We have previously presented an ultrasonic standing wave (USW) based multicellular tumor spheroid (MCTS) culture platform where 100 MCST can be formed, processed and imaged while being retained in micro-wells [1]. This platform was used to show MCTS characterization based on on-chip microscopy and off-chip flow cytometry [2]. Here, we present a high-content 3D image cytometry method of whole MCTS based on DNA staining where spatial and functional information can be assessed.
