened populations, we need informationon both the freshwater and marine phases of the salmon and sea trout life cycles.The Salmon Action Plan (SAP) 1997–2010 was adopted by IBSFC (InternationalBaltic Sea Fishery Commission), and states that by 2010 natural production inBaltic rivers should be >50% of the maximum production potential. To date, maximumnatural production levels have primarily been based on expert knowledge ratherthan empirical estimates (e.g. WGBAST 2008) . The Swedish Government nowrecognises the need for index rivers to obtain reliable estimates of abundance,productivity, population structure, and to collect the information on life-historydiversity needed to manage salmonid stocks.From 2005 to 2008, a pilot study was implemented in the River Sävarån (a small,unregulated forest river in northern Sweden), to monitor the downstream migrationsof salmon and trout, and explore its suitability as an index river. Rotary screwtraps were used to investigate the abundance of smolts as well as their timing, sizeand age, and to obtain samples to analyse the genetic composition of the stock. Parrdensities from electro-fishing surveys were compared with screw-trap data to determinewhether the two approaches produced similar smolt production estimates.

    • Experiments have been performed to investigate heat transfer in a circular vertical mini channel made of stainless steel (AISI 316) with internal diameter of 1.70 mm and a uniformly heated length of 245 mm using ammonia as working fluid. The experiments are conducted for a heat flux range of 15 to 350 kW/m(2) and mass flux range of 100 to 500 kg/m(2)s. The effects of heat flux, mass flux and vapour quality on the heat transfer coefficient are explored in detail. The experimental results show that the heat transfer coefficient increases with imposed wall heat flux while mass flux and vapour quality have no considerable effect. Experimental results are compared to predictive methods available in the literature for boiling heat transfer. The correlations of Cooper et al. [1] and Shah [3] are in good agreement with our experimental data.

Two phase frictional pressure drop variation with mass flux, vapour quality and heat flux was determined. The experimental results are compared to predictive methods available in literature for frictional pressure drop. The Homogeneous model and the correlation of Muller Steinhagen et al. [14] are in good agreement with our experimental data with MAD of 27% and 26% respectively.

    • This paper analyses environmental and socio-economic barriers for plantation activities on local and regional level and investigates the potential for carbon finance to stimulate the increased rates of forest plantation on wasteland, i.e., degraded lands, in southern India. Building on multidisciplinary field work and results from the model GCOMAP, the aim is to (1) identify and characterize the barriers to plantation activities in four agro-ecological zones in the state of Karnataka and (2) investigate what would be required to overcome these barriers and enhance the plantation rate and productivity. The results show that a rehabilitation of the wasteland based on plantation activities is not only possible but also anticipated by the local population and would lead to positive environmental and socio-economic effects at a local level. However, in many cases, the establishment of plantation activities is hindered by a lack of financial resources, low land productivity and water scarcity. Based on the model used and the results from the field work, it can be concluded that certified emission reductions such as carbon credits or other compensatory systems may help to overcome the financial barrier; however, the price needs to be significantly increased if these measures are to have any large-scale impact.
