    • BACKGROUND: Maternal diabetes (MD) is a risk factor for offspring to develop cardiovascular anomalies; this is of growing clinical concern since the number of women in childbearing age with compromised glucose homeostasis is increasing. Hyperglycemia abrogates cardiovascular development in vitro; however, a link to cardiovascular defects in diabetic offspring remains to be investigated. METHODS: We have studied cardiovascular development in offspring of MD rats by examining serial histological sections of GD 12.0-18.0 offspring. Development of pharyngeal arch artery malformations was analyzed and related to intracardiac anomalies. RESULTS: Pharyngeal arch artery and intracardiac defects were present in 27 of 37 MD GD 13.0-18.0 offspring. Early sixth arch arteries showed abrogated arteriogenesis, whereas fourth arch artery defects developed as a result of abnormal remodeling. Morphometrical analysis showed increased apoptosis in regressing artery segments and reduced apoptosis in persisting artery segments. Double outlet right ventricle with infundibular stenosis (tetralogy of Fallot) was predominantly found in combination with sixth artery defects and pulmonary atresia. As confirmed by morphometric analysis and three-dimensional (3D)-reconstructions, outflow tract defects coincided with endocardial cushion hypoplasia. Cases with teratology of Fallot additionally showed a shorter outflow tract. No relation with apoptosis or disturbed neural crest cell migration was found. CONCLUSIONS: Our data uniquely demonstrate mechanistic differences involved in the development of sixth and fourth artery anomalies. Whereas increased apoptosis induces fourth artery anomalies, pulmonary outflow obstruction abrogates sixth artery differentiation independent of apoptosis. The model presented allows analysis of diabetic conditions on cardiovascular development in vivo, essential for elucidating this teratology.

  • Deficiency of decidual IL-10 in first trimester missed abortion : A lack of correlation with the decidual immune cell profile

    • PROBLEM: To determine if first trimester missed abortion decidua is characterized by an altered immune cell profile and/or a modified interleukin (I L)-10 and interferon (IFN)-gamma production pattern compared with decidua from elective termination. METHOD OF STUDY: Flow cytometry and immunohistochemistry techniques were used to determine the decidual immune cell phenotypic profile and production pattern of IL-10 and IFN-gamma in cases of elective termination (n = 14) and missed abortion (n = 12). RESULTS: Both groups had a similar proportion of CD56(+) CD16(-),CD56(+) CD16(+), CD19(+), CD3(+), CD4(+), CD8(+), alphabeta T cells and gammadelta T cells. The majority of alphabeta and gammadelta positive T cells in both groups coexpressed the natural killer (NK) cell marker CD56, but lacked cell surface expression of CD3. Diminished decidual IL-10 staining was noted in 7/10 missed abortion cases compared with none of the elective termination cases (n = 12) (P = 0.007). A uniform decidual IFN-gamma staining pattern was observed in both groups. CONCLUSION: Decreased IL-10 production coupled with a sustained IFN-gamma presence noted in missed abortion compared with elective termination cases suggest that these cytokines may be important determinants in pregnancy outcome. In contrast, differences in the proportion of immune cells between both groups may not be a critical factor in early pregnancy loss. In normal pregnancy, decidual alphabeta and gammadelta positive T cells with reduced CD3 on their cell surface may be intrinsically restricted in T-cell receptor (TCR)-mediated activation.

    • Folate content in berries and the effect of cultivar on folate content in strawberries were investigated using a modified radioprotein-binding assay (RPBA) combined with solid phase extraction (SPE). With an SPE step prior to quantification the matrix effects in the RPBA assay were minimised. HPLC quantification was used to compare and characterise individual folate forms. High correlation (r = 0.9796) was shown and the mean difference between the RPBA and HPLC method was 6.7 4.8 mug/100 g. The predominating folate form was 6S-5-CH3-H-4-folate in all tested berries (>95%). Therefore, this biologically active folate form (6S-5-CH3-H(4)folate) must be chosen as calibrant because the RPBA response of the synthetic equimolar mixture of R- and S-diastereomers (6R,S-5-CH3-H(4)folate) was considerably higher, about 30%. The folate content in nine selected Swedish berries varied from 11 to 96 mug/100 g (fresh weight) and the highest folate content was found in rose hips (96 mug/100 g) and strawberries (74 mug/100 g). Moderate folate sources were raspberry (46 mug/100 g) and sea buckthorn (39 mug/100 g), whereas all other tested berries contained about 20 mug of folate/100 g or less. In strawberries, folate content varied significantly among eight different cultivars from 73 mug/100 g (Lina) to 99 mug/100 g (Melody).
